Resumo: The time of host-parasite interaction may influence the immune response against T. cruzi infection. But its peculiarities still demand more studies. In the present work we performed a kinetics of the cells that make up the innate immunity and stimulated them in vitro with T. cruzi antigens at times of 6 and 22 hours. The data were obtained by flow cytometry and analyzed using Flow-Jo software. Our data demonstrate that subsets of monocytes and neutrophils exert a role in the defense against infection. The group of inflammatory monocytes was found in the IND patients, however the neutrophils showed no differences. Another important point was that the recognition molecules, TLR-2 and 4, showed differences in expressions when compared at 6 and 22 hours, as well as CD-80 and HLA-DR, mainly in IND patients, together with higher production of IL-10. These findings reinforce the hypothesis of immunoregulation in these patients. However our analyzes in the CARD patients the recognition receptors were decreased along with IL-12 expression. In general, the greatest differences were observed when observed in the culture after 22 hours. It may indicate that the contact time of the host with the parasite directly influences the modulation of a more regulated or exacerbated immune response.

No presente trabalho realizamos uma cinética das células que compõem a imunidade inata e as estimulamos in vitro com antígenos do T. cruzi nos tempos de 6 e 22 horas. Os dados foram obtidos por citometria de fluxo e a análise dos mesmos realizada utilizando o software Flow-Jo. Nossos dados demonstram que as subpopulações de monócitos e neutrófilos exercem uma participação no processo de defesa contra a infecção. O grupo dos monócitos inflamatórios foi encontrado nos pacientes IND, entretanto os neutrófilos não apresentaram diferenças. Outro ponto importante foi observado que as moléculas de reconhecimento, TLR- 2 e 4, apresentam diferenças nas expressões quando comparadas nos tempos de 6 e 22 horas, assim como CD-80 e HLA-DR principalmente nos pacientes IND, juntamente com uma maior produção de IL-10. Esses achados reforçam hipótese da imunoregulação nesses pacientes. Entretanto nossas analises nos pacientes CARD os receptores de reconhecimento se apresentaram diminuídos juntamente com a expressão da IL-12. De forma geral as maiores diferenças encontradas foi quando observadas na cultura após 22 horas.
