Resumo: | Chronic Obstructive Pulmonary Disease (COPD) is an important public health problem and presents a high prevalence and a high economic and social cost when compared to other pulmonary diseases. However, several experimental models were proposed, aiming at a discovery of new therapeutic options. The use of mesenchymal stem cells (MSCs) has been the subject of several research studies and is treated by these as an important solution without fighting many diseases. Many advances in the field of cellular and molecular biology have contributed to the development in the techniques of repair and regeneration of organs and tissues injured by traumas, diseases and congenital deformities. This cells have possess the ability to preserve their own population and differentiate into cells from various tissues of the body. Low Level Laser Therapy (LLLT) is a relatively new therapy and has as main advantage the absence of side effects and possible use in the treatment of chronic lung diseases, besides having a low cost. In the present study we aimed to evaluate functional activity of structural, cells stimulated with 2.5% and 5% cigarette smoke extract (CSE) and incubated with human dental pulp mesenchimal stem cells (DPMSC). Epithelial cells (BEAS) were thawed and cultured. Three C57bl/6 mice were used for the primary culture of fibroblasts, which were irradiated or not with LLL, and incubated or not with DPMSC. The LLL parameters used were 30 mW, 660 and 808 nm, 60 seconds / well. The stem cells were extracted from the deciduous tooth pulp and cultured. All strains were subcultured until the necessary amount was obtained for the protocol. The effects of the association of LLL irradiation with DPMSC on the functional activity of lung cells induced by cigarette smoke extract through the release of cytokines (IL-6, IL-10, TNF-α and IFN-γ) and chemokine (IL-8) by ELISA. The results showed that in the BEAS culture there was an increase in levels of IL-10, IL-6 and IFN- in EFC+CTMDD+LBI when compared to EFC group. In the culture of fibroblasts, we observed an increase in all cytokines evaluated when the treatments associaten in relation to EFC group. Thus, studies on LLL and DPMSC have been promising, showing that they may increase in vitro the release of anti-inflammatory cytokines from fibroblasts and bronchial epithelial cells. Thus, the study about the functional activity of these cells may contribute to therapeutic strategies for COPD. |

Contudo, vários modelos experimentais têm sido propostos, objetivando a descoberta de novas opções terapêuticas. O uso de células-tronco mesenquimais (CTM) vem sendo objeto de estudo de várias pesquisas e é tratada por estes como uma importante solução no combate de muitas doenças. Muitos avanços na área da biologia celular e molecular vêm contribuindo para o desenvolvimento nas técnicas de reparo e regeneração de órgãos e tecidos injuriados por traumas, doenças e deformidades congênitas. Essas células possuem a capacidade de preservarem a sua própria população e se diferenciam em células de diversos tecidos do corpo. A terapia com Laser em Baixa Intensidade (LBI) é uma terapia relativamente nova e apresenta como principal vantagem a ausência de efeitos colaterais e possível utilização no tratamento das doenças pulmonares crônicas, além de possuir baixo custo. No presente estudo visamos avaliar alguns parâmetros em células estimuladas com extrato de fumaça de cigarro (EFC) 2,5% e 5%, irradiadas com LBI e incubadas com células-tronco da polpa dentária humana. As células epiteliais (BEAS) foram descongeladas e cultivadas. Três camundongos C57bl/6 foram utilizados para a cultura primária de fibroblastos, as culturas foram irradiadas ou não com LBI, e incubadas ou não com células-tronco. Os parâmetros do LBI utilizados foram de 30 mW, 660 nm e 808 nm, 60 segundos/poço. As células - tronco foram extraídas da polpa de dente decíduo e cultivadas. Todas as linhagens passaram por subcultivos até a obtenção da quantidade necessária para o protocolo. Foram avaliados os efeitos da associação da irradiação do LBI com CTMDD sobre a atividade funcional das células estruturais pulmonares induzidas por extrato de fumaça de cigarro através da liberação de citocinas (IL-6, IL-10, TNF-α e IFN- y) e quimiocina (IL-8), por ELISA. Os resultados mostraram que na cultura de BEAS houve um aumento dos níveis de IL-10, IL-6 e IFN-, no grupo EFC+CTMDD+LBI em relação ao grupo EFC. Já no sobrenadante da cultura de fibroblastos observamos um aumento de todas as citocinas avaliadas nos grupos com os tratamentos associados em relação ao EFC. Assim, os estudos acerca do LBI e da CTMDD têm sido promissores, mostrando que podem aumentar in vitro a liberação de citocinas anti-inflamatórias de fibroblastos e células epiteliais brônquicas.
