Resumo: A high incidence of intentional or accidental ingestion of paraquat (PQ) is related to the development of irreversible pulmonary fibrosis (PF). PF is a chronic and progressive disease characterized by injury to the lung parenchyma, inflammatory infiltrate and interstitial fibrosis. It is triggered by excessive and disordered deposition of collagen and other extracellular matrix components, resulting in severe changes in alveolar wall architecture. Due to the absence of an effective treatment, the objective of the study was to investigate the effect of treatment with vitamin D and/or photobiomodulation on the course of PF. C57BL6 mice were submitted to the induction of PF by the administration of Paraquat (10mg / kg, ip) and after 7 days of induction, the mice were treated with Vitamin D (5μg / kg, ip) and /or Photobiomodulation with LED (1x / day, 5 consecutive days, 150s). Treatment with vitamin D alone reduced the number of leukocytes in BAL as well as levels of IL-6, IL-17, TGF-beta, MMP-9, collagen deposition in lung tissue and tracheal reactivity. Furthermore, treatment with vitamin D increased levels of resolvin D1 in the BAL. Photobiomodulation reduced the number of cells in the BAL and increased the levels of resolvin D1. We also observed that the association between photobiomodulation and vitamin D did not cause an additive effect that justifies its use concomitantly. Thus, we conclude that in PQ-induced pulmonary fibrosis the treatment with vitamin D alone was more efficient than photobiomodulation and may emerge as a promising treatment tool for pulmonary fibrosis

A FP é uma doença crônica e progressiva caracterizada por lesão do parênquima pulmonar, infiltrado inflamatório e fibrose no interstício. É desencadeada pela excessiva e desordenada deposição de colágeno e outros componentes de matriz extracelular, resultando em severas mudanças na arquitetura da parede alveolar. Em função da ausência de um tratamento eficaz, o objetivo do estudo foi investigar o efeito do tratamento com vitamina D e/ou fotobiomodulação sobre o curso da FP. Para tanto, foram utilizados camundongos C57BL6 submetidos à indução de FP pela administração de Paraquat (10mg/Kg, ip) e após 7 dias da indução, os camundongos foram tratados com Vitamina D (5μg/kg, ip) e/ou Fotobiomodulação com LED (1x/dia, 5 dias consecutivos, 150s). O tratamento com vitamina D isoladadamente reduziu o número de leucócitos no LBA bem como os níveis de IL-6, IL-17, TGF-beta, MMP-9, depósito de colágeno no tecido pulmonar e reatividade traqueal. Ainda, o tratamento com vitamina D aumentou os níveis de resolvina D1 no LBA. Já a fotobiomodulação isolada reduziu o número de células no LBA e aumentou os níveis de resolvina D1. Observamos ainda que a associação da fotobiomodulação com vitamina D não causou efeito aditivo que justifique seu uso concomitantemente.
