Resumo: | Chronic oral inflammatory diseases (COID) are associated with systemic outcomes, in part due to modifications in the serum levels of inflammatory mediators. Considering that COID may lead to both local and systemic low-grade pro-inflammatory state, findings from observational studies have suggested an association between different parameters of oral health and physical fitness. The aim of this study was to evaluate the influence of apical periodontitis (AP) on physical training and fitness in Wistar rats, analyzing the impact of the inflammatory load of endodontic disease in the animals' physical conditioning. This study was structured according to the ARRIVE guidelines (Animal Research: Reporting of In Vivo Experiments). Forty-eight male Wistar rats, three months old and weighing between 400-450 g were used. The animals were divided into 6 groups (n = 8): sedentary rats, with AP induction in one tooth (1AP-SED); sedentary rats, with the induction of AP in four teeth (4AP-SED); sedentary rats, without AP induction (SED); exercised rats with physical training, with AP induction in one tooth (1PA-EX); exercised rats with physical training, with the induction of AP in four teeth (4AP-EX) and exercised rats with physical training, without induction of AP (EX). AP was induced through pulp exposure, keeping the pulp chamber open for 6 weeks. The physical training of the animals was carried out through a swimming protocol for 6 weeks. After the end of the experimental period, the animals were euthanized. For the assessment of physical fitness, the following tests were performed: maximum test of physical capacity and measurement of blood lactate concentration (before and after the training protocol); evolution of body weight and body mass index; adiposity, cardiac and muscular hypertrophy (soleus and gastrocnemius). The data obtained in the maximum tests were analyzed using the two-way ANOVA test with Tukey and Sidak post hoc. The EX-group showed the highest performance in the physical test, statistically different from the SED, 4PA-SED, 1PA-EX and 4PA EX groups (p ? 0.05). Regarding the maximum test of physical capacity, no differences were found among the 1PA-EX, 4PA-EX and SED groups (p > 0.05). No differences were found between groups in the assessment of blood lactate using the repeated measures ANOVA test. The evolution of the animals' body weight, over time, was evaluated using the ANOVA test of repeated measures and post hoc Tukey. The weight of all animals was increased during the initial 5 weeks of the experiment. From that period on, with the beginning of physical training, the animals of the sedentary groups followed the linear constant of weight gain, while in the trained animals the physical exercise attenuated the weight increase. However, there were no statistical differences (p > 0.05) in relation to body weight, regardless of the presence and amount of AP, between the trained animals. Regarding the Lee index and adiposity, the 1-way ANOVA test did not find differences between groups (p > 0.05). Regarding muscle parameters, an increase in cardiac muscle mass was observed in the EX-group compared to the SED and 1PA-SED groups (p ? 0.05). Present findings revealed that AP had a negative impact on the physical fitness of Wistar rats, suggesting a doseresponse effect in the relationship between the burden of endodontic disease and the physical capacity. |

Considerando que as DICB possuem efeito pr?-inflamat?rio, local e sist?mico, estudos observacionais sugerem a associa??o entre par?metros de sa?de bucal e aptid?o f?sica. O objetivo deste estudo foi avaliar a influ?ncia da periodontite apical (PA) no treinamento f?sico e na aptid?o f?sica, em ratos Wistar, analisando o impacto da carga inflamat?ria de origem endod?ntica no condicionamento f?sico dos animais. Este trabalho foi estruturado obedecendo as diretrizes ARRIVE (Animal Reserach: Reporting of In Vivo Experiments): Foram utilizados 48 ratos Wistar machos, com tr?s meses de idade e peso entre 400-450 g. Os animais foram distribu?dos em 6 grupos (n=8): ratos sem treinamento f?sico, com indu??o de PA em um dente (1PA-SED); ratos sem treinamento f?sico, com indu??o de PA em quatro dentes (4PA-SED); ratos sem treinamento f?sico, sem indu??o de PA (SED); ratos com treinamento f?sico, com indu??o de PA em um dente (1PA-EX); ratos com treinamento f?sico, com indu??o de PA em quatro dentes (4PA-EX) e ratos com treinamento f?sico, sem indu??o de PA (EX). A PA foi induzida atrav?s da exposi??o pulpar, mantendo a c?mara pulpar aberta por 6 semanas. O treinamento f?sico dos animais foi realizado atrav?s de um protocolo de nata??o durante 6 semanas. Ap?s o t?rmino do per?odo experimental, os animais foram eutanasiados. Para a avalia??o da aptid?o f?sica, foram realizados: teste m?ximo de capacidade f?sica e medidas da concentra??o de lactato sangu?neo (antes e ap?s o protocolo de treinamento); evolu??o do peso corporal e do ?ndice de massa corporal; ?ndice de adiposidade, hipertrofia card?aca e muscular (s?leo e gastrocn?mio) finais. Os dados obtidos nos testes m?ximos foram analisados atrav?s do teste de ANOVA de duas vias com post hoc de Tukey e Sidak. O grupo EX apresentou o melhor desempenho no teste e diferiu estatisticamente dos grupos SED, 4PA-SED, 1PA-EX e 4PA-EX (p ? 0,05). Os resultados de teste m?ximo dos grupos 1PA-EX e 4PA-EX n?o diferiu estatisticamente do grupo SED (p > 0,05). N?o foram encontradas diferen?as entre os grupos na avalia??o do lactato sangu?neo atrav?s do teste de ANOVA de medidas repetidas. A evolu??o do peso corporal dos animais, ao longo do tempo, foi avaliada atrav?s do teste de ANOVA de medidas repetidas e post hoc de Tukey. Todos os animais aumentaram de peso durante as 5 semanas iniciais do experimento. A partir desse per?odo, com o in?cio do treinamento f?sico, os animais dos grupos sedent?rios seguiram na constante linear de aumento de peso, enquanto, nos animais treinados foi poss?vel constatar que o exerc?cio f?sico atenuou este aumento. Por?m, n?o houve diferen?as estat?sticas (p > 0,05) em rela??o ao peso corporal, independente da presen?a e quantidade de PA, entre os animais treinados. Com rela??o ao ?ndice de Lee e de adiposidade o teste de ANOVA de 1 via n?o constatou diferen?as entre os grupos (p > 0,05). Em rela??o aos par?metros musculares, somente foi observado aumento da massa muscular card?aca, atrav?s do teste de ANOVA de 1 via e post hoc de Tukey no grupo EX em compara??o aos grupos SED e 1PA-SED (p ? 0,05).
