Estrutura sociogenética de ninhos de Euglossini (Hymenoptera: Apidae) e estrutura genética das populações urbanas de Euglossa cordata do estado de São Paulo

Resumo: | Euglossine bees (Hymenoptera: Apidae) are important pollinators in the Neotropical region. These bees are widely studied by collecting males in attractive baits, but there are few reports from females. In this work, we studied Euglossini bees using molecular tools, collecting bees in nests and adults in flowers at different levels, individual, nest and population. In Chapter 1, we use a non-lethal strategy for obtaining DNA from antenna conducting mark-recapture experiments. We found by a chi-square test the same chance to recapture the two groups of bees (with intact antennae vs removal antennae); this result suggests that the removal did not affect the survivor of bees. The DNA extracted from the antennas was successfully used for microsatellite analysis. In Chapter 2, we determine the sociogenetic structure of nests using microsatellite loci. Genetic analysis of Euglossa cordata, Euglossa townsendi and Eufriesea violacea nests revealed that brood is usually explained by a single mating, which corroborates the literature. In chapter 3, we used mitochondrial genes to verify the genetic structure of populations of Eg. cordata. The sequencing of mitochondrial genes cyt b and COI by direct PCR product showed high variation in several positions, showing two peaks or the mismatch between forward and reverse sequence. This result suggests the occurrence of heteroplasmy, and then, we have cloned these products. The clones showed even greater intra-individual variation, suggesting the occurrence of associated numts and heteroplasmy. Due to this variation, we could not establish the genetic structure of urban populations in Eg. cordata, although there is some evidence of interpopulation differentiation for mitochondrial haplotypes. In conclusion, this work has made contributions to a better understanding of the biology of Euglossine bees; employing molecular tools, we bring new information about familial and population genetics of this group of bees. |

Sao abelhas muito estudadas gracas as facilidades de obtencao de machos mediante o uso de iscas-odores; no entanto, poucos sao os estudos com femeas. Este trabalho teve o objetivo de estudar as abelhas euglossineas com o uso de ferramentas moleculares, coletando ninhos e adultos em flores para uma analise ao nivel do individuo, familia e populacao. No capitulo 1 e descrita uma estrategia nao letal para obtencao de DNA a partir da realizacao de experimentos de marcacao e recaptura coletando antenas de adultos de Euglossa cordata e Eulaema nigrita amostrados em flores de Thevetia peruviana. Foi demonstrada igual chance de recaptura nos dois grupos (antenas intactas vs. antenas seccionadas) pelo teste de qui-quadrado, sugerindo nao afetar a sobrevivencia das abelhas cujas antenas foram seccionadas. As antenas retiradas foram amplificadas com sucesso para microssatelites, confirmando a utilidade deste material como fonte de DNA para analises geneticas. No capitulo 2 e descrita a estrutura sociogenetica intranidal com a utilizacao de locos microssatelites. A analise genetica de ninhos de Eg. cordata, Eg. townsendi e Eufriesea violacea revelou que as progenies sao explicadas pelo acasalamento monandrico da femea, o que vem corroborar dados da literatura. No capitulo 3, utilizando genes mitocondriais, sao descritos os esforcos para verificacao da estrutura genetica das populacoes de Eg. cordata. O sequenciamento dos genes mitocondriais cyt b e COI por produto direto de PCR exibiu elevada variacao em varios sitios, apresentando dois picos numa mesma posicao ou a nao correspondencia entre as fitas forward e reverse, sugerindo a ocorrencia de heteroplasmia. Foi, entao, realizada a clonagem e nos clones foi detectada variacao intraindividual nas sequencias cyt b e COI, o que sugere a ocorrencia associada de numts e de heteroplasmia. A presenca desta variacao, cujo significado ainda esta por ser completamente elucidado, nao permitiu determinar a estrutura genetica das populacoes urbanas de Eg. cordata, embora haja indicios de diferenciacao interpopulacional para os haplotipos mitocondriais.
