Resumo: | The rapid development of biotechnology and increasing environmental awareness among producers and consumers are using organic products in the market's preference. It considered their physical-chemical properties to be the use of biosurfactants that are part of these potential compound groups in a variety of industrial and biotechnological applications, such as food additives, cosmetics and detergents. Compared to chemical surfactants they have many advantages as they are biodegradable and less toxic. In this sense, the present study aimed to select biosurfactant-producing microorganisms from two Candidas species (C. guilliermondii and C. lipolytica) cultivated in different carbon sources (glucose, sugar cane honey and frying residual oil)) and nitrogen (yeast extract, urea and millocin). After selection of the microorganism and production medium, the bio-surfer was tested for surface tension, emulsion activity and toxicity using the Artemia salina microcrustacean, vegetable seeds and onion (Allium cepa) as bioindicator. Then, Gas Chromatography (GC), Infrared (IR) and Nuclear Magnetic Resonance (NMR) analyzes were conducted for structural characterization of the biomolecule. According to the results obtained, it was observed that in Candida models cultivated in 5.0% oil, 5.0% milk and 5.0% residual frying oil were selected as the best biosurfactant producer after a reduction of water surface tension from 71 mN / m to 30mN / m and a yield of 21g / L with a Critical Micellar Concentration of 0.7 g / L. Isolated biosurfactant has no toxicity on microcrustaceans Artemia salina, Allium cepa and neither as vegetable seeds tested. Regarding the characterization revealed that the biosurfactant studied is a glycolipid. All the mayonnaise analyzed remained stable, with no pathogenic microorganisms. Biosurfactant isolated from C. guilliermondii has potential as a bioemulsifier for application in the food industry. |

Devido suas propriedades físico-químicas, os biossurfactantes tornam-se um grupo atrativo de compostos com uso potencial em uma variedade de aplicações industriais e biotecnológicas, como aditivos em alimentos, cosméticos e detergentes. Em comparação com os surfactantes químicos têm muitas vantagens, pois são, biodegradáveis e menos tóxicos. Nesse sentido, o presente estudo teve como objetivo a selecionar micro-organimos produtores de biossurfactantes a partir de duas espécies de Candidas (C. guilhermondi e C. lipolytica) cultivadas em diferentes fontes de carbono (glicose, melaço de cana e óleo residual de fritura) e nitrogênio (extrato de levedura, ureia e milhocina) e produzir biossurfactante com propriedades emulsificantes para aplicação como aditivo em sistemas alimentares. Para a seleção do micro-organismo e do meio de produção, os biossurfactantes produzidos foram testados quanto à tensão superficial, atividade de emulsificação e rendimento. O biossurfactante selecionado foi avaliado quanto à toxicidade usando o microcrustáceo Artemia salina, sementes de vegetais (Brassica oleracea, Solanum lycopersicum e Cucumis anguria) e da cebola (Allium cepa) como bioindicadores. Em seguida, análises por Cromatografia Gasosa (GC), Infravermelho (IV) e Ressonância Magnética Nuclear (RMN) foram conduzidas para a caracterização estrutural da biomolécula. A propriedade emulsificante do biossurfactante foi testada na formulação de sete tipos diferentes de molhos (tipo maionese). De acordo com os resultados obtidos foi observado que a Candida guillhermodii cultivada em meio contendo 5,0% de melaço, 5,0% de milhocina e 5,0% do óleo de fritura residual foi selecionado como o melhor produtor de biossurfactante, apresentando uma redução da tensão superficial da água de 71 mN/m para de 28mN/m e um rendimento de 21g/L com uma Concentração Micelar Crítica de 0,7 g/L. O biossurfactante isolado não apresentou toxicidade frente aos bioindicadores. A caracterização estrutural revelou que o biossurfactante estudado é um glicolipídeo. Todas as maioneses analisadas, contendo biossurfactante produzido por C. guilliermondii após um mês de armazenamento refrigerado mantiveram-se estáveis, com ausência de microorganismos patogênicos.
