Resumo: | INTRODUCTION Annexin-A1 (ANXA1) is a protein widely distributed in the body and is particularly abundant in epithelial cells and leukocytes. There is evidence of its participation in signal transduction, cell differentiation and proliferation. In the literature, there are conflicting reports on the expression of ANXA1 in breast cancer. In this study, we investigated the expression of ANXA1, its receptors, major posttranslational modifications and its correlation with neoplastic molecular subtypes. MATERIALS AND METHODS We used paraffin blocks of 42 patients with invasive ductal breast cancer diagnostic. ANXA1 protein expression, FPR1, FPR2, phosphorylated ANXA1 in the Ser27 and Tyr21 portion (ANXA1 and ANXA1-SER-TYR) and EGFR were quantified by mean optical density in the immunofluorescence. RESULTS Epithelial cells showed decreased expression of ANXA1 and increased of ANXA1-TYR in Luminal A, Luminal B/HER e Triple-Negative subgroups. FPR2 and EGFR showed a similar pattern to ANXA1 expression. However, FPR1 was not detected in normal epithelium or in tumor cells. CONCLUSION ANXA1 expression as well FPR2 and EGFR were heterogeneous between molecular breast cancer subgroups and inversely proportional to ANXA1-TYR expression revealing the paracrine effects of ANXA1 and involvement of its phosphorylated status in carcinogenesis between different molecular subtypes. |

Existem evidências de sua participação na transdução de sinais, na diferenciação e no controle do crescimento. Na literatura, há relatos discrepantes sobre a expressão da ANXA1 no câncer de mama. Nesse estudo, investigamos a expressão da ANXA1 no carcinoma ductal, seus receptores, principais modificações pós-traducionais e sua correlação com os subtipos moleculares da neoplasia. MATERIAL E MÉTODOS Foram utilizados blocos de parafina de 42 pacientes com diagnóstico de carcinoma ductal invasivo da mama. A expressão das proteínas ANXA1, FPR1, FPR2, ANXA1 fosforilada na porção Ser27 e Tyr21 (ANXA1-SER e ANXA1-TYR) e do EGFR foi quantificada por densidade óptica média na reação de imunofluorescência. RESULTADOS As células epiteliais neoplásicas apresentaram uma redução da expressão da ANXA1 e aumento da ANXA1-TYR nos subgrupos Luminal A, Luminal B/HER e Triplo Negativo. Os receptores FPR2 e EGFR apresentaram um padrão de expressão protéica semelhante ao da ANXA1. Já o receptor FPR1 não foi detectado no epitélio normal e nas células tumorais.
