Resumo: | Melasma is easy to diagnose, but difficult to cure. It affects women of childbearing age most, affecting only 10% of men. There are already several treatments on the market, but none that are 100% efficient since it has a high recurrence rate. The appearance of dark spots on the skin (melasma), in a way, interferes with the quality of life, since this condition affects the person's self-esteem. In the search for the efficiency of a treatment with less side effects, the use of nanotechnology has presented formulations that meet these needs. Thus, producing new formulations, on a nanoscale, can be an alternative to increase efficiency in treatment, since it improves hydration, skin permeation and can also reduce side effects, also increasing the effectiveness of the active ingredient. In this sense, this work aimed to produce, characterize and carry out studies, in vitro and in vivo, of a liposomal source containing α-arbutin, for application in the treatment of melasma. The formulations were supplied by the ethanol injection method and remained stable for up to 60 days when stored at room temperature and under refrigeration. The liposomal vesicles had an average diameter between 140 nm to 175 nm, polydispersion index below 0.20, zeta potential between -26.0 to -10.0 mV, and pH between 5.0-5.5, simulating the pH values of human skin. For skin biometrics studies, liposomal information, containing α-arbutin was incorporated in a gel-cream, showing stability is up to 60 days when stored at room temperature or under refrigeration. In vitro studies, carried out on keratinocytes, have shown that they do not alter the viability of normal skin cells, thus showing good safety for topical application. The results of skin evaluation, with women between 18 and 50 years old, demonstrated the voluntary safety for the use of the liposomal diet containing α-arbutin, incorporated in gel-cream, since none of the women arouses an adverse reaction to the nanotechnological product, in the period of proposed progress. |

Ele atinge mais as mulheres em idade fértil, afetando somente 10% dos homens. Já existem vários tratamentos no mercado, porém nenhum que seja 100% eficiente já que ele tem um alto índice de recidiva. O surgimento de manchas escuras na pele (melasma), de certa forma, interfere na qualidade de vida, uma vez que esta condição afeta a autoestima da pessoa. Na busca pela eficiência de um tratamento com menores efeitos colaterais, o uso da nanotecnologia tem apresentado formulações que atendem a estas necessidades. Assim, produzir novas formulações, em escala nanométrica, pode ser uma alternativa para aumentar a eficiência no tratamento, uma vez que melhora a hidratação, a permeação cutânea podendo ainda reduzir os efeitos colaterais, aumentando também a eficácia do ativo. Neste sentido, os objetivos deste trabalho foram produzir, caracterizar e realizar estudos in vitro e in vivo, de uma formulação lipossomal contendo α-arbutin, para aplicação no tratamento de melasma. As formulações foram obtidas pelo método de injeção de etanol e mantiveram-se estáveis por até 60 dias, quando armazenadas em temperatura ambiente e sob refrigeração. As vesículas lipossomais obtidas apresentaram diâmetro médio entre 140 nm a 175 nm, índice de polidispersão abaixo de 0,20, potencial zeta entre -26,0 a -10,0 mV e, pH entre 5,0-5,5, simulando os valores de pH da pele humana. Para estudos de biometria cutânea, a formulação lipossomal, contendo α-arbutin foi incorporada em um gel-creme, na qual teve estabilidade por até 60 dias, quando armazenada em temperatura ambiente ou sob refrigeração. Estudos in vitro, feitos em queratinócitos (HaCat), demonstraram não alterar a viabilidade das células normais da pele, mostrando assim ter uma boa segurança para aplicação tópica.
