Resumo: | Anxiety has increased significantly in the world population, which has encouraged the search for innovative alternatives that act in the treatment. Research with essential oil of Citrus aurantium L. (OEC) has shown results with anxiolytic effects in both animal and human groups. However, limitations regarding use, preservation, storage and dosage quantification affect the efficiency of the therapy. The potential use of cyclodextrin complexes as drug carriers has been successfully explored. The study aims to evaluate the anxiolytic potential of the inclusion complex 2-hydroxypropyl-βcyclodextrin (HP-β-CD) containing OEC. The preparation of the inclusion complex was developed by the lyophilization method using the Alpha 1-2 LD plus lyophilizer model. Limonene (LIM), the main OEC compound, with HP-β-CD in molar ratio (1: 1M) was dispersed in ethanol using a laboratory shaker for 36 hours at room temperature (25 ° C) to allow the formation of and avoid the loss of volatiles in the atmosphere. Nonclinical pharmacological tests were carried out using mice distributed in experimental groups and control groups submitted to the elevated plus maze test, perforated plate test, sphere hide test, and route-rod test. Satisfactory results of the anxiolytic effect of the OEC complexed in HP-β-CD were observed, with the indication of an potentiation of the effect with doses lower than 500 mg / kg and 250 mg / kg complexed, suggesting lower losses in the pharmacokinetic process. |

Pesquisas com óleo essencial do Citrus aurantium L. (OEC) tem apresentado resultados com efeitos ansiolíticos tanto em grupos animais como em grupos humanos. Porém limitações em relação ao uso, a sua preservação, estocagem e quantificação da dosagem afetam a eficiência da terapêutica. O uso potencial de complexos de Ciclodextrinas como carreadores de fármacos tem sido explorado com sucesso na produção de medicamentos, melhando as características físico-quimicas. O estudo tem como objetivo avaliar o potencial ansiolítico do complexo de inclusão 2- hidroxipropil-β-ciclodextrina (HP-β-CD) contendo OEC. A preparação do complexo de inclusão foi desenvolvida pelo método de liofilização usando o modelo de liofilizador Alpha 1-2 LD plus. O limoneno (LIM), principal composto do OEC, com HP-β-CD na razão molar (1: 1M) foi disperso em etanol usando um agitador de laboratório por 48 horas em temperatura ambiente (25° C) para permitir a formação de complexos e evitar a perda de voláteis na atmosfera. Foram realizados ensaios farmacológicos não clínicos utilizando camundongos distribuídos em grupos experimentais e grupos controle submetidos ao teste de labirinto de cruz elevado, teste da placa perfurada, teste de esconder esferas, e teste rota-rod.
