Resumo: | This work was developed in properties with creations of horses in the micro-regions of Itaguai and Serrana, both located in the state of Rio de Janeiro. The study aimed to evaluate the role of molluscs and trematodes in Neorickettsia risticii cycle in creations located in the micro-region of Itagua? and analyze the frequency of antibody against N. risticii in horses from Serrana micro-region, standing out the possible associated factors with the seropositivity of animals. Among thirteen properties studied in Itaguai micro-region were collected 410 specimens of limnic molluscs using hand net adapted by fine screen. The molluscs were exposed to the incandescent light (60 w) during 2-4 hours for research of trematodes larval forms. Subsequently, proceeded to molecular detection of N. risticii by Polymerase Chain Reaction in real time (qPCR) in DNA samples of molluscs as in samples of trematodes larval. Many taxons of molluscs were found: 71.70% (294/410) represented by Melanoides tuberculata; 10.97% (45/410) by Pomacea sp.; 3.65% (15/410) by Drepanotrema anatinum; 6.58% (27/410) by Biomphalaria tenagophila; 0.24% (1/410) by B. straminea; 5.60% (23/410) by Physa acuta and 1.21% (5/410) by Family Hydobriidae. A frequency of 3.17% (13/410) of molluscs parasited by trematodes was observed. According to the morphometric and biological characteristics observed in trematodes were classified the following cercariae types: Megalourous cercariae, Pleurolophocercous cercariae and Furcocercous cercariae. No amplification of the target DNA for N. risticii was observed in any DNA samples of molluscs and trematodes analyzed. Regarding to sero-epidemiological study of N. risticii in horses of Serrana micro-region, blood samples from 354 animals were collected, residing at 49 properties of region, by jugular venipuncture in tubes of 5mL without anticoagulant. Blood samples were subjected to centrifugation to obtainment of serums which were stored in freezer at -20?C until analysis moment. For detection of antibodies against N. risticii was used the Immunofluorescence Antibody Test (IFAT), and the samples were considered positive when reacted to N. risticii at 1:50 dilution. Between the municipalities of Petropolis and Teresopolis, was found a frequency of 5.9% of serologically reactive horses. Among the possible factors analyzed, it was found that gender, breed, age, zootechnical and sanitary management, presence of snails and flooded areas did not present association with seropositivity of horses by N. risticii (p>0.05). The region of origin or provenance of the animal was the only factor associated (p<0.05) with seropositivity for N. risticii in the area studied. The presence of animals serologically reactive to N. risticii in Petropolis and Teresopolis indicate the possible movement of the agent in one more geographical area of Brazil. Therefore, the research of molluscs and trematodes that sheltering the bacterium N. risticii may help in understanding the chain of Equine Monocytic Ehrlichiosis (EME) transmission, especially in regions with serological evidence of affected animals. O estudo teve por objetivo avaliar o papel de moluscos e tremat?deos no ciclo de Neorickettsia risticii nas cria??es localizadas na microrregi?o de Itagua? e analisar a frequ?ncia de anticorpo anti-N. risticii em equinos da microrregi?o Serrana, destacando os poss?veis fatores associados ? soropositividade dos animais. Das treze propriedades estudadas na microrregi?o de Itagua?, foram coletados 410 esp?cimes de moluscos l?mnicos, por meio de pu?? adaptado por tela fina. Os moluscos foram expostos ? luz incandescente (60 w) por 2-4 horas para pesquisa de formas larvais de tremat?deos. Posteriormente, procedeu-se ? detec??o molecular de N. risticii atrav?s da Rea??o em Cadeia da Polimerase em tempo real (qPCR) tanto nas amostras de DNA dos moluscos como nas amostras de larvas de tremat?deos. Foram encontrados diversos t?xons de moluscos sendo 71,70% (294/410) representados por Melanoides tuberculata; 10,97% (45/410) por Pomacea sp.; 3,65% (15/410) por Drepanotrema anatinum; 6,58% (27/410) por Biomphalaria tenagophila; 0,24% (1/410) por Biomphalaria straminea; 5,60% (23/410) por Physa acuta e 1,21% (5/410) pela Fam?lia Hydobriidae. Foi observada uma frequ?ncia de 3,17% (13/410) de moluscos parasitados por tremat?deos. De acordo com as caracter?sticas morfom?tricas e biol?gicas observada nos tremat?deos foram classificados os seguintes tipos cercariais: Megalourous cercariae, Pleurolophocercous cercariae e Furcocercous cercariae. N?o foi observada amplifica??o do DNA-alvo para N. risticii em nenhuma das amostras de DNA de moluscos e tremat?deos analisados. Com rela??o ao estudo soro-epidemiol?gico de N. risticii em equinos da Microrregi?o Serrana, foram coletadas 354 amostras de sangue dos animais, residentes em 49 propriedades da regi?o, atrav?s de venopun??o jugular em tubos de 5 ml sem anticoagulante. As amostras de sangue foram submetidas ? centrifuga??o para obten??o dos soros que foram armazenadas em freezer a -20 ?C at? o momento das an?lises. Para detec??o de anticorpo anti-N. risticii utilizou-se a Rea??o de Imunofluoresc?ncia Indireta (RIFI), e as amostras foram consideradas positivas quando reagiram ? N. risticii sob a dilui??o de 1:50. Dentre os munic?pios de Petr?polis e Teres?polis encontrou-se uma frequ?ncia de 5,9% dos equinos sorologicamente reativos. Dentre os poss?veis fatores analisados, constatou-se que sexo, ra?a, idade, manejo sanit?rio e zoot?cnico, presen?a de caramujos e ?reas alagadas n?o apresentaram associa??o com a soropositividade dos equinos por N. risticii (p>0,05). A regi?o de origem ou proced?ncia do animal foi o ?nico fator associado (p<0.05) ? soropositividade por N. risticii na ?rea estudada. A presen?a de animais sorologicamente reativos para N. risticii em Petr?polis e Teres?polis indica a circula??o do agente em mais uma ?rea geogr?fica do Brasil. Logo, a pesquisa de moluscos e tremat?deos que alberguem a bact?ria N. risticii pode auxiliar no entendimento da cadeia de transmiss?o da Erliquiose Monoc?tica Equina (EME), principalmente em regi?es com evid?ncias sorol?gicas de animais acometidos. This work was developed in properties with creations of horses in the micro-regions of Itaguai and Serrana, both located in the state of Rio de Janeiro. The study aimed to evaluate the role of molluscs and trematodes in Neorickettsia risticii cycle in creations located in the micro-region of Itagua? and analyze the frequency of antibody against N. risticii in horses from Serrana micro-region, standing out the possible associated factors with the seropositivity of animals. Among thirteen properties studied in Itaguai micro-region were collected 410 specimens of limnic molluscs using hand net adapted by fine screen. The molluscs were exposed to the incandescent light (60 w) during 2-4 hours for research of trematodes larval forms. Subsequently, proceeded to molecular detection of N. risticii by Polymerase Chain Reaction in real time (qPCR) in DNA samples of molluscs as in samples of trematodes larval. Many taxons of molluscs were found: 71.70% (294/410) represented by Melanoides tuberculata; 10.97% (45/410) by Pomacea sp.; 3.65% (15/410) by Drepanotrema anatinum; 6.58% (27/410) by Biomphalaria tenagophila; 0.24% (1/410) by B. straminea; 5.60% (23/410) by Physa acuta and 1.21% (5/410) by Family Hydobriidae. A frequency of 3.17% (13/410) of molluscs parasited by trematodes was observed. According to the morphometric and biological characteristics observed in trematodes were classified the following cercariae types: Megalourous cercariae, Pleurolophocercous cercariae and Furcocercous cercariae. No amplification of the target DNA for N. risticii was observed in any DNA samples of molluscs and trematodes analyzed. Regarding to sero-epidemiological study of N. risticii in horses of Serrana micro-region, blood samples from 354 animals were collected, residing at 49 properties of region, by jugular venipuncture in tubes of 5mL without anticoagulant. Blood samples were subjected to centrifugation to obtainment of serums which were stored in freezer at -20?C until analysis moment. For detection of antibodies against N. risticii was used the Immunofluorescence Antibody Test (IFAT), and the samples were considered positive when reacted to N. risticii at 1:50 dilution. Between the municipalities of Petropolis and Teresopolis, was found a frequency of 5.9% of serologically reactive horses. Among the possible factors analyzed, it was found that gender, breed, age, zootechnical and sanitary management, presence of snails and flooded areas did not present association with seropositivity of horses by N. risticii (p>0.05). The region of origin or provenance of the animal was the only factor associated (p<0.05) with seropositivity for N. risticii in the area studied. The presence of animals serologically reactive to N. risticii in Petropolis and Teresopolis indicate the possible movement of the agent in one more geographical area of Brazil. Therefore, the research of molluscs and trematodes that sheltering the bacterium N. risticii may help in understanding the chain of Equine Monocytic Ehrlichiosis (EME) transmission, especially in regions with serological evidence of affected animals.
