Resumo: The white shrimp Litopenaeus schmitti occurs in the Western Atlantic, from the Caribbean to Rio Grande do Sul, and it is a species of great commercial importance to the region of Sepetiba Bay - RJ. This study was made to analyze the main feature secretions of the vas deferens and the sperm viability in fresh prawns and post mortem and their usefullness for reproduction. The first study aimed to compare through the histological characterization the area of the seminiferous tubules in different seasons; comparing histologically and histochemically the secretions of the vas deferens (the proximal and medial regions) and the terminal ampoule, as well as the ultrastructure of spermatozoa in the regions of the white shrimp L. schmitti fresh and post mortem, characterizing the morphology of the sperm through the transmission electron microscopy and the reproductive system through the scanning electron microscopy. The animals came from the commercial fishing of Sepetiba Bay (Saco da Pombeba) latitude 23 ? 3'13 .91 "S and longitude 43 ? 54'44 .93" W. 21 adult males were bought post mortem (approximately 48 hours), every season, spring, summer, autumn and winter (totaling 84 animals), and three living adult males (fresh) for each season. The area of the testicles appeared higher in summer indicating that this is the period of the year of full reproductive activity and lower in winter, suggesting that this is the resting period of this species in the Sepetiba Bay - RJ. Through histological and ultrastructural analysis of sperm vas deferens and terminal ampoule of animals post mortem it was found that it is possible to use these spermatophore males to inseminate females as well as for their cryopreservation. The secretions present in the medial vas deferens, terminal ampoule and lumen spermatophore possess polysaccharides and proteins, indicating that they exert protective functions of sperm and supply of energy-rich substrates, increasing their survival time (viability) in the female. The sperm of the shrimp L. schmitti is composed of the main body, the acrosomal region and the thorn source acrosomal. The second study was divided into two tests, the first was to evaluate the feasibility of two protocols of freezing sperm and spermatophore mass (using 10% Glycerol) and the second, the efficiency of two cryoprotectants (Glycerol and 10% DMSO) in both mass sperm cryopreservation, as the spermatophore using the best protocol previously evaluated. There has been used a total of 55 male adult shrimps (post mortem) for the first experiment and 65 for the second. Adult males were equaly obtained as the ones from the first study. The protocol in which the temperature fell by 0.5oC/min to reach -32oC satisfactory results for cryopreservation, however, the protocol which fell 20oC/min to reach-120oC was unsatisfactory due to low sperm survival. Both Glycerol and DMSO may be used in cryopreservation of sperm mass.

Este trabalho teve como principal caracter?stica analisar as secre??es do vaso deferente e a viabilidade dos espermatozoides em camar?es frescos e post mortem para utiliza??o dos mesmos na reprodu??o. O primeiro estudo teve por objetivos comparar atrav?s da caracteriza??o histol?gica a ?rea dos t?bulos semin?feros nas diferentes esta??es do ano; comparar histol?gica e histoquimicamente as secre??es do vaso deferente (regi?es proximal e m?dio) e da ampola terminal, assim como a ultraestrutura dos espermatozoides nestas regi?es do camar?o branco L. schmitti frescos e post mortem, caracterizar morfologicamente os espermatozoides atrav?s de microscopia eletr?nica de transmiss?o e o sistema reprodutor pela microscopia eletr?nica de varredura. Os animais foram oriundos da pesca comercial da ba?a de Sepetiba (Saco da Pombeba) latitude 23? 3'13.91"S e longitude 43?54'44.93"O. Foram comprados 21 machos adultos post mortem (aproximadamente a 48h), em cada esta??o, na primavera, ver?o, outono e inverno (totalizando 84 animais), e tr?s machos adultos vivos (frescos) para cada esta??o do ano. A ?rea do test?culo se apresentou superior no ver?o indicando ser esta a ?poca do ano de plena atividade reprodutiva e inferior no inverno, sugerindo ser este o per?odo de repouso desta esp?cie na ba?a de Sepetiba - RJ. Por meio das an?lises histol?gicas e ultraestruturais dos espermatozoides do vaso deferente e da ampola terminal de animais post mortem constatou-se, que ? poss?vel a utiliza??o dos espermat?foros destes machos para a insemina??o de f?meas, assim como para a criopreserva??o dos mesmos. As secre??es presentes no vaso deferente m?dio, na ampola terminal e no l?men dos espermat?foros possuem polissacar?deos e prote?nas, o que indica que estas exercem as fun??es de prote??o dos espermatozoides e fornecimento de substratos ricos em energia, aumentando o tempo de sobreviv?ncia (viabilidade) dos mesmos na f?mea. O espermatozoide do camar?o L. schmitti ? composto por um corpo principal, a regi?o across?mica e o espinho de origem acrossomal. O segundo estudo foi dividido em dois ensaios, o primeiro teve por objetivo avaliar a viabilidade de dois protocolos de congelamento da massa esperm?tica e do espermat?foro (utilizando Glicerol a 10%) e o segundo a efici?ncia de dois crioprotetores (Glicerol e DMSO a 10%) na criopreserva??o tanto da massa esperm?tica, quanto do espermat?foro utilizando o melhor protocolo avaliado anteriormente. Foram utilizados um total de 55 camar?es machos adultos (post mortem) para o primeiro experimento e 65 para o segundo. Os machos adultos foram adquiridos de maneira semelhante ao do primeiro estudo. O protocolo em que a temperatura apresentou queda de 0,5oC/min at? atingir -32oC apresentou resultados satisfat?rios para a criopreserva??o, entretanto, o protocolo que apresentou queda de 20oC/min at? alcan?ar -120oC foi insatisfat?rio devido a baixa sobreviv?ncia esperm?tica. description The white shrimp Litopenaeus schmitti occurs in the Western Atlantic, from the Caribbean to Rio Grande do Sul, and it is a species of great commercial importance to the region of Sepetiba Bay - RJ. This study was made to analyze the main feature secretions of the vas deferens and the sperm viability in fresh prawns and post mortem and their usefullness for reproduction. The first study aimed to compare through the histological characterization the area of the seminiferous tubules in different seasons; comparing histologically and histochemically the secretions of the vas deferens (the proximal and medial regions) and the terminal ampoule, as well as the ultrastructure of spermatozoa in the regions of the white shrimp L. schmitti fresh and post mortem, characterizing the morphology of the sperm through the transmission electron microscopy and the reproductive system through the scanning electron microscopy. The animals came from the commercial fishing of Sepetiba Bay (Saco da Pombeba) latitude 23 ? 3'13 .91 "S and longitude 43 ? 54'44 .93" W. 21 adult males were bought post mortem (approximately 48 hours), every season, spring, summer, autumn and winter (totaling 84 animals), and three living adult males (fresh) for each season. The area of the testicles appeared higher in summer indicating that this is the period of the year of full reproductive activity and lower in winter, suggesting that this is the resting period of this species in the Sepetiba Bay - RJ. Through histological and ultrastructural analysis of sperm vas deferens and terminal ampoule of animals post mortem it was found that it is possible to use these spermatophore males to inseminate females as well as for their cryopreservation. The secretions present in the medial vas deferens, terminal ampoule and lumen spermatophore possess polysaccharides and proteins, indicating that they exert protective functions of sperm and supply of energy-rich substrates, increasing their survival time (viability) in the female. The sperm of the shrimp L. schmitti is composed of the main body, the acrosomal region and the thorn source acrosomal. The second study was divided into two tests, the first was to evaluate the feasibility of two protocols of freezing sperm and spermatophore mass (using 10% Glycerol) and the second, the efficiency of two cryoprotectants (Glycerol and 10% DMSO) in both mass sperm cryopreservation, as the spermatophore using the best protocol previously evaluated. There has been used a total of 55 male adult shrimps (post mortem) for the first experiment and 65 for the second. Adult males were equaly obtained as the ones from the first study. The protocol in which the temperature fell by 0.5oC/min to reach -32oC satisfactory results for cryopreservation, however, the protocol which fell 20oC/min to reach-120oC was unsatisfactory due to low sperm survival. Both Glycerol and DMSO may be used in cryopreservation of sperm mass.
