Resumo: This paper describes an outbreak of lysosomal storage disease in sheep induced by Sida planicaulis in the state of Rio de Janeiro. The consumption of S. planicaulis is responsible for this disease in cattle, horses, goat, sheep and captive deer, which occurs naturally, when the plant is abundant and there is shortage of other forage. The main toxic compound in this plant, Swainsonine, inhibits ?-manosidasis I e II enzymatic activitiy and causes glycoprotein storage in lysosomes. The storage provokes compression, deficiency in intracytoplasmatic transport, and finally, degenerative changes in various tissues, in which, nervous system is the most affected. Physical examination of the affected animals revealed proprioceptive d?ficit, motor incoordination, staggers and, neck and head intention tremors. ?Head-Raising Test? showed marked increase of head and limb tremors, with sequential difficulty in balance and tumble. Histopathology revealed severe distension of Purkinje cells, with foamy appearance, which eventually progressed to karyolysis and pyknosis, and intense vacuolization of pancreatic acinar and thyroid follicular cells. Additionally, there was vacuolization in neuronal bodies with dissolution of cytoplasm associated with marked proliferation of morphologically atypical astrocytes. Multiple axonal spheroids of variable sizes were noted. Positive reaction in lectin-histochemistry for lectins Con A, WGA, and sWGA was able to characterize the disease as a glycoproteinosis. Ultrastructural evaluation of pancreas sections evidenced numerous vacuoles up to 2,5?m in diameter, circled by 20nm wide membranes in acinar cells. In the present report, diagnosis of S. planicaulis toxicosis was established by epidemiological data, clinical and pathological findings, and confirmed by lectin-histochemistry and ultrastructural evaluation. Natural toxicosis in sheep by Sida planicaulis has not yet been described in the state of Rio de Janeiro.

A ingest?o de S. planicaulis Cav. (S. carpinifolia) ? respons?vel pelo aparecimento desta enfermidade em bovinos, equinos, caprinos, ovinos e cerv?deos cativos que ocorre, de forma natural, quando a planta predomina na pastagem e h? baixa oferta de outras forragens. O principal composto t?xico desta planta, a swainsonina, inibe atividade enzim?tica de ?-manosidase I e II que redunda no armazenamento de glicoprote?nas no interior de lisossomos. Este ac?mulo de subst?ncias promove compress?o, defici?ncia no transporte intracitoplasm?tico e finalmente degenera??o em diversos tecidos, dentre os quais o mais afetado ? o nervoso. O exame f?sico dos animais afetados revelou d?ficit proprioceptivo, incoordena??o motora, cambaleio ? movimenta??o e tremores de inten??o e na cabe?a. Houve aumento acentuado dos tremores em cabe?a e membros, com posterior dificuldade de se manter em esta??o e queda ao solo ap?s a realiza??o do ?Head-Raising Test?. A histopatologia evidenciou severa distens?o de neur?nios de Purkinje, com aspecto espumoso e eventual cari?lise ou cariopicnose e intensa vacuoliza??o de c?lulas acinares do p?ncreas e foliculares da tireoide. Adicionalmente, foram observadas vacuoliza??es em grandes corpos de neur?nios com dissolu??o do citoplasma associada ? marcada prolifera??o de astr?citos morfologicamente at?picos. Notou-se tamb?m m?ltiplos esferoides axonais de variados tamanhos. O exame lectino-histoqu?mico positivo para as lectinas Con A, WGA e sWGA foi capaz de caracterizar a enfermidade como uma glicoproteinose. A avalia??o ultraestrutural evidenciou numerosos vac?olos de at? 2,5?m de di?metro, delimitados por membranas de at? 20nm de espessura em c?lulas acinares do p?ncreas. O diagn?stico da intoxica??o por S. planicaulis foi estabelecido atrav?s dos dados epidemiol?gicos, achados cl?nico-patol?gicos e confirmados pelas avalia??es lectino-histoqu?mica e ultraestrutural. description This paper describes an outbreak of lysosomal storage disease in sheep induced by Sida planicaulis in the state of Rio de Janeiro. The consumption of S. planicaulis is responsible for this disease in cattle, horses, goat, sheep and captive deer, which occurs naturally, when the plant is abundant and there is shortage of other forage. The main toxic compound in this plant, Swainsonine, inhibits ?-manosidasis I e II enzymatic activitiy and causes glycoprotein storage in lysosomes. The storage provokes compression, deficiency in intracytoplasmatic transport, and finally, degenerative changes in various tissues, in which, nervous system is the most affected. Physical examination of the affected animals revealed proprioceptive d?ficit, motor incoordination, staggers and, neck and head intention tremors. ?Head-Raising Test? showed marked increase of head and limb tremors, with sequential difficulty in balance and tumble. Histopathology revealed severe distension of Purkinje cells, with foamy appearance, which eventually progressed to karyolysis and pyknosis, and intense vacuolization of pancreatic acinar and thyroid follicular cells. Additionally, there was vacuolization in neuronal bodies with dissolution of cytoplasm associated with marked proliferation of morphologically atypical astrocytes. Multiple axonal spheroids of variable sizes were noted. Positive reaction in lectin-histochemistry for lectins Con A, WGA, and sWGA was able to characterize the disease as a glycoproteinosis. Ultrastructural evaluation of pancreas sections evidenced numerous vacuoles up to 2,5?m in diameter, circled by 20nm wide membranes in acinar cells. In the present report, diagnosis of S. planicaulis toxicosis was established by epidemiological data, clinical and pathological findings, and confirmed by lectin-histochemistry and ultrastructural evaluation. Natural toxicosis in sheep by Sida planicaulis has not yet been described in the state of Rio de Janeiro.
