Resumo: Photodynamic Therapy (PDT) combined with the use of molecules containing the porphyrin ring associated with ruthenium transition metal has become an alternative method to the traditional treatment of melanoma. Here, we demonstrate DNA damage analyses, photocleavage properties and cytotoxicity of a free-based meso-terta-ruthenated porphyrin (H2RuTPyP). Cytotoxic and genotoxic potential of H2RuTPyP was investigated by colorimetric assay and by plasmid DNA photocleavage analysis followed by incubation with specific DNA repair enzymes, respectively. H2RuTPyP porphyrin efficiently induced DNA damage at the lowest concentration of 5.0 μM, resulting in its complete degradation at 15 μM. The presence of different scavengers for reactive oxygen species during exposure to visible light did not decrease the formation of DNA breaks, suggesting a hydrolytic mechanism for the breakdown of this molecule. In addition, the compound H2RuTPyP exhibited a higher cytotoxicity towards melanoma cell line compared to keratinocytes. Therefore, H2RuTPyP compound demonstrated to be a very promising effective photosensitizer, representing an alternative for the development of new treatments for melanoma by PDT.

Aqui, demonstramos as análises de dano ao DNA, propriedades de fotoclivagem e citotoxicidade de uma porfirina meso-tetra-rutenada de base livre (H2RuTPyP). O potencial citotóxico e genotóxico de H2RuTPyP foi investigado por ensaio colorimétrico de tetrazólio e pela análise de fotoclivagem de DNA plasmidial seguida de incubação com enzimas específicas de reparo de DNA, respectivamente. A porfirina H2RuTPyP induziu eficientemente danos ao DNA na concentração mais baixa de 5,0 μM, resultando em sua degradação completa a 15 μM. A presença de diferentes sequestradores para espécies reativas de oxigênio durante a exposição à luz visível não diminuiu a formação de quebras de DNA, sugerindo um mecanismo hidrolítico para a quebra desta molécula. Além disso, o composto H2RuTPyP exibiu citotoxicidade mais eleveda frente a linhagem celular de melanoma, em comparação com a de queratinócitos. Portanto, o composto H2RuTPyP motrou-se um fotossensibilizador eficaz muito promissor, representando uma alternativa para o desenvolvimento de novos tratamentos para o melanoma por meio da TFD.Coordenação de Aperfeiçoamento de Pessoal de Nível Superior - CAPESporUniversidade Federal de Santa MariaCentro de Ciências Naturais e ExatasPrograma de Pós-Graduação em Ciências Biológicas: Bioquímica ToxicológicaUFSMBrasilBioquímicaAttribution-NonCommercial-NoDerivatives 4.0 Internationalhttp://creativecommons.org/licenses/by-nc-nd/4.0/info:eu-repo/semantics/openAccessMelanomaTerapia fotodinâmicaPorfirina de rutênioGenotoxicidadePhotodynamic therapyRuthenium porphyrinGenotoxicityCNPQ::CIENCIAS BIOLOGICASDano ao DNA induzido por fotoclivagem e análise citotóxica de uma porfirina meso-tetra-rutenada sob irradiação por luz visívelDNA damage induced by photoclivage and cytoxotic analysis of a meso-tetra-rutenated porphyrin under visible light radiationinfo:eu-repo/semantics/publishedVersioninfo:eu-repo/semantics/doctoralThesisSchuch, André Passagliahttp://lattes.cnpq.br/4932611269622766Rodrigues Junior, Luiz CarlosRech, Virgínia CieloHörner, RosmariOliveira, Sara Marchesan dehttp://lattes.cnpq.br/7726613505527400Vizzotto, Bruno Stefanello200000000006600600600600600600600d93539a4-7b55-44e5-9c20-52601682b4f7e532cb9f-bc7c-466a-a578-2aeee155b18825a7c2c5-6e8f-4742-a3b0-98460d00e2c75223c71d-08cf-4fe9-a16b-f8577a94e351f4ee4a14-1e16-489c-a3ac-a234a7f2b6314075277b-8b36-46ac-b25d-abf48b5aa548reponame:Manancial - Repositório Digital da UFSMinstname:Universidade Federal de Santa Maria (UFSM)instacron:UFSMORIGINALTES_PPGCBBT_2020_VIZZOTTO_BRUNO.pdfTES_PPGCBBT_2020_VIZZOTTO_BRUNO.pdfTeseapplication/pdf2807339http://repositorio.ufsm.br/bitstream/1/22015/1/TES_PPGCBBT_2020_VIZZOTTO_BRUNO.pdf6f33b24f86c00dc26a76269c8be717b8MD51CC-LICENSElicense_rdflicense_rdfapplication/rdf+xml;
