Resumo: The organic solvents, also known as volatile organic compounds (VOCs) or volatile hydrocarbons, are widely used in a variety of industrial processes and consumer products. Gas station attendants (G.S.A.), as well as painters, are potentially exposed to these hydrocarbons such as benzene, toluene and xylene. Thus, the main of this work was to verify laboratory findings in workers exposed to these solvents. Blood and urine samples were collected to verify the biological indicators of exposure to these solvents, as well as inflammatory, genotoxicity and oxidative damage markers. The results showed an increase in malondialdehyde (MDA) levels in the G.S.A. group, while painters group presented an increase in nitric oxide (NO) levels. Among the genotoxicity markers, an increase in 8-hydroxydeoxyguanosine (8-OHdG) levels in G.S.A. and painters groups was observed and, in this last group, the increase was even greater. The G.S.A. group presented significant DNA damage compared to the control group, which was demonstrated by the comet assay. Among the inflammatory markers, interleukin-1 (IL-1), interleukin-6 (IL-6), tumor necrosis factor alpha (TNF-α) and interferon gamma (IFN-γ), the levels were increased in both groups compared with the control group, and painters showed higher levels of IFN-γ compared to the G.S.A. group. Interleukin-10 (IL-10) levels were significantly reduced in both groups of workers exposed, when compared to the unexposed group. Moreover, a significant increase was observed in alanine aminotransferase (ALT) activity in painters. Hematological parameters were changed in painters, since there was a reduction in platelet count and increase in mean corpuscular hemoglobin (MCH). The biological indicators of exposure to these solvents, trans, trans muconic acid (t, t mA) for benzene, hippuric acid (HA) for toluene and metilhipuric acid (mHA) for xylene, were not modified in G.S.A. and painters compared to control group. Despite these indicators remained within the maximum permissible values, significant changes were observed in other markers, revealing mainly changes in the specific markers for inflammation and genotoxicity.

Frentistas, assim como pintores, estão potencialmente expostos a estes hidrocarbonetos, tais como benzeno, tolueno e xileno. Assim, o objetivo deste trabalho foi verificar os achados laboratoriais em trabalhadores expostos a estes solventes. Amostras de sangue e urina foram recolhidas para verificar os indicadores biológicos da exposição a estes solventes, bem como marcadores de processo inflamatório, genotoxicidade e dano oxidativo. Os resultados demonstraram um aumento nos níveis de malondialdeído (MDA) no grupo de frentistas, enquanto que o grupo de pintores apresentou um aumento nos níveis de óxido nítrico (ON). Entre os marcadores de genotoxicidade, foi observado um aumento nos níveis de 8-hidroxideoxiguanosina (8-OHdG) no grupo de frentistas e nos pintores, sendo que, neste último grupo, o aumento foi ainda maior. O grupo de frentistas também apresentou dano significativo ao DNA com relação ao grupo controle, que foi demonstrado pelo teste do cometa. Entre os marcadores inflamatórios, a interleucina-1 (IL-1), a interleucina-6 (IL-6), fator de necrose tumoral alfa (TNF-α) e interferon gama (IFN-γ), os níveis mostraram-se aumentados em ambos os grupos, em comparação com o grupo controle, sendo que pintores apresentaram maiores níveis de IFN-γ em comparação aos frentistas. Os níveis de interleucina 10 (IL-10) foram reduzidos significativamente em ambos os grupos de trabalhadores expostos, quando comparados ao grupo não exposto. Além disso, foi observado um aumento significativo na atividade da alanina aminotransferase (ALT) em pintores. Os parâmetros hematológicos foram alterados nos pintores, que apresentaram uma redução na contagem de plaquetas e um aumento da hemoglobina corpuscular média (HCM). Os indicadores biológicos de exposição a estes solventes, ácido trans, trans mucônico (t, t ma) para o benzeno, ácido hipúrico (AH) para o tolueno e ácido metilhipúrico (mHA) para o xileno, não apresentaram alterações significativas nos frentistas e pintores em relação aos controles. Apesar destes indicadores mantiveram-se dentro dos valores máximos admissíveis, mudanças expressivas foram observadas em outros marcadores, revelando principalmente alterações nos marcadores específicos para a inflamação e genotoxicidade.porUniversidade Federal de Santa MariaCentro de Ciências da SaúdePrograma de Pós-Graduação em Ciências FarmacêuticasUFSMBrasilAnálises Clínicas e ToxicológicasAttribution-NonCommercial-NoDerivatives 4.0 Internationalhttp://creativecommons.org/licenses/by-nc-nd/4.0/info:eu-repo/semantics/openAccessExposição ocupacionalSolventes orgânicosBenzenoToluenoXilenoOccupational exposureOrganic solventsBenzeneTolueneXyleneCNPQ::CIENCIAS DA SAUDE::FARMACIAAvaliação de diferentes marcadores de dano oxidativo, genotoxicidade e inflamação em trabalhadores expostos a solventes no município de Santa Maria, RSEvaluation of different markers of oxidative damage, genotoxicity and inflammation in workers exposed to solvents in Santa Maria, Rio Grande do Sul, Brazilinfo:eu-repo/semantics/publishedVersioninfo:eu-repo/semantics/masterThesisSilva, José Edson Paz dahttp://lattes.cnpq.br/9154529811247631Luchese, Cristianehttp://lattes.cnpq.br/3420684025232526Brucker, Natáliahttp://lattes.cnpq.br/7188237428821146http://lattes.cnpq.br/7223209525186178Silva, Lilian Marquezini da400300000005600f31cbb2c-a6b3-42b5-99ec-4f7ceeff614960d85652-3b42-4995-baec-771abe4e4a3b6394c49e-1d1b-44ff-89fb-e3d4c9a0f1317e1d71bf-4010-48ae-bd49-256dee844c6dreponame:Manancial - Repositório Digital da UFSMinstname:Universidade Federal de Santa Maria (UFSM)instacron:UFSMORIGINALDIS_PPGCF_2015_SILVA_LILIAN.pdfDIS_PPGCF_2015_SILVA_LILIAN.pdfDissertação de Mestradoapplication/pdf1348905http://repositorio.ufsm.br/bitstream/1/17541/1/DIS_PPGCF_2015_SILVA_LILIAN.pdfd701a781f73a8a00c5f2d784fdc9be00MD51CC-LICENSElicense_rdflicense_rdfapplication/rdf+xml;
