Syzygium cumini no diabetes mellitus iatrogênico em ratos: prevenção e tratamento

Resumo: The objective of the present study was to investigate the possible antidiabetic properties of Syzygium cumini. The plant is often used in popular medicine. The first experiment evaluated the efficiency of the prophylactic use of Syzygium cumini against the diabetogenic effect of alloxan in rats. Syzygium cumini was offered as tea, prepared with plant leaves, at the concentration of 20 g/l. Animals received the tea as a substitute for water for 16 days before induction and for the following 3 days of the induction protocol. Fifteen female rats received the tea and another equally numbered group served as control, receiveing water. At days 17, 18 and 19 of the study diabetes was induced by daily administration of 60 mg/Kg of alloxan in all animals. The ingestion of the tea during 16 days had a hyperglycemic effect. Four days after the last dose of alloxan the plasma glucose levels of Syzygium cumini pretreated animals increased 48,86% in relation to pre-induction values. In the control group the post-induction increase was 177,96%. Additionally, pretreatment with the tea prevented the weight loss observed in the control group. These results indicate a protective effect of Syzygium cumini against diabetes mellitus induced by alloxan in rats. The second study was conducted to assess the antihyperglycemic effect of Syzygium cumini in an experimental diabetes model. The tea was prepared from leaves of Syzygium cumini, in the concentration of 200 g/l and administrated in a single dose of 25 ml/kg of body weight in alloxan-induced diabetic rats. The animals were submitted to an oral glucose tolerance test, one hour after tea ingestion. The tea ingestion improved glucose tolerance, decreasing acute hyperglycemia starting at 60 minutes after ingestion of glucose and contributing to the return to fasting levels, after 120 minutes of glucose load. These results demonstrate the antihyperglycemic effect of Syzygium cumini in diabetic rats, suggesting its potential use on the control of postprandial hyperglycemia in patients with diabetes mellitus type 1.

Em um primeiro experimento, a influência da planta sobre a eficiência diabetogênica do aloxano foi avaliada. Syzygium cumini foi oferecido na forma de chá, preparado a partir das folhas da planta, na concentração de 20g/l, como substituto da água, antes e durante os dias da administração do aloxano, para um grupo de 15 ratas adultas. Durante este período, outro grupo, numericamente igual, de ratas recebeu água como única fonte hídrica. Nos dias 17, 18 e 19 do experimento foi realizada a indução do diabetes mellitus, através da administração diária de 60 mg/Kg de aloxano, em todos os animais. A ingestão ad libitum da planta demonstrou, ao final de 16 dias de tratamento, ter um efeito hiperglicemiante. No quarto dia após a última dose de aloxano, os níveis glicêmicos dos animais pré-tratados com Syzygium cumini sofreram uma elevação de 49,86% em relação ao período anterior à indução, enquanto que esta taxa atingiu um índice de 177,96 % no grupo controle, tratado somente com aloxano. Além disso, o tratamento prévio com a planta evitou a perda de peso corporal, apresentada pelo grupo controle. Esses resultados indicam o efeito protetor do Syzygium cumini contra diabetes mellitus induzido por aloxano em ratos. Em um segundo experimento, foi avaliado o efeito anti-hiperglicemiante do chá de Syzygium cumini em um modelo experimental de diabetes. O chá foi preparado a partir das folhas Syzygium cumini, na concentração de 200 g/l, e administrado, na dose única de 25 ml /kg de peso corporal, para ratos com diabetes mellitus induzido por aloxano. Uma hora após terem ingerido a planta, os animais foram submetidos a um teste de tolerância oral à glicose. A ingestão da planta minimizou a flutuação na glicemia decorrente da carga de glicose, diminuindo a hiperglicemia aguda a partir dos 60 minutos após o desafio e favorecendo o seu retorno para níveis próximos aos de jejum, após ter decorrido 120 minutos do desafio.
