Resumo: | Ceratocystia fimbriata Ellis & Halsted is a pathogen responsible for the disease wilt ceratocistis and has caused losses in many important crops in Brazil, such as Eucalyptus spp., Ficus carica, Mangifera indica, Hevea brasiliensis, Gmelina arborea and Acacia mearnsii. Ceratocystis fimbriata s.l. currently considered a "species complex" containing multiple cryptic species, many of which are not yet described. Thus, the objective of this study was to identify the species of the genus Ceratocystis associated with the disease Ceratocystis wilt in kiwi (Actinidia deliciosa) correlating morphological characteristics and phylogenetic isolates of C. fimbriata causing wilt ceratocysti in kiwifruit in Brazil with brazilian isolates from different hosts and others belonging to the clades of North America and Asia. To study whether there is host specialization, through inoculations at different eucalyptus clones and cultivars of kiwifruit, and identify eucalyptus clones and kiwi cultivars resistant. For this, samples were collected from soil and plant material of plants of kiwi with symptoms typical of the disease. To obtain the isolates, we used the method bait carrot, then immediately was obtained culture monoaspórica. Were performed phylogenetic analysis, morphological identification, mycelial growth at different temperatures, host specialization. The causal agent of wilt kiwi is C. fimbriata, which belongs to clade Latin America. The ideal temperature for development is 20-250C. It did not occur to the host specialization of C. fimbriata isolates studied. The kiwifruit cultivars are susceptible to C. fimbriata. Eucalyptus dunnii and clone 32864 were highly resistant to C. fimbriata. |

Ceratocystis fimbriata é considerado, atualmente, como um “complexo de espécies” contendo várias espécies crípticas, muitas das quais ainda não descritas. Deste modo, o objetivo neste trabalho foi identificar as espécies do gênero Ceratocystis associadas à doença murcha-de-ceratocystis em de kiwi (Actinidia deliciosa), relacionando características morfológicas e molecular, com isolados de diferentes hospedeiros brasileiros e outros pertencente ao clados da América do Norte e Ásia, também estudar se existe especialização por hospedeiro, por meio de inoculações em diferentes clones de eucalipto e Eucalyptus dunnii e cultivares de kiwi, e identificar materiais genéticos de eucalipto e cultivares de kiwi resistentes. Para isso, foram coletadas amostras de solo e material vegetal de plantas de kiwi que apresentavam sintomas típicos da doença. Para a obtenção dos isolados, utilizou-se o método isca de cenoura, logo em seguida, foi obtida a cultura monoascospóricas. Foram realizadas análises de molecular e morfológica, crescimento micelial em diferentes temperaturas, especialização por hospedeiro. O agente causal da murcha-de-kiwi é Ceratocystis fimbriata, pertencente ao clado da América Latina. A temperatura ideal para o seu desenvolvimento é 20-250C e não ocorreu especialização por hospedeiro para os isolados de Ceratocystis fimbriata estudados. As cultivares de kiwi são suscetíveis e Eucalyptus dunnii e o clone 32864 apresentaram-se altamente resistentes a C. fimbriata.Coordenação de Aperfeiçoamento de Pessoal de Nível Superior - CAPESporUniversidade Federal de Santa MariaCentro de Ciências RuraisPrograma de Pós-Graduação em Engenharia FlorestalUFSMBrasilRecursos Florestais e Engenharia FlorestalAttribution-NonCommercial-NoDerivatives 4.0 Internationalhttp://creativecommons.org/licenses/by-nc-nd/4.0/info:eu-repo/semantics/openAccessMurcha-de-ceratocystisKiwiResistênciaEpidemiologiaActinidia deliciosaEucalyptus sp.Ceratocystis wiltResistanceEpidemiologyCNPQ::CIENCIAS AGRARIAS::RECURSOS FLORESTAIS E ENGENHARIA FLORESTALMorfologia, caracterização molecular e patogenicidade de Ceratocystis fimbriata no Rio Grande do SulMorphology, molecular characterization and pathogenicity of Ceratocystia fimbriata in Rio Grande do Sulinfo:eu-repo/semantics/publishedVersioninfo:eu-repo/semantics/doctoralThesisMuniz, Marlove Fatima Briãohttp://lattes.cnpq.br/3148312031889388Alfenas, Acelino CoutoManfio, Candida ElizaMenezes, Josiane PachecoBrackmann, Aurihttp://lattes.cnpq.br/7128401163875834Piveta, Graziela500200000003600600600600600600600085fc8dc-a963-42f9-b156-961f0fc14b447169e32c-4723-445e-a190-da3345a0ae2c59b697be-7139-410f-8109-bca9a0af56286a7f33e0-caaa-4eba-8c4c-4790f7dce0870b518a78-8366-4cf7-aebd-cc7a2f1ea1bde6d627e7-3660-4771-a64c-9e3da0e009e1reponame:Biblioteca Digital de Teses e Dissertações do UFSMinstname:Universidade Federal de Santa Maria (UFSM)instacron:UFSMCC-LICENSElicense_rdflicense_rdfapplication/rdf+xml;
