Resumo: | Clinical, epidemiological and experimental evidences certify the role of trans fatty acids in the genesis of non-transmissible chronic diseases. The atherogenic effects of trans fatty acids produced by industrialization process are well known in adult subjects. However, little is known about its effects on maternal-infantile and infantile-juvenile populations. Therefore, the objective of this study was to evaluate the effects of trans fatty acid intake on blood lipid profile and its deposition in the liver, adipose tissue and serum of Wistar rats after birth, weaning and growth states. Hence, 40 female rats and their male offspring were used, 20 newborn, 20 weaning and 20 young adults were separated into control and trans groups in each phase. The animals during pregnancy and during growth of both groups received AIN-93 diet with modifications. The source of trans fatty acids was hydrogenated vegetable fat at 5% of the energy of the diet. After birth, weaning and growth, the rats were euthanized and the abdominal adipose tissue, liver and serum were collected. It was determined the lipid profile by gas chromatography after lipid extraction by direct methylation. It was analyzed the levels of serum total cholesterol, HDL-cholesterol, triacylglycerol and glucose as well as the liver morphometric analysis and hepatic- somatic index. The data were analyzed by t-test of Student and analysis of variance ANOVA, and the Pearson correlation to evaluate the relation between essential fatty acids. The results showed that trans fatty acid accumulated in higher proportion in the liver and adipose tissue of the newborn rats (P<0.05). However, the accumulation of trans fatty acids in the liver did not affect the synthesis of long chain fatty acids, probably due to the adequacy of the requirement of essential fatty acids. There was negative and significant correlations (P<0.001) between trans fatty acids and linoleic acid in the adipose tissue of weaning and young adult rats, and between linolenic and aracdonic acid in the adipose tissue of young adult rats. Fasting blood glucose was higher in young adult rats fed trans fatty acids than the control (P=0.004). The liver morphometric analysis showed higher lipid accumulation in rats fed trans fatty acids than the control group in all phases, and the difference was significant between newborn and weaning rats (P<0.01). The hepatic-somatic index showed significant difference between groups in the young adult rats (P<0.05). In conclusion, trans fatty acids accumulated in the adipose tissue, liver and serum at different ways in the analyzed phases, and promoted hyperglycemia in young adult rats and higher accumulation of lipids in the liver of newborn rats. |

Evidências científicas comprovam, através de dados clínicos, epidemiológicos e experimentais, o risco do consumo de ácidos graxos trans para a saúde. Dessa forma, os apelos atuais levam a determinar a diminuição dos ácidos graxos trans nos produtos industrializados. Discussões são realizadas sobre rotulagem dos alimentos, diferentes efeitos à saúde provocados pelos ácidos graxos trans industrializados e naturais, bem como aspectos sobre a qualidade e custos para remoção desses ácidos graxos. Conhecem-se os ácidos graxos trans, produtos da hidrogenação de óleos ou da biohidrogenação de ruminantes, pelos seus efeitos aterogênicos em indivíduos adultos. Entretanto, pouco se sabe sobre seus efeitos na população materno-infantil e infanto-juvenil. Dessa forma, objetivando avaliar os efeitos do consumo de ácidos graxos trans no perfil lipídico sangüíneo e sua deposição no fígado, tecido adiposo e soro de ratos machos Wistar em três fases da vida (recém-nascidos, desmamados e adultos-jovens), foi desenhado este estudo. Para tal, foram utilizadas 40 ratas Wistar prenhas e suas crias, 20 ratos machos recém-nascidos, 20 desmamados e 20 adultos-jovens, separados em grupo controle e trans, em cada fase. As ratas prenhas e ratos adultos-jovens do grupo controle e trans receberam dieta AIN 93 com modificações. Como fonte de ácidos graxos trans foi utilizada a gordura vegetal hidrogenada, correspondente a aproximadamente 5% do valor calórico total da dieta. Após o nascimento, desmame e crescimento (fase adulto-jovem) os ratos foram submetidos à eutanásia e o tecido adiposo abdominal, fígado e soro foram extraídos. Determinou-se o perfil lipídico por cromatografia gasosa. Foram realizadas determinações no soro de: colesterol total, HDL , triglicerídeos e glicemia de jejum, bem como, análises morfométrica do fígado e determinação do índice hepatossomático. Os dados foram analisados pelos testes t de Student e análise de variância ANOVA, sendo utilizado correlações de Pearson para avaliar as relações entre ácidos graxos trans e ácidos graxos essenciais. Os resultados mostraram que no fígado e tecido adiposo os ácidos graxos trans acumularam em maior proporção nos ratos recém-desmamados, com diferença significante (P<0,05). Entretanto, esse acúmulo de ácidos graxos trans no fígado, não provocou alteração na síntese de ácidos graxos de cadeia longa, provavelmente em função do atendimento das necessidades de ácidos graxos essenciais. Houve, também, correlações negativas e significantes (P< 0,001) entre os isômeros trans e ácido graxo linoléico no tecido adiposo dos ratos recém-desmamados e adultos-jovens, e entre ácidos graxos linolênico e araquidônico no tecido adiposo dos ratos adultos-jovens. A glicemia de jejum foi maior em ratos adultos-jovens do grupo trans do que nos ratos do grupo controle (P=0,004). A análise morfométrica do fígado apresentou maior acúmulo de gordura nos ratos do grupo trans do que no grupo controle nas três fases, com diferenças significativas entre os grupos recém-nascidos e recém-desmamados (P< 0,001). O índice hepatossomático apresentou diferença significante entre os grupos, nos ratos adultos-jovens (P<0,05).
