Resumo: | This study aimed to evaluate the effects of CLA and/or vitamin E supplementation in the progression of atherosclerosis and in the fatty acids profile of tissues of LDLR -/- mice, a model for familial hypercholesterolemia. The animals were fed with a semi purified diet (CT) or with an atherogenic diet (AT), during six weeks. After this period, in order to evaluating the atherosclerosis development degree, a group of animals was euthanized. The other animals were fed during eight weeks later with a semi purified diet supplemented with 1% of vitamin E (TOC), 1% of 10t,12c- CLA (CLA) or with 1% of vitamin E + 1% of 10t,12c-CLA (CLA/TOC). The weight and the diet consumption were monitored weekly and daily, respectively. Fatty acid serum profile was determined, as well as liver and feces concentrations of α-tocopherol and cholesterol. Peroxidation in liver was evaluated by hydroperoxides analysis. The fatty acids profiles of the liver, adipose tissue and small intestine were analyzed by gas chromatography. The total area of the atherosclerotic lesion and the aorta obstruction percentile were estimated. Serum lipids profile showed total cholesterol, LDL and triacylglycerols smaller concentrations in the supplemented with vitamin E groups. In these groups the cholesterol excretion and the hepatic α-tocopherol concentrations were larger than in others. However, in the group that received with vitamin E only, was found the biggest degree of hepatic lipids peroxidation. The fatty acids profiles of tissues reflected the dietary fatty acids composition. Althought, in none tissues analyzed was possible to identify the incorporation of 10t,12c-CLA. Probably the CLA was not detected due to its rapidly metabolization. In all of the experimental groups, the polyunsaturated fatty acids percentage in the adipose tissue and intestine was larger than in CT group. The use of 10t,12c-CLA isomer individually did not impede the atherosclerotic lesions development in the aorta. On the other hand, the dietary supplementation with vitamin E, as well as the vitamin E and 10t,12c-CLA association significantly reduced the total area of the atherosclerotic lesion and the aorta obstruction percentile. Showing the better action of vitamin E than 10t,12c-CLA in reduce the atherosclerosis progression, despite their antioxidant effect. |

Os animais foram alimentados com dieta semipurificada (CT) ou com dieta aterogênica (AT), durante seis semanas. Após este período, um grupo sofreu eutanásia para avaliação do grau de desenvolvimento da aterosclerose e os outros animais foram alimentados por oito semanas com dieta semipurificada suplementada com 1% de vitamina E (TOC), 1% de 10t,12c-CLA (CLA) ou com 1% de vitamina E + 1% de 10t,12c-CLA (CLA/TOC). O peso dos animais foi monitorado semanalmente e o consumo alimentar diariamente. O perfil de lipídios séricos foi determinado, bem como as concentrações de α-tocoferol e de colesterol no fígado e nas fezes. A peroxidação lipídica no fígado foi determinada pela análise de hidroperóxidos. Os perfis de ácidos graxos do fígado, tecido adiposo e intestino delgado foram analisados por cromatografia gasosa. Avaliou-se a área total da lesão aterosclerótica e o percentual de obstrução da aorta dos diferentes grupos experimentais. As concentrações séricas de colesterol total, de LDL e de triacilgliceróis foram menores nos grupos suplementados com vitamina E. Nestes grupos, a excreção de colesterol foi maior que nos demais e também foram observadas as maiores concentrações hepáticas de α-tocoferol. Por outro lado, no grupo suplementado com vitamina E foi encontrado o maior grau de peroxidação dos lipídios hepáticos. Os perfis de ácidos graxos dos tecidos refletiram a composição de ácidos graxos das dietas. Entretanto, em nenhum dos tecidos analisados foi possível identificar a incorporação do 10t,12c-CLA. Aparentemente, o CLA não foi detectado em função da sua rápida metabolização. Em todos os grupos experimentais, os percentuais de ácidos graxos poliinsaturados no tecido adiposo e no intestino, foram maiores em relação ao grupo CT. A utilização isolada do isômero 10t,12c-CLA não impediu o desenvolvimento de lesões ateroscleróticas na aorta. Por outro lado, a suplementação dietética com vitamina E, assim como a associação de vitamina E e 10t,12c-CLA, reduziram significantemente a área total da lesão e o percentual de obstrução da aorta.
