Resumo: | In the world, Brazil stands out as a major producer and consumer in the world of common bean, Phaseolus vulgaris, however has low productivity due to several factors among them, low technological level used in the cultivation and lack of care mainly plant grain during the storage of . Large losses in the storage of bean (P. vulgaris) are attributed to grain legumes weevil causing the decline of their nutritional value. The main enzymes related to energy and protein metabolism of bruchid are serine and cysteine proteases and α-amylase. The different levels of activities of digestive enzymes suggested a higher metabolic efficiency. Therefore, the process of resistance may be attributed to better adaptation to the natural inhibitors present in the plant leading to reproductive success and perpetuation of the species. Here, we report a phylogeography study that investigated the genetic diversity and an enzyme study that examined the energy metabolism in of Acanthoscelides obtectus. The phylogeography study was based on two genes of mitochondrial genome, COI and rRNA16S, which revealed that populations from either Mexico or Peru consisted of one lineage each. Brazilian populations contained two distinct mitochondrial lineages, both with low genetic diversity, and possibly originated from the Mesoamerican lineage. Probably these lineages came to Brazil only once forming two ancestral lines. Alternatively, they were introduced repeatedly from the same parental source. One of the lines is geographically restricted to South Brazil, whereas the other lineage is widespread all over Brazil. Comparative analysis of the proteolytic and amidolytic activity of cysteine and serineprotease between four study populations (Sobradinho, Viçosa, Cacoal, and Bom Jesus do Amparo) revealed Sobradinho with a higher activity of serine and cysteine proteases. Most likely, Sobradinho exhibited more efficient metabolism in relation to the protease inhibitors present in seeds. The population Viçosa showed higher activity of amylase followed by Bom Jesus do Amparo, while Cacoal and Sobradinho had lower activity of amylase and did not differ statistically. |

The population Viçosa showed higher activity of amylase followed by Bom Jesus do Amparo, while Cacoal and Sobradinho had lower activity of amylase and did not differ statistically.No cenário mundial, o Brasil destaca-se como grande produtor e consumidor mundial do feijão comum, Phaseolus vulgaris, entretanto apresenta baixa produtividade devido a alguns fatores dentre eles, baixo nível tecnológico empregado no cultivo e a falta de cuidados fitossanitários principalmente durante o armazenamento dos grãos. Grandes perdas no armazenamento de feijão (P. vulgaris) são atribuídas aos carunchos de grãos leguminosos causando a diminuição do seu valor nutricional. As principais enzimas relacionadas ao metabolismo protéico e energético dos bruquídeos são as serino e cisteíno proteases e a α-amilase. Os níveis diferenciados de atividades destas enzimas digestivas sugeriram uma maior eficiência metabólica. Portanto, o processo de resistência pode ser atribuído a uma melhor adaptação aos inibidores naturais presentes na planta levando ao sucesso reprodutivo e à perpetuação da espécie. No intuito de conhecer a diversidade genética e algumas características enzimáticas de populações do Acanthoscelides obtectus, foram realizados estudo filogeográfico e das enzimas do metabolismo protéico e energético deste inseto. O estudo filogeográfico foi baseado em dois genes do DNA mitocondrial, COI e rRNA16S, os quais permitiram inferir que os haplótipos oriundos das populações mexicanas e peruanas formaram duas linhagens independentes, ambas com alta diversidade genética. Nas populações brasileiras foram identificadas duas linhagens mitocondriais distintas, ambas com baixa diversidade genética e, possivelmente, originadas da linhagem mesoamericana. Provavelmente estas linhagens chegaram ao Brasil apenas uma única vez formando duas linhagens ancestrais ou alternativamente, existiram dois eventos distintos de introdução a partir de uma mesma fonte. Uma das linhagens está geograficamente restrita à região sul e a outra está distribuída por todo o Brasil. A análise da atividade proteolítica de cisteíno e serino-proteases e atividade amidolítica do caruncho revelou que entre as quatro populações estudadas (Sobradinho, Viçosa, Cacoal e Bom Jesus do Amparo), a Sobradinho apresentou maior atividade de serino e cisteínoproteases quando comparada às demais populações. Provavelmente, Sobradinho apresenta metabolismo mais eficiente frente aos inibidores de proteases presentes nas sementes de feijão.
