Caracterização bioquímica e funcional de fatores de transcrição envolvidos na resposta a estresses biótico e abiótico em soja

Resumo: | Responses to environmental stress in plants lead to a dramatic reprogramming of gene expression favoring stress responses at the expense of normal cellular functions. Transcription factors are master regulators of gene expression at the transcriptional level and control the activity of these factors alters the transcriptome of the plant, which leads to metabolic and phenotypic changes in response to stress. Functional analysis of transcription factors is very important to elucidate the role of these transcriptional regulators in different signaling cascades. In this study, we describe the functional characterization of important transcription factors in the molecular responses during environmental stresses, such as reticulum and osmotic stress, and during pathogen attack. Here, in the first chapter, we describe a novel transcription factor, GmERD15 (Glycine max Early Responsive to Dehydration 15), which is induced by ER stress and osmotic stress to activate the expression of NRP (Asparagine-rich protein) genes. In the second chapter, we describe the identification of four bZIP genes in soybean, GmbZIP62, GmbZIP105, GmbZIPE1 e GmbZIPE2, responsive to Phakopsora pachyrhizi, the Asian Soybean Rust causing agent. The understanding of these transcription factors functions in these response signaling cascades is crucial to the management of stress tolerance in crops, and will eventually lead to the development of genetically manipulated crop varieties with improved stress tolerance. |

Fatores de transcrição são reguladores fundamentais da expressão gênica a nível transcricional, e o controle de suas atividade altera o transcriptoma da planta, determinando mudanças metabólicas e fenotípicas em resposta ao estresse. Análises funcionais de fatores de transcrição são, portanto, muito importantes para a elucidação do papel destes reguladores transcricionais em diferentes cascatas de sinalização. Neste estudo, nós descrevemos a caracterização funcional de fatores de transcrição nas respostas moleculares durante estresses ambientais, tais como estresses no retículo e estresse osmótico, e durante ataque de patógenos. No primeiro capítulo, nós descrevemos um novo fator de transcrição, GmERD15 (Glycine max Early Responsive to Dehydration 15), o qual é induzido por estresse no retículo e osmótico para ativar a expressão de genes NRP (Asparagine-rich protein). No segundo capítulo, descrevemos a identificação de quatro fatores bZIP de soja, GmbZIP62, GmbZIP105, GmbZIPE1 e GmbZIPE2, responsivos à infecção por Phakopsora pachyrhizi, agente causador da ferrugem asiática.
