Resumo: The common bean (Phaseolus vulgaris L.) is a major component of the diet in Brazil and an important primary source of protein and iron. Brazil is the largest common bean producer and the yield of this crop in the country could be higher if it were not, among other reasons, for the numerous diseases affecting this crop. Among the fungal diseases affecting the aerial portion of the plant are the angular leaf spot, caused by Pseudocercospora griseola, and anthracnose, caused by Colletotrichum lindemuthianum. These diseases can cause losses of more than 70% depending on weather conditions, degree of resistance of the cultivar and pathogenicity of the fungal isolates. Understanding how plants respond to pathogen attack is of fundamental importance for the development of resistant cultivars that can ensure the sustainability of agricultural production. This work aimed to identify proteins differentially expressed by the common bean genotype AND 277 in response to P. griseola and C. lindemuthianum. Two independent experiments were conducted. In the first one, leaf samples were collected and evaluated 12, 24 and 48 hours after inoculation (h.a.i.) with P. griseola race 31.31. In the second one, leaf samples were collected and evaluated 12 and 48 h.a.i. of C. lindemuthianum race 73 of. Protein samples were extracted by the phenol-SDS method and separated by two-dimensional electrophoresis. Proteins differentially expressed between inoculated and non-inoculated plants were excised from the gels, cleaved with trypsin and analyzed by mass spectrometry. In the first experiment, 13 differentially expressed proteins were identified, five were detected only in inoculated plants, three showed increased expression and five had reduced expression upon inoculation. These proteins are associated with the defense process, hormone synthesis, photosynthesis, metabolism of reactive oxygen species, cellular respiration and porphyrin synthesis. In the second experiment, 35 differentially expressed proteins were identified, 13 of which were present only in inoculated plants and six only in non- inoculated plants, 10 had increased expression and six were reduced upon inoculation. These proteins participate in cellular detoxification processes, defense against pathogens, metabolism of reactive oxygen species, photosynthesis, cellular respiration, protein biosynthesis and signaling. These results contribute for a better understanding of the defense response of common bean to these pathogens. The identified proteins and their corresponding genes (candidate genes) can be used as tools breeding programs to develop resistant plants.

O Brasil é o maior produtor dessa leguminosa. Um dos fatores que limitam a produtividade brasileira é o grande número de doenças. Dentre as doenças fúngicas da parte aérea destacam-se a mancha angular causada por Pseudocercospora griseola e a antracnose, causada por Colletotrichum lindemuthianum. Essas doenças podem causar perdas superiores a 70% dependendo das condições climáticas, suscetibilidade da cultivar e patogenicidade dos isolados. A compreensão de como as plantas respondem ao ataque de patógenos é de fundamental importância para o desenvolvimento de cultivares resistentes que possam garantir a sustentabilidade da produção agrícola. Com este trabalho objetivou-se identificar proteínas diferencialmente expressas pelo genótipo de feijoeiro comum AND 277 em resposta a P. griseola e C. lindemuthianum. Foram realizados dois experimentos independentes. No primeiro, foram avaliadas amostras de folhas coletadas 12, 24 e 48 horas após a inoculação (h.a.i.) da raça 31.31 de P. griseola. No segundo, foram avaliadas amostras de folhas coletadas 12 e 48 h.a.i. da raça 73 de C. lindemuthianum. Amostras de proteínas foram extraídas pelo método fenol-SDS e separadas por eletroforese bidimensional. Proteínas diferencialmente expressas entre plantas inoculadas e não inoculadas com o patógeno foram excisadas dos géis, clivadas com tripsina e analisadas por espectrometria de massa. No primeiro experimento, foram identificadas 13 proteínas diferencialmente expressas, sendo que cinco foram detectadas somente nas plantas inoculadas, três apresentaram aumento de expressão e cinco tiveram redução da expressão após inoculação. Essas proteínas estão associadas a processos de defesa, síntese de hormônios, fotossíntese, metabolismo de espécies reativas de oxigênio, respiração celular e síntese de porfirinas. No segundo experimento, foram identificadas 35 proteínas diferencialmente expressas, sendo que 13 estavam presentes somente nas plantas inoculadas e seis somente nas plantas não inoculadas, 10 proteínas tiveram aumento de expressão e seis tiveram redução após a inoculação. Essas proteínas participam de processos de detoxificação celular, defesa contra patógenos, metabolismo de espécies reativas de oxigênio, fotossíntese, respiração celular, biossíntese de proteínas e sinalização. Esses resultados contribuem para um melhor entendimento do mecanismo de resposta de defesa do feijoeiro comum a esses patógenos.
