Resumo: The present study aimed to evaluate Eucalyptus urophylla x E. globulus and Eucalyptus grandis x E. globulus clones in relation to micropropagation by axillary shoot proliferation in the in vitro establishment and multiplication phases, and the rooting of mini-cuttings in the clonal propagation of these hybrids. In the in vitro establishment phase, contamination by fungi and bacteria, browning and nodal segment shoot inductions of 21 Eucalyptus urophylla x E. globulus clones and eight Eucalyptus grandis x E. globulus clones were evaluated in different in vitro introductions. In the in vitro multiplication phase, the multiplication rate of 21 Eucalyptus urophylla x E. globulus clones and six Eucalyptus grandis x E. globulus clones was evaluated in successive subcultures using MS and JADS culture media in different in vitro introductions. In the mini- cuttings, the survival and emission of roots at the bottom of the dibble tube in the green house and shade house was evaluated, and the rooting and number of roots of apical and intermediary mini-cuttings in the shade house, treated with IBA, in nine Eucalyptus urophylla x E. globulus clones and two Eucalyptus grandis x E. globulus clones. In the in vitro establishment phase, shoot induction varied among clones, being satisfactory for most of the genotypes, while the browning rate and bacterial and fungal contamination did not compromise the establishment of the cultures. In the in vitro multiplication phase, a differentiated response for the multiplication rate was noted, with clones presenting good in vitro multiplication rates and recalcitrant others for micropropagation. The MS and JADS culture media had positive effects, with greater multiplication rate tendencies for the MS media, where the multiplication rate of the subcultures varied with the tendency of an initial increase and subsequent fall in most of the clones and in vitro introductions. As for the mini-cuttings, there was no significant effect of IBA in any of the studied characteristics. The survival rate of the mini-cuttings was high for most of the clones and the rooting rate varied among clones and the types of mini-cuttings with superior results for apical mini- cuttings. A high variability of responses to rooting was observed among the studied clones, with clones presenting a high rate of rooting and number of roots for minicuttings, while some clones still lacked adjustments to the mini- cuttings propagation process.

Na fase de estabelecimento in vitro, foram avaliadas a contaminação por fungos e bactérias, oxidação e a indução de brotações em segmentos nodais de 21 clones de Eucalyptus urophylla x E. globulus e oito clones de Eucalyptus grandis x E. globulus em diferentes introduções in vitro. Na multiplicação in vitro, avaliou-se a taxa de multiplicação de 21 clones de Eucalyptus urophylla x E. globulus e seis clones de Eucalyptus grandis x E. globulus em sucessivos subcultivos nos meios de cultura MS e JADS, em diferentes introduções in vitro. Na miniestaquia, avaliou-se a sobrevivência e a emissão de raízes no fundo do tubete na casa de vegetação e de sombra, e o enraizamento e número de raízes na casa de sombra de miniestacas apicais e intermediárias, tratadas com AIB, em nove clones de Eucalyptus urophylla x E. globulus e dois clones de Eucalyptus grandis x E. globulus. No estabelecimento in vitro, a indução de brotações foi variável entre clones, sendo satisfatória para a maioria dos genótipos, enquanto a ocorrência de oxidação e contaminação por bactérias e fungos não comprometeu o estabelecimento das culturas. Na fase de multiplicação in vitro, houve resposta diferenciada quanto à taxa de multiplicação, com clones apresentando boas taxas de multiplicação in vitro e outros recalcitrantes à micropropagação. Os meios de cultura MS e JADS tiveram efeitos positivos, com tendência de maiores taxas de multiplicação para o meio MS, sendo que ao longo dos subcultivos a taxa de multiplicação variou com tendência de aumento inicial e posterior queda na maioria dos clones e introduções in vitro. Quanto à miniestaquia, não houve efeito significativo do AIB em nenhuma das características estudadas. A sobrevivência das miniestacas foi alta para a maioria dos clones e as taxas de enraizamento variaram entre clones e entre os tipos de miniestacas, com resultados superiores para miniestacas apicais.
