Resumo: Melatonin is responsible for circadian regulation and control of sleep, being found at greatest concentration in the body at night. Disorders in its production may be responsible for insomnia. A substantial decrease in melatonin production occurs with age and often affects the sleep quality of elderly individuals. It is therefore important to observe and regulate the production of melatonin in the body, since insomnia and sleep disorders may have diverse negative effects on health and productivity of individuals. One method of increasing melatonin in the organism is through food, since the compound is found in many different foods including mustard, fennel, and milk. However in these foods melatonin is found in low concentrations and its increase by the addition of melatonin to the food is not allowed. Therefore, the objective of the present study was to obtain milk with elevated melatonin concentrations by night milking. The profiles of individuals with and without insomnia were studied, as well as the main symptoms of sleep disorders. Results showed that there is a large niche market composed especially of elderly women, for the development of natural products which have the properties of improving sleep quality. Different periods (15h00min. and 2h 00min,) of milking were tested in order to define the period in which the milk obtained had the highest concentrations of melatonin. Milk obtained at 2h 00min presented melatonin concentrations nine times greater than the milk obtained at 15h 00min. Biological assays with Wistar rats were conducted to test the 3 formulations developed (milk obtained from milking at15h 00min, milk obtained from milking at 2h 00min, milk obtained from milking at 2h 00min and supplemented with tryptophan) in relation to the increase in blood melatonin and urinary sulphatoxymelatonin. Milk obtained by night milking successfully increased blood melatonin and urinary sulphatoxymelatonin, but the formulation that showed the best results by increase in the indicators quality of sleep was that obtained at 2h 00min and supplemented with tryptophan. For development of the new product a study of melatonin stability when subjected to heat treatment was necessary. This study was conducted by assessing the kinetics of thermal destruction of melatonin in milk. Results indicate that it is stable, and that the heat treatment used in milk cause little loss of melatonin. The non-sensory characteristics (presence of melatonin and information on the benefits of consuming melatonin) were tested in two groups (individuals younger than 50 and older than 50 years). The results show that information on the presence of melatonin had a positive impact on acceptance of the milk in both groups. Knowledge of the benefits of consuming melatonin also positively influenced the two groups, however the group composed of individuals over 50 was more strongly influenced. By means of a joint analysis of factors, a blue packaging for the product was developed which was designed with stars and pouring milk, given the name night-time milk and informative text was also printed on the packaging. Thus, an innovative and unique product was developed from night milking of the animals in the climate conditions of Brazil, with the addition of tryptophan, to improve sleep quality of elderly people and add value to milk without large investments on the part of producers, in both a sustainable and viable fashion.

Distúrbio em sua produção pode ser responsável pela insônia. Uma queda substancial na produção de melatonina acontece com a idade, podendo prejudicar a qualidade do sono dos idosos. Desta forma, é importante a observação e a regulação da produção da melatonina no organismo, uma vez que a insônia e os distúrbios do sono causam diversos malefícios à saúde e à produtividade das pessoas. Uma forma de aumentar a melatonina no organismo é por meio da alimentação, uma vez que o composto é encontrado em diversos alimentos como mostarda, erva-doce e leite. Porém, nestes alimentos a melatonina se encontra em baixas concentrações e o seu aumento, pela adição de melatonina no alimento, não é permitido. Assim, o presente trabalho objetivou a obtenção de leite com concentração de melatonina aumentada por meio das ordenhas noturnas. Foram estudados o perfil dos portadores e não portadores de insônia, bem como os principais sintomas dos distúrbios do sono. Os resultados mostraram que há um nicho de mercado, composto principalmente por idosos e mulheres, para o desenvolvimento de produtos naturais que tenham como propriedade melhorar a qualidade do sono da população. Diferentes horários (15h00min. e 2h00min.) de ordenha foram estudados a fim de identificar o período em que o leite obtido apresenta maior concentração de melatonina. O leite obtido por meio de ordenha realizada às 2h00min. (39,43 pg/mL) apresentou concentração de melatonina nove vezes superior ao leite obtido às 15h00min. (4,03ρg/mL). Ensaio biológico com ratos wistar foi realizado com objetivo de testar as 3 formulações desenvolvidas (leite obtido na ordenha das 15h00min., leite obtido na ordenha das 2h00min., leite obtido na ordenha das 2h00min. adicionado de triptofano) em relação ao aumento da melatonina sanguínea e sulfatoximelatonina urinária. O leite obtido por meio da ordenha noturna foi eficiente no aumento de melatonina sanguínea e sulfatoximelatonina urinária, porém a formulação que apresentou melhores resultados no aumento dos indicadores de qualidade do sono foi o leite ordenhado às 2h00min. e acrescido de triptofano. Para o desenvolvimento do novo produto, foi necessário estudar a estabilidade da melatonina a tratamentos térmicos. Este estudo foi realizado, utilizando-se a cinética de destruição térmica da melatonina no leite. Os resultados indicaram que os tratamentos térmicos usados em leite causam perdas insignificantes da melatonina. As características não sensoriais (presença de melatonina, e informações sobre os benefícios do consumo de melatonia) foram testadas em dois grupos (menores de 50 anos e maiores de 50 anos). Os resultados mostraram que a informação da presença de melatonina causa impacto positivo na aceitação do leite em ambos os grupos. O conhecimento dos benefícios do consumo de melatonina também influenciou positivamente os dois grupos, porém, o grupo de pessoas maiores de 50 anos foi influenciado mais fortemente. Por meio da análise conjunta de fatores, foi definida uma embalagem para o produto desenvolvido, de design azul com estrelas e desenho de leite derramando, com o nome leite noturno e com texto informativo.
