Resumo: A total of 120 isolates of Hemileia vastatrix, obtained from two coffee crop systems (conventional and organic) and six counties from two main production regions in Minas Gerais (Zona da Mata and Sul de Minas), sampled and multiplied in 2005, were characterized by microsatellite and RAPD markers. After checking DNA integrity of the samples by ITS 1 and ITS 4 primer pairs, we assessed the genetic diversity of all isolates with three microsatellite primer pairs and three RAPD primers. Bands from microsatellite primers were monomorphic, whereas bands from RAPD primers were polymorphic. Over all loci, the genetic diversity (h) was 0.28 and the genetic differentiation (Gst) was 0.22, therefore the Gst was considered high. Higher h (0.29) and Gst (0.25) values were estimated for the crops under organic system. When regions were compared, for Zona da Mata we estimated high h (0.27), but low Gst (0.17) values. The highest h and number of migrants (Nm) values were estimated for Tombos (0.29) and Poços de Caldas (11.90) counties, respectively. The cluster analysis did not show any structure regarding to the origin, crop system or production region of the isolates.

The cluster analysis did not show any structure regarding to the origin, crop system or production region of the isolates.Utilizaram-se os marcadores microssatélite e RAPD para a caracterização de 120 isolados de Hemileia vastatrix obtidos em 2005, de cafeeiros de seis municípios, sob dois sistemas de cultivos (convencional e orgânico), de duas das principais regiões cafeicultoras de Minas Gerais (Zona da Mata e Sul de Minas). Após avaliar a integridade das amostras de DNA dos isolados, com o par de primers ITS 1 e ITS 4, empregaram-se três pares de primers microssatélites e três primers RAPD para estudar a diversidade genética de H. vastatrix. Obtiveram-se padrões de bandas monomórficas, com os primers microssatélites, e polimórficas, com os primers RAPD. Considerando todos os locos, a diversidade gênica (h) foi de 0,28, a diferenciação genética (Gst) de 0,22 e, portanto, o Gst foi considerado alto. Nas lavouras sob cultivo orgânico, ocorreram maiores valores de h (0,29) e Gst (0,25). Comparando-se as regiões de cultivo, na Zona da Mata ocorreram maior h (0,27), porém menor Gst (0,17). Os valores de h e do número de migrantes (Nm) máximos ocorreram nos municípios de Tombos (0,29) e Poços de Caldas (11,90), respectivamente.
