Resumo: | Selenium (Se) is a trace element with antioxidant property and important enzimatic functions as selenocysteine. The low Se status is positively related with the incidence of chronic diseases, including cancer. Food components of the Brazilian diet, such as Brazil nut, naturally rich in selenium, and beans, with attributed functional properties to some of its constituents as fiber, phytate, and tannins are promising in the reduction of the risk of these chronic diseases. The objective of the current study was to evaluate the bioavailability of Se in 5 bean cultivars (Ouro Branco, Diamante Negro, BRS Radiante, Pérola, and Talismã), and in Pérola beans fertilized with Se. The study also aimed to evaluate the functional potential of black beans and Brazil nut in reducing the risk of colon cancer in rats. In the bioavailability study the bean cultivars were cooked with their soaking water; then, they were dried in oven, milled and analyzed for Se content. Female adult rats received their experimental diets containing the same levels of Se provided by the beans for 28 days. One control group with Se and another control group without Se were also included in the study. The activity of plasma and liver Glutathione Peroxidase (GPx) and the erythrocyte Se were determined. There was no difference between bean cultivars and beans fertilized with Se and its control in terms of Se bioavailability, as measured by liver and plasma GPx activity. To evaluate the anticarcionogenic effect of black beans and Brazil nut on colon cancer, 45 male F344 rats were distributed in 5 groups and fed with hyperlipidic diest for 12 weeks. One group received control AIN-93 diet without Se (CN), another group received AIN-93 diet supplemented with 4 mg Se kg-1 (CP), other two groups were fed the same Se dose of CP diet, provided either by Brazil nut (C) or Brazil nut + beans (F+C) and the last group received diet with beans (F). The carcinogen DMH and its vehicle (control) were administred to the animals in two consecutive doses at weeks two and three after diet introduction. Plasma Se and the activity of liver, erythrocyte and plasma GPx and liver Glutathione S-transferase (GST) were measured. Cell differenciation, cell proliferation and morphological characteristics of cript and colon were biomarkers for the risk of cancer. The diets CP, C, F and F+C promoted the same expression levels of GPx and GST, higher cell differentiation and lower cell proliferation of colon mucosa, in relation to CN diet. No change was observed with DMH administration. Nutritional overdoses of Se provided by SeMet, Brazil nut or their combination with beans (C+F) and the supplied bean separately (F) promoted homeostase of colon development, and avoided changes in cell differentiation and proliferation in DMH-treated rats. This suggests a protective role of these food or its components on colon cancer. The results seem to be due to multiple mechanisms of action. |

O baixo estado nutricional de Se tem mostrado correlacionar-se positivamente com a incidência de doenças, entre elas o câncer. Alimentos componentes da dieta brasileira, como a castanha-do-Pará, naturalmente rica em selênio, e o feijão, com propriedades funcionais atribuídas à presença de constituintes como fibra, fitatos e taninos tem mostrado efeitos promissores na redução do risco de doenças crônicas. Este estudo teve como objetivo avaliar a biodisponibilidade de Se em 5 cultivares de feijão (Ouro Branco, Diamente Negro, BRS Radiante, Pérola e Talismã) e em feijão do cultivar Pérola submetido ao enriquecimento com Se bem como o potencial funcional do feijão e da castanha-do-Pará na redução do risco de câncer de cólon em animais. No ensaio de biodisponibilidade, feijões dos 5 cultivares e do cultivar enriquecido com Se e seu controle foram cozidos com água de maceração, secos em estufa, triturados e quantificados quanto ao teor de Se. Ratas adultas Wistar receberam, por 28 dias, dietas experimentais com o mesmo nível de Se fornecidos pelos feijões e dietas controle com e sem adição do nutriente. Foram determinados a atividade enzimática de Glutationa Peroxidase (GPx) plasmática e hepática bem como a concentração eritrocitária de Se. Os cultivares de feijão estudados e o cultivar Pérola adubado com Se e seu controle não apresentaram diferenças entre si na biodisponibilidade do nutriente medida pela atividade GPx plasmática e hepática, não estando, portanto, condicionada por fatores antinutritivos presentes no alimento. Na avaliação do potencial anticarcinogênico do feijão e da castanha-do-Pará sobre o câncer de cólon, ratos machos F344 (45) foram divididos em cinco grupos e alimentados com dietas hiperlipídicas constituindo-se nos controles AIN-93 sem adição de selênio (CN) e AIN-93 suplementada com 4 mg de Se por kg de dieta (CP), dietas contendo a mesma dose de selênio da dieta CP fornecidos pela castanha-do-Pará de forma isolada (C) ou combinada com feijão (F+C) e dieta com feijão (F) por um período de 12 semanas. O carcinógeno de cólon dimetilhidrazina (DMH) foi aplicado a seis animais de cada grupo em duas doses consecutivas na segunta e terceira semanas após a introdução das dietas experimentais. Três animais de cada grupo receberam apenas a solução veículo constituindo-se nos controles. Foram medidos a atividade das enzimas GPx plasmática, eritrocitária e hepática e Glutationa S-transferase (GST) hepática e a concentração plasmática de Se. Diferenciação celular, proliferação celular e características morfológicas da cripta e do cólon foram utilizados como biomarcadores do risco de câncer. As dietas CP, C, F e F+C promoveram o mesmo nível de expressão das enzimas GPx e GST, maior grau de diferenciação celular e menor grau de proliferação celular da mucosa colônica em relação à dieta CN, não sendo observadas alterações de padrão quando da aplicação do DMH. A administração de doses supranutricionais de Se na forma de SeMet ou fornecida pela castanha-do-Pará e a utilização de feijão de forma isolada ou em combinação com a castanha favoreceu a homeostase do desenvolvimento colônico evitando alterações na diferenciação e proliferação celular em ratos tratados com DMH, sugerindo um papel protetor destes alimentos no câncer de cólon.
