Resumo: | The relevance of ticks as parasites lies not only on their effects on their hosts, but also the great ability to act as vectors of pathogens, which include viruses, bacteria, protozoa and nematodes. Traditionally, identification of ticks was based on morphological characters and it is labor intensive, and depended on the expertise of specialists, the state of preservation of the material and the stage of development of the specimen. Because it adopts molecular identification of ticks, this work represents a pioneering effort in Brazil. Sequence analyses involving amplification of the second internal transcribed spacers (ITS2) of rDNA were performed for identification of genera and species of ticks. The design of specific primers and PCR standardization was carried on for species considered as major epidemiological issues. ITS2 regions were considered as useful for molecular identification and showed low intraspecific variation. Three out of four designed sets of primers were effective for genera identification. Trials for species-specific primers were successful for three species of Amblyomma, resulting in amplicons of 404 bp, 389 bp and 320 bp for A. cajennense, A. oblongoguttatum and A. parvum, respectively. Sensitivity of the speciesspecific primers was successfully tested in A. cajennense. It was concluded that these results contributed to the efficient molecular identification of ticks species that are of great national relevance. |

It was concluded that these results contributed to the efficient molecular identification of ticks species that are of great national relevance.A importância parasitológica dos carrapatos reside não somente na sua ação espoliativa sobre o hospedeiro, como também na grande capacidade de atuar como vetores de agentes patogênicos, entre os quais se incluem vírus, bactérias, protozoários e nematóides. Tradicionalmente, a identificação de carrapatos baseada em características morfológicas é uma atividade extremamente trabalhosa, demorada, passível de dúvidas, dependente de pessoal especializado e com limitação quanto à sua possibilidade de realização, dependendo do estado de conservação e do estágio de desenvolvimento do espécime. Assim, o presente trabalho é pioneiro no Brasil, no sentido de adotar uma estratégia molecular para a identificação de carrapatos. Os objetivos foram buscar marcadores moleculares capazes de identificar e distinguir diferentes gêneros e espécies de carrapatos, envolvendo amplificação dos espaçadores internos transcritos secundários (ITS2) e seqüenciamento dos mesmos. Além disso, objetivou-se o desenho de primers específicos e a padronização de PCRs direcionadas às espécies de maior importância epidemiológica. Foi observado que os ITS2 são regiões úteis para a identificação molecular por apresentarem baixa variabilidade intraespecífica. Três dos quatro conjuntos de primers desenhados para gêneros demonstraram ser eficientes para identificação, enquanto que nos ensaios para primers espécie-específicos foi obtido êxito para três espécies do gênero Amblyomma, resultando em amplicons de 404 pb, 389 pb e 320 pb característicos para as espécies A. cajennense, A. oblongoguttatum e A. parvum, respectivamente.
