Resumo: | Ram semen was diluted (200 x 106 sperm/mL) in Tris-egg yolk (5% glycerol), added of myicetin (0.1, 10, 20, 30, 40, 100, 200, 300, 400 and 1000 nM) and frozen (-196 ° C). After thawing (37°C/30s), kinetics, iMPA, PMM, iROS, LPO, eMP and fertility in vivo (0 and 100 nM myricetin) were evaluated. The analyzes of 0, 1, 10, 100 and 1000 nM of myricetin showed: 10nM myricetin showed lower % RAP (P≤0.05), than 1000nM. Samples of control showed higher (P≤0.05) VAP than 10nM, while samples with 10, 100 and 1000nM of myicetin showed higher BCF (P≤0.05), when compared to control. Miricetin 1000nM presented higher percentage (P <0.05) of cells with LPO, than the control. For 0, 20, 30, 40, 100, 200, 300 and 400 here was no treatment effect (P>0.05) for kinetics and flow cytometry (P> 0.05), there were also no interactions between treatment and time (P> 0.05). For both there was effect of the incubation time, in some evaluated parameters, the PM, VCL, VSL, VAP, LIN, ALH, percentage of cells with intact plasma and acrosomal membrane, oxidative stress and membrane stability were reduced (P <0.05) in all groups, while BCF (P <0.05) increased in all groups.. The pregnancy rate in the 100 nM myricetin group was 50% (5/10) and that of the 10% control (1/10). In the in vitro analysis, it was concluded that myricetin has no antioxidant effect on cryopreserved spermatozoa. However, it improves the pregnancy rate of inseminated sheep. |

However, it improves the pregnancy rate of inseminated sheep.Sêmen ovino foi diluído (200 x 106 espermatozoides/mL) em Tris-gema de ovo (5% de glicerol), acrescido de miricetina (0, 1, 10, 20, 30, 40, 100, 200, 300, 400 e 1000 nM) e congelado (-196 °C). Após descongelação (37°C/30s), avaliou-se cinética, iMPA, PMM, iROS, LPO, eMP e fertilidade in vivo (0 e 100 nM miricetina). As analises de 0, 1, 10, 100 e 1000 nM de miricetina evidenciaram: miricetina 10nM apresentou menor %RAP (P≤0,05), do que 1000nM. Amostras do controle apresentaram maior (P≤0,05) VAP que o 10nM, enquanto amostras com 10, 100 e 1000nM de miricetina evidenciaram maior BCF (P≤0,05), quando comparadas ao controle. Miricetina 1000nM apresentou maior percentual (P<0,05) de células com LPO, do que o controle. Para 0, 20, 30, 40, 100, 200, 300 e 400 não houve efeito do tratamento (P>0,05) para cinética e citometria de fluxo (P>0,05), também não houve interações entre tratamento x tempo (P>0,05). Para ambos houve efeito do tempo de incubação, em alguns parâmetros avaliados a MP, VCL, VSL, VAP, LIN, ALH, porcentual de células com membrana plasmática e acrossomal intactas, estresse oxidativo e estabilidade de membrana foram reduzidos (P <0,05) em todos os grupos, enquanto que o BCF (P <0,05) aumentou em todos os grupos. A taxa de prenhez no grupo 100 nM de miricetina foi de 50% (5/10) e a do controle 10% (1/10). Nas análises in vitro, conclui-se que a miricetina não tem efeito antioxidante sobre os espermatozoides criopreservados.
