Resumo: | After the menopause there is an increased in the incidence of hypertension, insulin resistance, type 2 diabetes and morbidity. In other hand, aerobic exercise training has been recommended as an important therapy for the prevention and treatment of cardiometabolic disorders. Recently, resistance exercise training has been recommended as a complement to aerobic (combined training) for the treatment of cardiovascular and metabolic diseases. However, the physiological effects of this approach is not well understood. Moreover, the mechanisms involved in the possible benefits of combined exercise training have been few studied after ovarian hormone deprivation, especially in the presence of cardiometabolic disorders. Thus, the aim of this study was to evaluate the effects of exercise training combined in hypertensive ovariectomized rats submitted to fructose overload. Wistar rats and spontaneously hypertensive rats (SHR) were divided in: control (C), hypertensive (HS), hypertensive ovariectomized (HOS) and hypertensive ovariectomized submitted to fructose overload sedentary (HOFS) and trained (HOFT). Initially, the maximum load test and resistance training protocol in ladder adapted to rats were standardized. The combined exercise training was performed on a treadmill and ladder adapted to rats in alternated days. Cardiovascular function, autonomic cardiovascular modulation, metabolic and physical capacity were evaluated, as weel as the relationship of these variables with cardiac oxidative stress and inflammation. The association of hypertension and ovariecyomy induced increase in arterial pressure (AP ) (HOS: 164??5 vs. HS: 146??6 mmHg) and body weight in relation to hypertensive rats. The association of risk factors (hypertension + ovariectomy + fructose) induced an additional increase in the AP (HOFS: 174??4 mmHg), increased heart rate (HOFS: 403??12 vs. HOS: 348??16 bpm) and blood triglycerides (HOFS: 160??8 vs. HOS: 125??6.4 mg/dl), induced an impairment in the insulin sensitivity and cardiovascular autonomic modulation, oxidative stress (redox ratio of glutathione: HOFS: 8.94??0.8 vs. HOS: 13.00??1,4) and inflammation (TNF-??- HFOS: 65.8??9.9 vs. HOS: 31.7??8.6 pg/mg protein) of these animals. Moreover, the combined exercise training was able to reduce the AP (HOFTc: 158??4 mmHg) and heart rate (HOFTc: 303??5 bp), to normalize the triglyceride levels (HOFTc: 137??4.7 mg/dL), insulin sensitivity, vascular autonomic modulation. Moreover training reduced cardiac lipoperoxidation and increased antioxidant defense and redox balance of glutathione (HOFTc: 11.94 ?? 1.1) and reduced the TNF- ?? (HOFTc: 33.1??4.9 mg/dL) and increased the IL-10. The results of this study demonstrate that the association of risk factors promoted additional impairment in metabolic, cardiovascular, autonomic, inflammatory and oxidative stress parameters; and the combined exercise training was effective in reduce and/or normalize these dysfunctions. |

Por outro lado, o treinamento f??sico aer??bio tem sido recomendado como uma importante terapia para a preven????o e o tratamento de disfun????es cardiometab??licas, em fun????o dos reconhecidos benef??cios dessa abordagem em diferentes popula????es. Mais recentemente, o treinamento f??sico resistido vem sendo recomendado em complemento ao aer??bio (treinamento combinado) nas diretrizes de tratamento de doen??as cardiovasculares e metab??licas, no entanto, pouco se sabe sobre os benef??cios fisiol??gicos dessa abordagem. Al??m disto, os mecanismos envolvidos nos eventuais benef??cios do treinamento f??sico din??mico combinado foram muito pouco estudados ap??s a priva????o dos horm??nios ovarianos, principalmente em presen??a de disfun????es cardiometab??licas. Desta forma, o objetivo do presente estudo foi avaliar os efeitos do treinamento f??sico din??mico combinado em ratas ooforectomizadas hipertensas submetidas ?? sobrecarga de frutose. Foram utilizadas ratas Wistar controles (CS) e ratas espontaneamente hipertensas (SHR): submetidas (HOS) ou n??o (HS) ?? priva????o dos horm??nios ovarianos e ooforectomizadas submetidas ?? sobrecarga cr??nica de frutose sedent??rias (HOFS) ou ao treinamento f??sico combinado (HOFTc). Inicialmente foi padronizado um teste de carga m??xima e um protocolo de treinamento resistido em escada para ratos. O treinamento f??sico combinado foi realizado em esteira e escada adaptadas para ratos em dias alternados. Par??metros de fun????o cardiovascular, de regula????o auton??mica cardiovascular, metab??licos e de capacidade f??sica e sua rela????o com altera????es em par??metros de estresse oxidativo e de inflama????o foram avaliados no tecido card??aco. Foi observado que a associa????o (hipertens??o+ooforectomia) induziu aumento da press??o arterial (PA) (HOS: 164??5 vs. HS: 146??6 mmHg) e peso corporal em rela????o as ratas hipertensas. J?? a associa????o de fatores de risco (hipertens??o+ooforectomia+frutose) induziu aumento adicional PA (HOFS: 174??4 mmHg), aumento da frequ??ncia card??aca (HOFS: 403??12 vs. HOS: 348??16 bpm) e dos n??veis de triglic??rides sangu??neos (HOFS: 160??8 vs. HOS: 125??6,4 mg/dl), preju??zo na sensibilidade ?? insulina e na modula????o auton??mica cardiovascular, al??m de preju??zos no perfil de estresse oxidativo (raz??o redox da glutationa: HOFS: 8,94??0,8 vs. HOS: 13.00??1,4) e inflamat??rio (TNF-??: HOFS: 65,8??9,9 vs. HOS: 31.7??8.6 pg/mg de prote??na) desses animais. Por outro lado, o treinamento f??sico combinado foi capaz de reduzir a PA (HOFTc: 158??4 mmHg), promover bradicardia de repouso (HOFTc; 303??5 bpm), normalizar os n??veis de triglic??rides (HOFTc: 137??4,7 mg/dL), a sensibilidade ?? insulina, modula????o auton??mica vascular, reduzir o dano a membrana celular, aumentar a defesa antioxidante e o balan??o redox da glutationa (HOFTc: 11,94??1,1), al??m de reduzir o TNF-?? (HOFTc: 33,1??4,9 mg/dL) e aumentar IL-10. Dessa forma, os resultados do presente estudo demonstram que a associa????o de fatores de risco promoveu preju??zos adicionais em par??metros metab??licos, cardiovasculares, auton??micos, de estresse oxidativo e inflamat??rios, de metaboliza????o e que o treinamento f??sico combinado foi eficaz em atenuar e/ou normalizar tais disfun????es.Made available in DSpace on 2015-04-22T17:20:43Z (GMT).
