No Brasil, sua distribuição é heterogênea, sendo observada em todos os estados e, em Pernambuco apresenta-se como doença endêmica. Suas manifestações clínicas variam, sendo a forma cutânea a forma mais comum. A resposta imunológica desenvolvida pelo hospedeiro desempenha um importante papel na cura clínica da doença ou na sua progressão, e os linfócitos T, bem como células NK são fundamentais, por serem fontes produtoras de citocinas e por atuarem na resposta imune imediata à infecção. Dessa maneira, os objetivos desse estudo foram caracterizar os pacientes quanto aos aspectos clínicos, epidemiológicos e laboratoriais, identificar no sangue periférico de pacientes com LTA as células NK e NKT, linfócitos T CD8⁺ e T CD4⁺ e associar o perfil fenotípico observados com a evolução clínica dos pacientes avaliados. A imunofenotipagem foi feita por citometria de fluxo, avaliando moléculas presentes na superfície de células mononucleares do sangue periférico de pacientes com a doença ativa após o período de 72h de cultura, após a estimulação com antígeno total de L. brasiliensis. Os resultados desse trabalho, demostraram que células NK, NKT e T CD8+, estiveram presentes em maior quantidade no sangue desses pacientes. Sendo importantes na imunidade imediata à LTA. Além disso, foi visto que antígeno total de L. (V.) braziliensis é capaz de induzir uma resposta imune específica por parte dos pacientes com LTA, nos períodos iniciais da infecção e que o balanço entre células da imunidade inata e adaptativa contribuem para a proteção à doença. O estudo mostrou que a resposta imunológica desenvolvida pelo hospedeiro na LTA é dinâmica, e contribui em futuros estudos para o desenho apropriado de intervenções imunológicas em pacientes e da produção de vacinas.FACEPEAmerican Cutaneous Leishmaniasis (LTA) is a neglected disease with a worldwide distribution, which affects mainly the populations of lower socioeconomic status. In Brazil, its distribution is heterogeneous, being observed in all states, and in Pernambuco, it is an endemic disease. Its clinical manifestations vary, and the cutaneous form is the main common form. The immune response developed by the host plays an important role in the clinical cure of the disease or in its progression, and T lymphocytes as well as NK cells are fundamental because they are sources of cytokine and because they act on the immediate immune response to the infection. Therefore, the objectives of this study were to characterize the patients in the clinical, epidemiological and laboratory aspects, to identify in the peripheral blood of patients with ACL, NK and NKT cells, CD8+ and CD4+ T lymphocytes and associate the phenotypic profile observed with the clinical evolution of the patients. Immunophenotyping was done by flow cytometry, evaluating the molecules on the surface of peripheral blood mononuclear cells of patients with an active disease, after the period of 72h of culture. The results of this work demonstrated that NK, NKT and CD8 + T cells were present in a larger quantity in the blood of these patients. Being important in the immediate immunity in ACL. In addition, it was seen that total antigen of L. (V.) braziliensis is able to induce a specific immune response by the patients with ACL in the initial periods of infection, and that the balance between cells of innate and adaptive immunity contributes to the disease protection.
