Resumo: This study aimed to evaluate the influence of the diversity of nitrogen-fixing bacteria on the productivity of 6 tree species native to the Atlantic Forest in microcosm conditions. Two experiments were conducted in a glasshouse at Embrapa Agrobiologia. The first experiment simulated a community with 3 legume and 3 non-legume species and it was carried out in trays containing 45 kg of substrate (soil:sand:vermiculite v:v:v). These species were tested individually in 1.5 kg pots in the second experiment. Both experiments followed a randomized block design, with 6 replicates. The following treatments were tested: inoculation of plants with two levels of rhizobial richness (3 and 9 strains), and two controls, one receiving mineral fertilization and another that received neither inoculation or fertilization. The rhizobia inoculated are recommended for the legume species used in the experiments, that is, Albizia pedicellaris, Piptadenia gonoacantha, and Pseudopiptadenia contorta. Plant height was measured at 30, 60, 90, 120, 150 and 180 days after transplanting. Plants were collected after 180 days and dried at 65 ? C until constant weight and the masses of dry shoot and root, as well as shoot total N, were measured. Further, nodule occupancy was evaluated. The levels of diversity of nitrogen fixing bacteria for some species of legumes had a positive effect on average height , accumulation of root mass and leaf N content . This effect was not observed for the variable dry matter of shoots. Additionally, we isolated and characterized, phenotypic and genotypically, bacteria isolated from nodules Pseudopiptadenia contorta , Albizia pedicellaris and Piptadenia gonoacantha. The Morphophysiological characterization demonstrated considerable diversity among the isolates. PCR amplification using the primer BOX- A1R revealed a high genotypic diversity among isolates. Seven major groups were formed. The molecular analysis of 16S rRNA gene sequences revealed that these bacteria belong to the genera Burkholderia, Bradyhrhizobium, Rhizobium and Mesorhizobium. A group of isolates did not cluster closely to species already described, thus presenting a potential for the description of new species. Nodule occupancy by the inoculated bacteria in forest legume Albizia pedicellaris and Piptadenia gonoacantha in the microcosm differed from that in individual pots.

Foram conduzidos dois experimentos em casa de vegeta??o na Embrapa Agrobiologia. O primeiro foi realizado em bandejas contendo 45 kg de substrato simulando uma comunidade juvenil com 3 esp?cies de leguminosas e 3 de n?o leguminosas em crescimento conjunto. O segundo foi realizado com as mesmas esp?cies, por?m as plantas foram individualizadas em vasos de 1,5 kg de substrato. Em ambos os experimentos foi utilizado o delineamento de blocos inteiramente casualisados com 6 repeti??es e constitu?dos dos seguintes tratamentos: controle n?o inoculado, controle nitrogenado e 2 n?veis de riqueza de esp?cies de riz?bio (1 ou 3 estirpes recomendadas para cada leguminosa). As estirpes de riz?bio inoculadas inclu?ram bact?rias recomendadas para Albizia pedicellaris, Piptadenia gonoacantha e que nodulam Pseudopiptadenia contorta. O substrato utilizado foi uma mistura autoclavada na propor??o 1:1:1 (v:v:v) de areia, vermiculita e horizonte B de um Argissolo textura arenosa. A altura das plantas foi avaliada aos 30, 60, 90, 120, 150 e 180 dias ap?s o transplantio. O material vegetal foi seco em estufa de circula??o de ar for?ada a 65?C, at? peso constante, determinando-se a mat?ria da parte a?rea seca. Este material foi mo?do e analisado para o teor de nitrog?nio foliar. Al?m disso, foi realizada a avalia??o da ocupa??o nodular, amostrando alguns n?dulos de cada leguminosa e tratamento. A caracteriza??o morfofisiol?gica demonstrou uma consider?vel diversidade entre os isolados. A amplifica??o por PCR usando o iniciador BOX-A1R para a amplifica??o de regi?es flanqueadas por sequ?ncias repetitivas de DNA (BOX-PCR) foi obtida com sucesso sendo verificada uma alta diversidade genot?pica entre os isolados. Sete grupos principais foram formados. As an?lises de sequ?ncias moleculares do gene 16S DNAr revelou uma diversidade de esp?cies de riz?bio onde foram encontrados 4 g?neros diferentes nodulando as tr?s esp?cies florestais: Burkholderia, Bradyhrhizobium, Rhizobium e Mesorhizobium. Um grupo de isolados se destacou dos demais ressaltando uma grande potencialidade para ser identificado como uma nova esp?cie nodulando leguminosas florestais. A ocupa??o nodular pelas bact?rias inoculadas nas leguminosas florestais Albizia pedicellaris e Piptadenia gonoacantha foi diferente em condi??o de microcosmo quando comparadas ?s plantas individualizadas. Tanto em crescimento conjunto ou individualizado a leguminosa Pseudopiptadenia contorta n?o apresentou nodula??o. Os n?veis de diversidade de bact?rias fixadoras de nitrog?nio para algumas esp?cies de leguminosas causou um efeito positivo na altura m?dia, ac?mulo de massa de raiz e teor de nitrog?nio foliar. Additionally, we isolated and characterized, phenotypic and genotypically, bacteria isolated from nodules Pseudopiptadenia contorta , Albizia pedicellaris and Piptadenia gonoacantha. The Morphophysiological characterization demonstrated considerable diversity among the isolates. PCR amplification using the primer BOX- A1R revealed a high genotypic diversity among isolates. Seven major groups were formed. The molecular analysis of 16S rRNA gene sequences revealed that these bacteria belong to the genera Burkholderia, Bradyhrhizobium, Rhizobium and Mesorhizobium. A group of isolates did not cluster closely to species already described, thus presenting a potential for the description of new species. Nodule occupancy by the inoculated bacteria in forest legume Albizia pedicellaris and Piptadenia gonoacantha in the microcosm differed from that in individual pots.
