Resumo: | In this work it was studied the liquid-liquid extraction process, using aqueous two phase systems (ATPS) in order to fractionate whey proteins α-lactoalbumin (α-la), β-lactoglobulin (β-lg) and glycomacropeptide (GMP). The evaluated ATPS were composed by PEG 2000 and inorganic salts (potassium phosphate, sodium citrate and lithium sulfate). Initially a fractionated factorial design was applied in order to evaluate the influence of the factors (pH, temperature, PEG concentration, salt concentration and NaCl concentration) on the proteins partition. The results weresubmitted to a variance analysis and it was found that pH and NaCl molar concentration affected significantly (p<0,05) the partition of α-la and β-lg. After that, a central composite rotatable design (CCRD) was used to test different levels of pH and NaCl molar. The results obtained from the CCRD were submitted to a regression analysis and response surface analysis in order to determine the appropriate conditions to optimize the partition of such proteins, for the different salts used. On the systems PEG/citrate it was verified that for pH values higher, the increasing of NaCl leaded to higher partition coefficient values of β-lg. For α-la the increment of the pH increase the partition coefficient while GMP presented a maximum partition coefficient (K=0,912) in pH 6,35 e 0,441 mol L-1 of NaCl. For the PEG/phosphate systems, the increasing of NaCl leaded the partition coefficient of β-lg to lower values, while for α- la it was found a condition of minimum in pH 6,0 and 0,441 mol L-1 of NaCl. On the PEG/sulfato ATPS this time, it was obtained a minimum for the partition coefficient of β-lg (K=0,031) in pH pH 5,67 and 0,276 mol L-1 of NaCl. The fractionation experiments were carried out on PEG/potassium phosphate (13% w/w for both), pH 5,43 and 0,290 molar of NaCl, at 25°C, using different dilutions of whey (pure, 1:1 and 1:3). The extraction yield for the aimed proteins, the purification factor for β–lactoglobulin and reduction rate for α-lactoalbumin were determinate on each system after 3 steps of liquid-liquid extraction. It was observed that among the ATPS tested, the ones which used pure whey presented higher reduction rate for α-lactoalbumin (5,56) on the bottom phase and higher purification factor β-lactoglobulin (96,25 %) on the salt phase after tree extraction stages. This study showed that ATPS can be used to fractionate the proteins α-lactoalbumin, β-lactoglobulina and glycomacropeptide form in natura whey. |

This study showed that ATPS can be used to fractionate the proteins α-lactoalbumin, β-lactoglobulina and glycomacropeptide form in natura whey.Neste trabalho, estudou-se o processo de extração líquido-líquido em sistemas aquosos bifásicos (SAB) para a otimização do fracionamento das proteínas do soro de leite α-lactoalbumina (α-la), β-lactoglobulina (β-lg) e glicomacropeptídeo (GMP). Os SAB avaliados foram compostos por polietilenoglicol (PEG) 2000 gmol-1 e sais (fosfato de potássio, citrato de sódio e sulfato de lítio). Inicialmente foi avaliado a influência dos fatores pH, temperatura, concentração de PEG, concentração de sal e concentração de NaCl sobre a partição das proteínas, aplicando-se um delineamento fatorial fracionado. Os resultados obtidos foram submetidos a análise de variância, sendo verificado que o pH e a concentração molar de NaCl afetaram significativamente (p<0,05) a partição da α-la e β-lg. Em seguida, foi empregado um delineamento composto central rotacional (DCCR), testando diferentes níveis dos fatores pH e concentração molar de NaCl. Os resultados obtidos no DCCR foram submetidos as análises de regressão e superfície de resposta, sendo determinadas as melhores condições para a otimização da partição das proteínas, em cada tipo de sal. Para os sistemas PEG/citrato, verificou-se que para valores de pH acima de 5,6, o aumento da concentração de NaCl nos sistemas promoveu um aumento no coeficiente de partição da β-lg. Para a α-la, o incremento do pH proporcionou um aumento do coeficiente de partição, enquanto para o GMP foi obtido um valor máximo de coeficiente de partição (K=0,912) em pH 6,35 e 0,441 molL-1 de NaCl). Para os sistemas PEG/fosfato, o acréscimo de NaCl aos sistemas promoveu uma redução no valor do coeficiente de partição da β-lg, e para a α-la, foi obtida uma condição de minimização de seu coeficiente de partição. Já para o SAB PEG/sulfato, em pH 5,67 e 0,276 molL-1 de NaCl foi obtido um mínimo de coeficiente de partição para a β-lg (K=0,031). Avaliou-se o fracionamento das proteínas do soro in natura em SAB PEG/fosfato de potássio (ambos 13 % em massa), em pH 5,43 com 0,290 molL-1 de NaCl, a 25οC, em função das diferentes diluições do soro (integral, 1:1 e 1:3). Em cada sistema determinou-se o rendimento de extração das proteínas-alvo, fator de purificação da β-lg e a taxa de redução da α-la na fase salina, após 3 estágios de extração líquido-líquido. Verificou-se que dentre os sistemas avaliados, os SAB adicionados de soro integral, apresentaram: maior taxa de redução da α-la (5,56) na fase inferior e maior fator de purificação da β-lg (96,25 %) na fase salina, após três estágios de extração.
