Resumo: | Aqueous two phase systems (ATPS) have been used with great success in processes of separation, concentration and purification of bioactive molecules, since they are a variant of conventional liquid-liquid extraction, which favor the stability of biomolecules. As the water component present in the largest amount in both phases, these systems provide an environment for the biomolecules mild preserving thus its stability and avoiding loss of molecular biological activities. Therefore, this study was obtained new equilibrium data for systems formed by ionic liquid ([C4min] [BF4]) + Li2SO4 + water, ionic liquid ([C4min] [BF4]) + MnSO4 + water; copolymer (F68) + C4H4NaO6 + water, and copolymer (F68) + C4H4K2O6 + water at 25 ° C. For systems consisting of ionic liquid and sulfate it was observed that by changing the cation of the sulphate added to the system, there was a tendency of increased region to the biphasic curve Li2SO4 higher concentrations of salt. In the case of copolymer systems formed of organic and salt were also significant shift of the two phase region. For the system in which it was used C4H4NaO6 the region with the formation of two phases is greater. Subsequently, was used these new data in the study of partition of the whey proteins α-lactalbumin (α-la) and β-lactoglobulin (β-lg). In this step sought to partition the proteins so obtain two fractions, one rich in β-lg and the other rich in α-la. Were studied the effects of the types of salts used in training systems on the partition coefficient of the protein of interest. The systems formed by [C4min] [BF4], Li2SO4 and water, able to partition the proteins, where the highest concentration of α-la was detected in the lower phase (rich in ionic liquid). Already β-lg partitioned to saline phase (upper phase). For systems consisting of [C4min] [BF4], MnSO4 and water, exclusively for both proteins separated to lower phase. However, during the experiments it was observed protein precipitation at the interface systems. Systems already formed by copolymer (F68) C4H4NaO6 and water, and copolymer (F68) C4H4K2O6 and water separated proteins of interest only for the saline phase (lower phase), in other words, the systems were not able to separate the proteins of interest in two distinct phases. |

Sendo a água o componente presente em maior quantidade em ambas as fases, estes sistemas fornecem um ambiente ameno para as biomoléculas preservando, assim, a sua estabilidade molecular e evitando perdas das atividades biológicas. Assim sendo, neste trabalho obteve-se novos dados de equilíbrio para sistemas formados por líquido iônico ([C4min][BF4]) + Li2SO4 + água; líquido iônico ([C4min][BF4]) + MnSO4 + água; copolímero (F68) + C4H4NaO6 + água; e copolímero (F68) + C4H4K2O6 + água na temperatura de 25 °C. Em seguida, empregou-se estes novos dados no estudo de partição das proteínas do soro α-lactalbumina (α-la) e β-lactoglobulina (β-lg). Para os sistemas compostos por líquido iônico e sulfato foi observado que ao mudar o cátion do sulfato adicionado ao sistema houve uma tendência do aumento da região bifásica para a curva do Li2SO4, em concentrações maiores de sal. No caso do sistemas constituídos por copolímero e sal orgânico também houve deslocamento significativo da região bifásica. Para o sistema no qual utilizou-se C4H4NaO6, a região com formação de duas fases foi maior. Foram estudados os efeitos dos tipos de sais utilizados na formação dos sistemas sobre o coeficiente de partição das proteínas de interesse. Os sistemas formados por [C4min][BF4], Li2SO4 e água conseguiram particionar as proteínas, nos quais a maior concentração de α-la foi detectada na fase inferior (rica em líquido iônico). Já a β- lg particionou para a fase salina (fase superior). No caso dos sistemas constituídos por [C4min][BF4], MnSO4 e água, ambas proteínas particionaram exclusivamente para a fase inferior. Entretanto, verificou-se precipitação de proteínas na interface dos sistemas.
