Resumo: This study was developed at Ranário Experimental in Universidade Federal de Viçosa, Viçosa (MG) from April 6th, 2009 to June 7th, 2009, and aimed to assess the seminal characteristics and analyze the morphology of semen and sperm peppermint frog (Leptodactylus labyrinthicus). We collected eight male rã-pimenta, suitable for reproduction, in Viçosa, Cajuri, Timóteo and Governador Valadares. The assumed temperature was 25.0 ± 1.5 Cº with two photoperiods: LD 12:12 (LD = Light Dark) and 14:10 LD. For induction of spermiogenesis, we used the hormone analogue buserelin acetate (GnRHa) at a dose of 0.4 mg (= 0.1 mL), charged in the coelomic cavity of the animal with the aid of a hypodermic needle 25, 0 x 0.7 mm. This procedure was performed for two consecutive days: five doses on the first day with an hour interval between doses, and three doses in the second, with the first dose at the same time the day before and 1 h interval between doses. On the first day were collected five of the semen specimens of frog pepper (one, two, three, four and five hours after the first hormone application). To verify the effect of treatment was used an optical microscope with bright field (400x increase), which examined slides with semen. It was considered positive the slides with the presence of spermatozoa. An aliquot of fresh semen from each animal, corresponding to a value of one drop (approximately 0.05 mL) was placed on a glass slide and the tax rate, a cover slip. Force, motility and sperm concentration were avaluated. The animals responded positively to the hormone only the second day (5 h of treatment the first day and after 24 h, two more hours of treatment), totaling 26 h to collect semen, photoperiod of 14: 10 LD. In Plant Anatomy Laboratory, Universidade Federal de Viçosa, we used the light microscope Olympus BX-40 and held image capture, which was used for the analysis of 28 sperm, with the help of the program analyzes the image IMAGE PRO PLUS®, the head area and total length, head, tail and the membrane surrounding the head. The relevant time minimum-maximum (average) were: the head area: 15.57 to 50.15 (29.80) μm2, total length: 26.89 -76.06 (21.21) μm and the membrane surrounding the head.

Oito machos de rãs-pimenta, aptos à reprodução, de Viçosa, Cajuri, Timóteo e Governador Valadares foram coletados. A temperatura adotada foi de 25,0 ± 1,5 °C, com dois fotoperíodos: 12:12 LD (LD = Light Dark) e 14:10 LD. Utilizou-se o hormônio análogo acetato de buserelina (GnRHa) na dosagem de 0,4 μg (= 0,1 mL) para indução a espemiação, aplicado na cavidade celomática do animal com uma seringa hipodérmica com agulha de 25,0 x 0,7 mm. Esse procedimento foi realizado durante dois dias consecutivos: cinco doses no primeiro dia, com intervalo de uma hora entre doses, e três doses no segundo, com a primeira dose no mesmo horário do dia anterior e com intervalo de 1 h entre doses. No primeiro dia cinco coletas do sêmen dos exemplares de rã-pimenta (uma, duas, três, quatro e cinco horas após a primeira aplicação do hormônio) foram realizadas. O efeito do tratamento foi avaliado com microscópio óptico de luz com campo claro (aumento de 400x), onde se analisaram lâminas com sêmen. Foi considerado como positivo as lâminas com presença de espermatozoides. Uma alíquota do sêmen in natura de cada animal, correspondente ao valor de uma gota (aproximadamente 0,05 mL), foi colocada sobre uma lâmina de vidro e, sobre a alíquota, uma lamínula. O vigor, motilidade e concentração espermática foram avaliados. Hormônio induziu somente no segundo dia (5 h de tratamento no primeiro dia e, após 24 h, mais duas horas de tratamento), totalizando 26 h para coletar o sêmen, com fotoperíodo de 14:10 LD. Um fotomicroscópio Olympus BX-40 foi utilizado no Laboratório de Anatomia Vegetal da Universidade Federal de Viçosa,e realizou-se a captura de imagem, de 28 espermatozóides com auxílio do programa de análises de imagem IMAGE PRO PLUS®, obtendo-se a área da cabeça e comprimentos total, da cabeça, cauda e membrana que envolve a cabeça.
