Resumo: | The treatments for Chagas disease and leishmaniasis are not very efficient and have many side effects. There is an urgent need for new drugs for the chemotherapy of Chagas disease and leishmaniasis. Phytotherapy has been an alternative of great interest for the development of new drugs. Together with this, Brazil has one of the largest, if not the largest, biodiversity in the world. The objective of this research was to test the trypanocidal and leishmanicidal activities of the crude extracts of the bark, leaves and flowers of Tabebuia avellanedae. The parts of the vegetable were collected in the region of Franca / SP and the extraction was carried out in the Chemistry laboratory of UNIFRAN. For extraction, hydroalcoholic solvent (ethanol / H²O 96.4 ° GL) was used. The extracts were aliquoted and tested in four concentrations (8 µg / mL, 32 µg / mL, 128 µg / mL and 512 µg / ML) of each extract diluted in dimethylsulfoxide (DMSO), were evaluated for each parasite used. trypanocidal trypomastigote forms of Trypanossoma (Schizotrypanum) cruzi (Cepa Y), and to verify leishmanicidal activity promastigote forms of Leishmania (Viannia) braziliensis were used. The 'in vitro' tests were carried out at the Parasitology Laboratory of the Faculty of Pharmaceutical Sciences of Ribeirão Preto, University of São Paulo (FCFRP-USP). The method used to determine the activity of the extrater was colorimetric by 3- (4,5-dimethylthiazol-2-yl) -2,5-diphenyltetrazolium bromide (MTT). It was observed that the crude extract of the bark of Tabebuia avellanedae showed greater trypanocidal and leishmanicidal activity with an IC50 of 697.0 ug / mL and 774.2 uh / mL, respectively, when compared to the extracts of the leaves and flowers of Tabebuia avellanedae, which did not demonstrate significant trypanocidal and leishmanicidal activity. When comparing the results with the respective positive controls, we observed that the activities were of little significance for the tested extracts. Although both protozoa belong to the Trypanosomatidae family, it is suggested that the differences observed for the activities must be related to the biological differences between the two species of parasite. We conclude that new experiments with the bark, leaf and flower of Tabebuia avellanedae are necessary to prove its trypanocidal and leishmanicidal action, mainly through phytochemical analysis and separation of pure substance from the extracts to prove its activity. |

Há uma necessidade urgente por novos medicamentos para a quimioterapia da doença de Chagas e leishmanioses. A Fitoterapia vem sendo uma alternativa de grande interesse para o desenvolvimento de novos fármacos. Unido a isso, o brasil possui uma das maiores, se não a maior, biodiversidade do mundo. O objetivo desta pesquisa foi testar as atividades tripanocida e leishmanicida dos extratos brutos da casca, folhas e flores de Tabebuia avellanedae. As partes do vegetal foram coletadas na região de Franca/SP e a extração realizada no laboratório de Química da UNIFRAN. Para extração foi utilizado solvente hidroalcoólico (etanol/H²O 96,4° GL). Os extratos foram aliquotados e testados em quatro concentrações (8ug/mL, 32 ug/mL, 128 ug/mL e 512 ug/ML) de cada extrato dilluídos em dimetilsufóxido (DMSO), foram avaliadas para cada parasito utilizado, Para verificação da atividade tripanocida foram utilizadas formas tripomastigotas de Trypanossoma (Schizotrypanum) cruzi (Cepa Y), e para verificação da atividade leishmanicida foram utilizadas formas promastigotas de Leishmania (Viannia) braziliensis. Os ensaios 'in vitro' foram realizados no Laboratório de Parasitologia da Faculdade de Ciências Farmacêuticas de Ribeirão Preto da Universidade de São paulo (FCFRP-USP). O método realizado para determinar a atividade do extratro foi colorimétrico pelo 3-(4,5-dimetiltiazol-2-il)-2,5-difeniltetrazolium brometo (MTT). Foi observado que o extrato bruto da casca de Tabebuia avellanedae apresentou maior atividade tripanocida e leishmanicida apresentando IC50 de 697,0 ug/mL e 774,2 uh/mL, respectivamente, quando comparado com os extratos das folhas e flores de Tabebuia avellanedae, que não demonstraram uma atividade tripanocida e leishmanicida significativa. Quando comparamos os resultados com os respectivos controles positivos, observamos que as atividades foram pouco significativas para os extratos testados. Apesar de ambos os protozoários pertencerem à família Trypanosomatidae, sugere-se que as diferenças observadas para as atividades devem estar relacionadas às diferenças biológicas entre as duas espécies de parasita.
