Resumo: Homeopathy consists of drug therapy that uses its own methods to fully interpret and treat the patient. Tropism medication is one that has in its pathogenesis characteristics similar to the patient's site or tissue, due to its electivity. The objective of this work was to investigate the influence of homeopathic medicines (MH) with bone tropism, in the culture of stem cells from human exfoliated deciduous teeth (SHED). The homeopathic medicines Calcarea carbonica (CC), Calcarea phosphorica (CP) and Symphytum officinale (SO) were tested, all in potency 30 CH; administered daily in wells containing SHED, cells validated in the Dominici Protocol and already plated. In the negative control (C-) ultrapure water by reverse osmosis was used in the wells containing SHED, while in the positive control (C +) the conventional culture medium (DMEM) was used. The 3 MH were subjected to tests of 24, 48 and 72 hours. All assays were performed in biological duplicate (from the same sample, cells in different subcultures or passages, P4 and P5), and in triplicate (n = 3) for each pass. The following cell assays were used: viability (Neutral Red), cytotoxicity (MTT) and proliferation (Crystal Violet). Statistical analysis was performed with α = 0.05 (SPSS Statistics). In P4, in the 72 h final time, the CP and SO groups showed the highest cell viability; while in P5 there were no differences among groups. In the proliferation assay, in P4, the CC and SO groups had the lowest total averages (among 24, 48 and 72 h); in P5, C+ showed the highest cell proliferation in 72 h, while the other groups showed no differences among them. As for cytotoxicity in P4, the SO presented the highest cytotoxicity in 24 h, in 48 h it was similar to the others with the exception of C +, and in 72 h presented cytotoxicity similar to C-. In P5, SO demonstrated the highest cytotoxicity only at 72 h. At Scanning Electron Microscopy (SEM), the SO showed an aspect of ultramorphology suggestive of a greater number of cells than the C +, with cells adhered and spread, confluent and in close association, without cell death. Further research was suggested, including the osteoinduction test, and the immunofluorescence test to detect specific bone molecules possibly secreted to the extracellular matrix. It can be concluded that the concentrations and potency of the tested MH influenced the cultivation of stem cells, without significant negative effects, suggesting new studies based on the same conditions.

Medicamento de Tropismo é aquele que tem em sua patogenesia características semelhantes ao local ou tecido do doente, devido à sua eletividade. Medicamentos homeopáticos (MH) podem ser investigados quanto ao estímulo sobre células-tronco para fins de terapia celular. Assim, o objetivo deste trabalho foi realizar uma breve revisão da literatura e investigar a influência de MH com tropismo ósseo, no cultivo de células-tronco mesenquimais da polpa de dente decíduo (stem cells from human exfoliated deciduous teeth, SHED). Foram testados os medicamentos homeopáticos Calcarea carbonica (CC), Calcarea phosphorica (CP) e Symphytum officinale (SO), todos na potência 30 CH; administrados diariamente em poços (placa de 96) contendo SHED, devidamente validadas no Protocolo de Dominici e já plaqueadas. No controle negativo (C-) foi usada a água ultrapurificada por osmose reversa em poços contendo SHED, e no controle positivo (C+) com SHED foi usado o meio de cultivo convencional (DMEM). O período dos ensaios foi de 24, 48 e 72 horas. Todos os ensaios foram realizados em duplicata biológica (da mesma amostra, células em subculturas ou passagens distintas, P4 e P5), e em dupla triplicata (n=6) para cada passagem. Os seguintes ensaios celulares foram usados: viabilidade (Vermelho Neutro), proliferação (Cristal Violeta) e citotoxicidade (MTT). A análise estatística foi realizada com α = 0,05. O SO com C+ foram plaqueados em placa de 24 poços para avaliação em Microscopia eletrônica de Varredura (MEV), em triplicata. Em P4, no tempo final de 72 h, os grupos CP e SO apresentaram a maior viabilidade celular; enquanto em P5 não houve diferenças entre os grupos. Quanto à proliferação, em P4 os grupos CC e SO apresentaram as menores médias totais (entre 24, 48 e 72 h); em P5, o C+ apresentou a maior proliferação celular em 72 h, enquanto os outros grupos não apresentaram diferenças entre si. Quanto à citotoxicidade, em P4 o SO apresentou a maior citotoxicidade em 24 h, sendo que em 48 h foi semelhante aos demais, com exceção do C+; em 72 h apresentou maior citotoxicidade, mas semelhante ao C-; em P5 o SO apresentou a citotoxicidade em 72 h. No entanto, à MEV o SO demonstrou aspectos morfológicos e ultraestruturais sugestivos de maior número de células do que o C+, com células aderidas e espraiadas, confluentes e em íntima associação, sem morte celular. Foram sugeridas novas pesquisas, incluindo-se o teste de osteoindução, e de imunofluorescência para detecção de moléculas ósseas específicas, possivelmente secretadas para a matriz extracelular.
