Resumo: Asthma affects a significant portion of the population, with a high social and economic cost. Photobiomodulation therapy (FBM) is a treatment that demonstrates efficacy in lung diseases by reducing inflammatory symptoms, with low cost and without side effects. The objective was to evaluate the effects of FBM therapy in an experimental model of chronic lung inflammation induced by ovalbumin (OVA). Balb/C mice were divided into 6 groups: Control, OVA, and OVA+FBM (OVA+1J, OVA+3J, OVA+5J, OVA+7.5J). Inflammation was induced by OVA sensitization (day 0 and day 14) and orotracheal challenge from day 21 (3 days/week for 5 weeks). One hour after the challenge, irradiation with a diode laser (660nm, 100mW) was applied, using different irradiation times (10s, 30s, 50s and 75s), at three different points, one below the trachea and one in each lung lobe. Twenty-four hours after the last treatment, the animals were anesthetized and euthanized for collection of bronchoalveolar lavage (BAL) and lungs. Parameters such as total and differential cell counts, BAL cytokine levels, mucus production, collagen deposition, quantification of macrophages, neutrophils, eosinophils in the lung, as well as analyzing the rate of T lymphocytes (CD4 and CD25) and T controllers ( Foxp3) and IL-10 release by cell assays. The data were left to the One-way ANOVA test followed by the Newman-Keuls post-test. Significance levels adjusted to 5% (p<0.05). Therapy with FBM was able to reduce the pulmonary inflammation observed by reducing the total and differential number of cells in the BAL, mainly with regard to the number of eosinophils, in addition we verified the decrease in the levels of the pro-inflammation cytokines IL-4, IL- 5 and IL-13 in BAL in the group treated with FBM, as well as the increase in IL-10 levels. for the values of CD4+IL-10+ lymphocytes we witnessed an increase in all groups treated with FBM. These results revealed that FBM therapy had a beneficial effect on cell migration to the lung, as well as the stimulation of IL-10 production. We also noticed a reduction in the deposition of mucus and collagen in the airways in the groups submitted to FBM therapy with the energies of 3J and 5J. Therefore, we conclude that the use of FBM therapy in chronic airway inflammation attenuated the inflammatory process, as well as related functional and pulmonary parameters, probably due to the increase in regulatory T cells.

A terapia com fotobiomodulação (FBM) é um tratamento que demonstra eficácia em doenças pulmonares por reduzir parâmetros inflamatórios, com baixo custo e sem efeitos colaterais. O objetivo foi avaliar os efeitos da terapia FBM em modelo experimental de inflamação alérgica pulmonar crônica induzida por ovalbumina (OVA). Camundongos Balb/C foram divididos em 6 grupos: Controle, OVA, e OVA+FBM (OVA+1J, OVA+3J, OVA+5J, OVA+7,5J). A inflamação foi induzida por sensibilização com OVA (dia 0 e dia 14) e desafio orotraqueal a partir do dia 21 (3 dias/semana por 5 semanas). Uma hora após o desafio foi aplicado a irradiação com laser de diodo (660nm, 100mW), utilizando diferentes tempos de irradiação (10s, 30s, 50s e 75s), em três pontos distintos, um abaixo da traquéia e um em cada lobo pulmonar. Vinte e quatro horas após o último tratamento, os animais foram anestesiados e eutanasiados para a coleta de lavado broncoalveolar (LBA) e dos pulmões. Parâmetros como contagem de células totais e diferenciais, níveis de citocinas no LBA, produção de muco, deposição de colágeno, quantificação de macrófagos, neutrófilos, eosinófilos no pulmão, bem como analisamos a porcentagem de linfócitos T (CD4 e CD25) e células T reguladoras (Foxp3) e a liberação de IL-10 por essas células. Os dados foram submetidos ao teste One-way ANOVA seguido do pós-teste de Newman-Keuls. Níveis de significância ajustados para 5% (p<0,05). A terapia com FBM foi capaz de reduzir a inflamação pulmonar observada pela redução do número total e diferencial de células no LBA, principalmente com relação ao número de eosinófilos, além disso verificamos diminuição dos níveis das citocinas pró-inflamatórias IL-4, IL-5 e IL-13 no LBA no grupo tratado com FBM, bem como o aumento dos níveis de IL-10.Observamos o aumento de CD4+CD25+Foxp3+ para os grupos tratados com laser (3J, 5J e 7,5J), Já para os valores de linfócitos CD4+IL-10+ presenciamos um aumento em todos os grupos tratados com FBM. Esses resultados revelaram que a terapia com FBM apresentou um efeito benéfico na migração celular para o pulmão, bem como a estimulação da produção de IL-10. Notamos também a redução da deposição de muco e de colágeno nas vias aéreas nos grupos submetidos à terapia com FBM com as energias de 3J e 5J.
