Efeito da laserterapia na biomodulação da osteogênese em defeitos críticos confeccionados em calota craniana de ratos

Resumo: The presented study aims at evaluating, through histological and morphometrical analysis, the biomodulation of the process of osteogenesis in critical defects made in skull bones of rats, submitted to infrared laser diodo therapy (GaAIAs). There were used 40 rats, weight between 300 and 500 grams, distributed randomly in two groups, the control (GC-I and II) and the experimental (GT-I and II) and 4 subgroups according to the period of observation of the animals. For the control and experimental group, there were destinated 20 animals, equally distributed. The subgroups received a low-level laser therapy (LLLT) in a critical-size bone defect of 4mm made in the right parietal bone. In the group GC-I and II all the cirurgical procedures were made, but without using laser. In the group GT-I and II was used infrared laser (λ= 830 nm, 2 J/cm2, 90mW, 27 s) in a ponctual and steady way. The protocol for radiation was established within breaks of 48 hours, beginnig right after the suture of the cirurgical procedure and following it until six days after the surgery. The animals were sacrified at 07 and 21 days after the surgery. For histological analysis, it was used the commom technique of HE. The plates were studied according to descriptive and morphometric analysis. The tissues evaluated, in the descriptive analysis, included a new formation of bone.For morphometrical analysis, there were determinated the average areas of new forming trabeculated bones related to the total area of the defect. The obtained results show that, according to a morphometric analysis, the positive biomodulation in bone of the groups GT-I and II presented a bigger area of trabeculated bone when compared to groups which did not received LLLT. The results allow to suggest that low-level laser therapy acts like biomodulation in bone, stimulating the osteogenesis, being able to be used in the process of bone repairing.

Foram utilizados 40 ratos machos da linhagem Wistar, com peso entre 300 a 500 gramas, distribuídos aleatoriamente em dois grupos, o controle (GC-I e II) e o experimental (GT-I e II), e quatro subgrupos de acordo com o período de observação dos animais. Para os grupos controle e experimental destinaram-se 20 animais, igualmente distribuídos. Os subgrupos experimentais receberam a terapia laser de baixa potência (LLLT), em um defeito ósseo crítico de 4mm confeccionado no osso parietal direito do animal. No grupo GC-I e II todo o protocolo cirúrgico foi realizado, porém sem a aplicação do laser. No grupo GT-I e II foi utilizado o laser infravermelho (λ= 830 nm, 2 J/cm2, 90mW, 27 s), de forma pontual e contínua. O protocolo de radiação foi estabelecido com intervalos de 48 horas, iniciando-se imediatamente após a sutura do procedimento cirúrgico e seguindo-se até o sexto dia de pós-operatório. Os animais foram mortos aos 07 e 21 dias após o procedimento cirúrgico. Para análise histológica, executou-se o processamento de rotina para a técnica de HE.As lâminas foram estudadas segundo análise descritiva e morfométrica. O fenômeno tecidual avaliado, na análise descritiva, incluiu a neoformação óssea. Para a análise morfométrica determinaramse as médias das áreas de trabeculado ósseo neoformado em relação a área total do defeito. Os resultados obtidos demonstraram que, segundo a análise morfométrica, a biomodulação óssea positiva evidenciada nos grupos GT-I e II apresentou maior área de trabeculado ósseo quando comparada aos grupos não submetidos à LLLT. Os resultados permitiram sugerir que a laserterapia de baixa potência no protocolo estabelecido atua como biomoduladora óssea, estimulando a osteogênese, podendo ser utilizada como coadjuvante no processo de reparo ósseo.Made available in DSpace on 2011-12-27T14:14:07Z (GMT).
