Resumo: In this study, we evaluated the interaction of Diabrotica speciosa (Germar) (Coleoptera: Chrysomelidae) and six commercial cultivars of bean (Phaseolus vulgaris), in order to characterize antixenosis and/or antibiosis resistance. The cultivars BRS Campeiro, IPR Uirapuru, BRS Esplendor, IPR Tuiuiu, IPR Tiziu and FTS Soberano were grown in the greenhouse and used in laboratory bioassays. To evaluate antixenosis, free-choice bioassays were performed in petri dishes, releasing six newly emerged adults of D. speciosa and offering to them six leaf discs (30 mm diameter), one of each cultivar, during 24 hours. In addition, no-choice bioassays were performed in plastic containers whith a leaf disc (30 mm diameter) of each cultivar were offered as the only source of food to one adult. Each foliar disc were evaluated before and after consumption by D. speciosa, registering the area damaged. The antibiosis was evaluated in insects biology studies, which were done in plastic cages with one newly-emerged couple (N=30), registering the survival and oviposition of adults at intervals of 48 hours.The leaf trichomes were analyzed, inserting 20 leave`s seccions (1 mm²) in scanning electron microscope, and registering the number of hookedtrichomes, acicular trichomes and glandular trichomes on abaxial (N = 10) and adaxial (N = 10) surfaces. In free-choice bioassays adults showed less preference to cultivars IPR Tuiuiú, IPR Uirapuru, BRS Esplendor, consuming less leaf area. On the other hand, in no-choice bioassays insects also fed up all cultivars, and the consumption area was similar. The isolated analysis of biological parameters did not differ among the insects that fed the different genotypes. From the fertility life table it was obtained difference in the mean interval between generations (IMG) as the BRS Esplendor genotype showed a lower average range compared to BRS Campeiro. And the net reproductive rate (Ro) of BRS Campeiro genotype differed from the others because the insects fed with this genotype showed higher reproduction. As to the trichomes, IPR Tuiuiu genotype had the lowest density acicular trichomes on the adaxial side face, and the glandular trichomes and hooked did not differ in the side faces of leaves. In the leaf center BRS Campeiro genotypes IPR Tuiuiu and BRS Esplendor had the greatest density of glandular trichomes in adaxial face, and in the abaxial face only the BRS Campeiro and the IPR Tuiuiu remained with higher density of this tricoma followed by IPR Tiziu. For acicular trichomes BRS Campeiro differed presenting the lowest density in the face adaxial. Hooked trichomes showed higher densities in the abaxial face of genotypes IPR Tiziu, and IPR Tuiuiu and IPR Uirapuru. The total trichomes (adaxial + abaxial) did not differ between the genotypes on the side, but the center genotypes IPR Tiziu, IPR Tuiuiú and IPR Uirapuru were the densest. In this study, there were no differences that can say resistance to D. speciosa in the evaluated treatments, however it was obtained are presented important information about the feeding behavior of the bean resistance to D. speciosa in bean and life table, which may provide a basis for future work in the area.

Bioensaios de laboratório foram realizados com as cultivares BRS Campeiro, IPR Uirapuru, BRS Esplendor, IPR Tuiuiú, IPR Tiziu e FTS Soberano, as quais foram cultivadas em casa de vegetação. Para avaliar a ocorrência de antixenose, foram realizados bioensaios com chance de escolha, oferecendo um disco foliar (30 mm de diâmetro) de cada cultivar em recipientes plásticos circulares, liberando-se seis adultos para a livre escolha e alimentação durante 24 horas. Além disso, foram realizados bioensaios sem chance de escolha, utilizando um recipiente plástico contendo um disco foliar (30 mm de diâmetro) de cada cultivar, como única fonte de alimentação para os adultos. Estes discos foram avaliados antes e após alimentação, com relação a área foliar consumida. Para caracterizar a ocorrência de antibiose, foram realizados estudos da biologia dos insetos, avaliando-se a longevidade e a fecundidade, a partir da montagem de casais e manutenção em gaiolas durante toda a fase adulta, os quais foram observados em intervalos de 48 horas, registrando-se a sobrevivência e o número de ovos depositados. Para a caracterização e contagem dos tricomas foliares das cultivares, vinte secções (1 mm²) na lateral e vinte secções no centro (1 mm²) dos folíolos foram analisadas em microscópio eletrônico de varredura (MEV), registrando-se o número de tricomas unciformes, aciculares e glandulares nas faces abaxial (N=10) e adaxial (N=10). Nos bioensaios com chance de escolha, os adultos de D. speciosa apresentaram menor preferência em relação a cultivar IPR Tuiuiú, IPR Uirapuru, BRS Esplendor, consumindo menor área foliar. Por outro lado, em bioensaios sem chance de escolha os insetos alimentaram-se igualmente de todas as cultivares avaliadas, sendo que o consumo em área foi semelhante. As análises isoladas dos parâmetros biológicos não apresentaram diferenças entre os insetos que se alimentaram dos diferentes genótipos. A partir da tabela de vida de fertilidade obteve-se diferença no intervalo médio entre gerações (IMG), pois o genótipo BRS Esplendor apresentou um intervalo médio menor em relação ao BRS Campeiro. E a taxa liquida de reprodução (Ro) do genótipo BRS Campeiro diferiu dos demais, pois os insetos alimentados com esse genótipo apontaram maior reprodução. Quanto aos tricomas, o genótipo IPR Tuiuiú apresentou a menor densidade de tricomas aciculares na face adaxial lateral, sendo que os tricomas glandulares e unciformes não diferiram nas faces da lateral do folíolo. No centro foliar os genótipos BRS Campeiro, IPR Tuiuiú e BRS Esplendor retrataram a maior densidade de tricomas glandulares na face adaxial, sendo que na face abaxial apenas o BRS Campeiro e o IPR Tuiuiú permaneceram com maior densidade deste tricoma seguido do IPR Tiziu. Para os tricomas aciculares o BRS Campeiro se diferenciou apresentando a menor densidade na face adaxial. Tricomas unciformes mostraram densidades maiores na face abaxial dos genótipos IPR Tiziu, IPR Tuiuiú e IPR Uirapuru. O total de tricomas (abaxial+adaxial) não apresentou diferenças entre os genótipos na lateral, entretanto no centro os genótipos IPR Tiziu, IPR Tuiuiú e IPR Uirapuru foram os mais densos. No presente estudo, não houve diferenças que possam afirmar resistência a D. speciosa nos tratamentos avaliados. Contudo, obtiveram-se informações importantes sobre o comportamento alimentar de D. speciosa em feijão e tabela de vida, as quais podem fornecer subsídios para futuros trabalhos na área.Made available in DSpace on 2016-09-20T12:12:44Z (GMT).
