Resumo: In Brazil, the optimal climate conditionsenable banana production in the entire country. However, some phytopatogens such as fungi, nematodes, insects and v?rus compromise yield and fruit quality. Among the v?rus diseases, banana streak v?rus (BSV) is the most important, whereas more than fifteen different species have been identified worldwide. The objectives of the present study were to identify BSV species in germplasm bank of Embrapa and in different banana production regions in Brazil by molecular characterization and indexation using IC-PCR(Immunocapture-PCR) + DNase, as well as map the presence of infective fragments in commercial germplasm and those of genome BB used in the crosses. Leaves of 300 banana accessions from the Embrapa ex situ germplasm collection and 39in vitro samples, were collected. Two-hundred and twenty-seven samples from eight different banana production regions in Brazil, were also collected. Twenty-three species-specific primers for detection of endogenous BSV (eBSV) for species Goldfinger(eBSGFV), Imov? (eBSIMV) and Obinol'Ewai (eBSOLV) were also used for the analysis of incidence and prevalence of viral species. The analysis of the 300 accessions of BAG showed that most samples were negative for eBSV and of the positive banana samples for eBSV, 20 were infected by episomal BSV of the BSOLV and BSGFV species. None of the analyzed samples contained BSV episomal of the BSIMV species. Most of the samples analyzed from the eight different banana producing regions in Brazil was positive for the presence of at least one of the eBSV species and only samples from the State of S?o Paulo and one from Santa Catarina presented episomal BSV. The study with hybrids developed by Embrapa identified presence of the infective fragment GF7 associated to the BSGFV species. Regarding the presence of the infective fragment associated to the BSOLV species, only the Thap Maeo cultivar was positive. The other genotypes amplified only the non-infective fragment, OL2. All eight accessions with BB genome presented either the infective fragment for the BSOLV species as well as for the BSGFV species. The IC-PCR + DNAse indexation confirmed the low presence of episomal BSV in the analyzed samples.

Entre as viroses mais comuns, destaca-se o v?rus das estrias da bananeira (BSV), que possui mais de quinze diferentes esp?cies identificadas em todo o mundo. O presente estudo teve como objetivos realizar um levantamento das esp?cies de BSV presentes no Banco Ativo de Germoplasma de Banana da Embrapa e em diferentes regi?es produtoras de banana do Brasil, por meio da caracteriza??o molecular e indexa??o a partir do uso da t?cnica IC-PCR (Immunocapture-PCR) +DNase, bem como mapear a presen?a de fragmentos infectivos no germoplasma comercial e diploides BB com potencial de uso em cruzamentos. Para tanto, foram coletadas amostras de folhas de 300 acessos de bananeira da cole??o de germoplasma da Embrapa provenientes do cultivo ex situ e 39 amostras do cultivo in vitro. Tamb?m foram coletadas 237 amostras provenientes de oito diferentes regi?es produtoras de banana do Brasil. Para a an?lise de incid?ncia e preval?ncia de esp?cies do v?rus, foram utilizados 23 iniciadores esp?cie-espec?ficos para detectar a presen?a do v?rus das estrias da bananeira end?geno (eBSV), a saber Goldfinger (eBSGFV), Imov? (eBSIMV) e Obinol'Ewai (eBSOLV). A an?lise dos 300 acessos do BAG permitiu observar que a maioria apresentou resultados negativos quanto ? presen?a do eBSV. Das amostras de bananeira positivas, 20 apresentaram infec??o por BSV epissomal das esp?cies BSOLV e BSGFV. Nenhuma amostra apresentou o BSV epissomal da esp?cie BSIMV.Dentre os acessos do cultivo in vitro indexados n?o foi constatado a presen?a do BSV epissomal das esp?cies BSGFV e BSIMV. Tamb?m se observou que a maioria das amostras analisadas das oito diferentes regi?es produtoras do Brasil foi positiva para a presen?a de ao menos uma esp?cie de eBSV e apenas as amostras dos Estados de S?o Paulo e outra de Santa Catarina apresentaram a presen?a do BSV epissomal. O estudo com os h?bridos desenvolvidos pela Embrapa identificou a presen?a do fragmento infectivo GF7 associado ? esp?cie BSGFV. Em se tratando da presen?a do fragmento infectivo associado ? esp?cie BSOLV, apenas a cultivar Thap Maeo mostrou-se positiva. Os outros gen?tipos amplificaram apenas o fragmento n?o infectivo OL2. Observou-se que todos os oito acessos de genoma BB apresentam tanto o fragmento viral infeccioso para a esp?cie BSOLV quanto o fragmento infeccioso para a esp?cie viral BSGFV.
