Resumo: This study evaluated the antifungal and antibiofilm potential of the hydroalcoholic extract of the bark of Anadenanthera colubrina vell. Brenan, known as Angico and White Angico. The antifungal activity was initially evaluated by the microdilution technique in broth with minimum inhibitory concentration (MIC) and Minimum Fungicide Concentration (CFM). The potential antibiofilm was tested in mature biofilms formed by Candida species and analyzed by counts of CFU / mL and scanning electron micrograph (SEM). In vitro toxicity was analyzed in Raw 264.7 macrophages and systemic toxicity was evaluated in vivo in the Galleria mellonella model. A strong antifungal potential of the extract against strains of the genus Candida was observed, with the following MIC values: Candida albicans ATCC 5314 and ATCC 90028 (19.5 μg / mL), Candida tropicalis ATCC 750 (5000 μg / mL), Candida Glabrata ATCC 90030 (1250 μg / ml), Candida parapsilosis ATCC 22019 and Candida krusei ATCC 6258 (39 μg / ml). The treatment with the extract reduced the number of CFU / mL and was able to cause structural alteration in the biofilm and cellular destruction, observed in SEM, presenting a promossora antibiofilm activity against Candida. A. colubrina presented moderate toxicity in vitro in RAW 264.7 macrophages culture. In the in vivo assay the extract did not affect the viability of the larvae at doses below 100 mg / kg, presenting low toxicity. Considering its high antifungal potential and its low in vivo toxicity, A. colubrina extract is a strong candidate for the development of a new drug for the treatment of oral candidiasis.

Brenan, conhecida como angico e angico branco. A atividade antifúngica foi inicialmente avaliada pela técnica da microdiluição em caldo com obtenção da concentração inibitória mínima (CIM) e Concentração Fungicida Mínima (CFM). O potencial antibiofilme foi testado em biofilmes maduros formados por espécies de Candida e analisados por contagem de UFC/mL e microcopia eletrônica de varredura (MEV). A toxicidade in vitro foi analisada em macrófagos Raw 264.7 e a toxicidade sistêmica foi avaliada in vivo em modelo de Galleria mellonella. Observou-se um forte potencial antifúngico do extrato contra cepas do gênero Candida, obtendo-se os seguintes valores da CIM: Candida albicans ATCC 5314 e ATCC 90028 (19.5 μg/mL), Candida tropicalis ATCC 750 (5000 μg/mL), Candida glabrata ATCC 90030 (1250 μg/mL), Candida parapsilosis ATCC 22019 e Candida krusei ATCC 6258 (39 μg/mL). O tratamento com o extrato reduziu o número de UFC/mL e foi capaz de causar alteração estrutural no biofilme e destruição celular, observadas em MEV, apresentando uma promossora atividade antibiofilme contra Candida. A. colubrina apresentou moderada toxicidade in vitro em cultura de macrófagos RAW 264.7. No ensaio in vivo o extrato não afetou a viabilidade das larvas em doses abaixo de 100 mg/kg, apresentando baixa toxicidade.
