Resumo: Of the several species that make up the Candida genus, some have been shown to be associated with cases of human infection, with Candida albicans being the main pathogen, despite studies confirming cases of infection with other species of the genus. There are several drugs available for the treatment of oral candidiasis, and nystatin is the drug of first choice. However, in addition to its known side effects, nystatin does not present considerable residual action, which impairs treatment efficiency. Given this, this work aims to develop a mucoadhesive gel for the treatment of oral candidiasis, constituted by extract of S. brasiliensis, encapsulated in maltodextrin nanoparticles. After obtaining the extract by maceration followed by rotary evaporation, the assay was performed to verify the antifungal activity of the same, by the microdilution method in broth, using 96-well microplates. This was done with the assay to elucidate the possible mechanism of action of the extract on the wall and fungal cell membrane, by the microdilution method. Then, the phytochemical prospection was carried out, searching for the main phytocomponents of the plant, by spectrophotometry, in order to associate their presence with the antifungal action of the plant. Subsequently, the microparticles of maltodextrin were developed by Spray Drier, encapsulating the extract, varying the flow rate of the pump. Subsequently, the microparticle that had the best fungal activity was selected to follow the steps of thermal compatibility, formulation and evaluation of its effectiveness.

Existem diversos fármacos disponíveis para o tratamento da candidíase oral, sendo que a nistatina é o medicamento de primeira escolha, porém, além de seus conhecidos efeitos colaterais, a nistatina não apresenta ação residual considerável, o que prejudica a eficiência do tratamento. Visto isso, este trabalho objetiva desenvolver um gel mucoadesivo para o tratamento de candidíase oral, constituído por extrato de S. brasiliensis, encapsulado em nanopartículas de maltodextrina. Após a obtenção do extrato por maceração seguido de evaporação rotativa, foi realizado o ensaio para verificação da atividade antifúngica do mesmo, pelo método de microdiluição em caldo, utilizando microplacas de 96 cavidades. Feito isto, seguiu-se com o ensaio para elucidação do possível mecanismo de ação do extrato sobre a parede e membrana celular fúngica, pelo método de microdiluição. Em seguida, realizou-se a prospecção fitoquímica, buscando os principais fitocomponentes da planta, por espectrofotometria, objetivando associar a presença desses à ação antifúngica do vegetal. Posteriormente, desenvolveu-se as micropartículas de maltodextrina, por Spray Drier, encapsulando-se o extrato, variando-se a vazão da bomba. Posteriormente, foi selecionada a micropartícula que melhor apresentou atividade fúngica para que se seguissem as etapas de compatibilidade térmica, formulação e avaliação de sua eficácia.Coordenação de Aperfeiçoamento de Pessoal de Nível Superior - CAPESapplication/pdfUniversidade Estadual da ParaíbaPrograma de Pós-Graduação em Ciências Farmacêuticas - PPGCFUEPBBRPró-Reitoria de Pós-Graduação e Pesquisa - PRPGPPró-Reitoria de Pós-Graduação e Pesquisa - PRPGPMucosadhesive gelOral candidiasisCIENCIAS DA SAUDEPlantas medicinaisCandidíase oralGel mucoadesivoSchinopsis brasiliensisDesenvolvimento de gel mucoadesivo para o tratamento de candidíase oral a partir do extrato de Schinopsis brasiliensis Englerinfo:eu-repo/semantics/publishedVersioninfo:eu-repo/semantics/masterThesisPereira, Jozinete Vieirahttp://lattes.cnpq.br/8041850574045487Vargas, Mara Rubia Winter dehttp://lattes.cnpq.br/5244707551037970Medeiros, Ana Claudia Dantas dehttp://lattes.cnpq.br/0640671064146120http://lattes.cnpq.br/3822476112643957Araújo, René Monteiroinfo:eu-repo/semantics/openAccessporreponame:Repositório Institucional da Universidade Estadual da Paraíba (UEPB)instname:Universidade Estadual da Paraíba (UEPB)instacron:UEPBLICENSElicense.txtlicense.txttext/plain;
