Resumo: The study aimed to characterize the resistance of F2:3 progenies of maize inoculated with Colletotrichum graminicola, genetic parameters and map genomic regions associated with quantitative resistance alleles to anthracnose stalk in tropical maize determining the effects of each locus on resistance/susceptibility C. graminicola. Individuals of the F2 generation were selfed to generate F2:3 progenies, which were evaluated phenotypically for resistance to anthracnose stalk in three experiments. The treatments were consisted of 121 F2:3progenies, parental lines (L04-2 and L99-4) and the F1 generation, which were used as control. The experimental plots were arranged in a randomized block design with three replications. Each plot consisted in one row of 3 m in length with rows spaced 0.90 m apart, with a total of 15 plants per plot. The plants were inoculated in the anthesis stage by injecting 1 mL of spore suspension into the first or second elongated internode above ground. The data was collected 30 days after inoculation by evaluating the internal lesion length (CINT), external lesion length (CEXT) in cm and the number of discolored internodes (INT). From the phenotypic evaluations of 121 F2:3 progenies genetic parameters were associated with resistance to all three forms of assessment (CINT, INT and CEXT) in each experiment. The association of microsatellites and AFLP markers to resistance alleles was performed using multiple regression. AFLP and SSR loci used to form the construction of the linkage map by MAPMAKER version 3.0. Detection of the QRL was performed by composite interval mapping method using QTLCartographer program. The experimental results showed the existence of high genetic variability among the F2:3 progenies for resistance/susceptibility to C. graminicola. The forms of assessment of anthracnose stalk (CINT, INT and CEXT) were equally effective in discriminating susceptible from resistant individuals. The high broad-sense heritability confirms the great contribution of genetic variance for resistance, which ensures genetic gain for artificial selection in this pathosystem. The AFLP marker E55534 individually explained the greatest proportion of the phenotypic variation for the internal length (- 28.43%), external lesion length (- 27.70%) and the number of discolored internodes (- 27.45%) in F2:3 progenies of maize. The composite interval mapping identified 31 QRLs to anthracnose stalk segregating in the mapping population, which were mapped across the 10 linkage groups. This is the first report of the QRL to anthracnose stalk mapped on chromosomes 2, 3 and 10. At large number of mapped QRL is suggested that the inheritance of resistance in this tropical germplasm is more complex, conditioned by a large number of genes, some large phenotypic effects and the vast majority with small effects for resistance in this pathosystem.

Os indivíduos da geração F2 foram autofecundados para gerar as progênies F2:3, as quais foram avaliadas fenotipicamente para resistência à antracnose do colmo em três experimentos de campo. Os tratamentos foram constituídos de 121 progênies F2:3, linhagens parentais (L04-2 e L99-4) e da geração F1, os quais foram utilizados como tratamentos controle. O delineamento experimental foi o de blocos aleatorizados, com três repetições. Cada parcela foi constituída de 3 m de comprimento e 0,90 m na entrelinha, com aproximadamente 15 plantas por parcela. As plantas dos experimentos foram inoculadas no estádio de florescimento pleno da cultura, através da injeção de 1mL da suspensão de esporos, no meio do primeiro ou segundo internódio visível acima do solo. A avaliação dos sintomas foi realizado 30 dias após a inoculação, através da avaliação do comprimento interno de lesão (CINT), comprimento externo de lesão (CEXT) em cm e do número de internódios descoloridos (INT). A partir das avaliações fenotípicas das 121 progênies F2:3 foram estimados os parâmetros genéticos associados a resistência para as três características avaliadas (CINT, CEXT e INT) em cada experimento. A associação dos marcadores microssatélites e AFLPs a alelos de resistência foi realizada através da regressão linear múltipla. Locos SSR e AFLPs foram utilizados na construção do mapa de ligação através do MAPMAKER versão 3.0. A detecção dos QRLs foi realizada pelo método de mapeamento por intervalo composto utilizando o programa QTLCartographer. Os resultados experimentais evidenciaram existência de grande variabilidade genética entre as progênies F2:3 de milho para resistência/suscetibilidade à C. graminicola. As formas de avaliação da antracnose do colmo (CINT, CEXT e INT), foram igualmente eficientes em discriminar os indivíduos resistentes dos suscetíveis. Os elevados coeficientes de herdabilidade no sentido amplo confirmam a grande contribuição da variância genética para resistência, o que assegura ganhos genéticos com a seleção artificial neste patossistema. A regressão linear múltipla possibilitou identificar marcadores microssatélites e AFLPs altamente associados a alelos de resistência à C. graminicola. O marcador AFLP E55534 explicou individualmente a maior proporção da variação fenotípica para o comprimento interno (- 28,43%), comprimento externo de lesão (- 27,70%) e para o número de internódios descoloridos (- 27,45%) nas progênies F2:3 de milho. O mapeamento por intervalo composto identificou 31 QRLs à antracnose do colmo segregando na população de mapeamento, os quais foram mapeados entre os 10 grupos de ligação. Este é o primeiro relato de QRLs à antracnose do colmo mapeados nos cromossomos 2, 3 e 10. Pelo grande número de QRLs mapeados sugere-se que a herança da resistência neste germoplasma tropical seja mais complexa, condicionada por um grande número de genes, sendo alguns de grande efeito fenotípico e a grande maioria com pequenos efeitos para a resistência neste patossistema.Made available in DSpace on 2017-07-21T19:59:45Z (GMT).
