The aim of this study was to evaluate the importance of the IL-1B (+3954) polymorphism for severe chronic periodontitis in comparison with healthy patients. Genotypic and allelic frequencies related to this polymorphism were also evaluated in a general population with unknown periodontal status. Of a total of 186 selected patients from Rio de Janeiro State, 50 had severe chronic periodontitis (PC) (51,16  5,37), 35 were controls (C) (43,97  9,60) e 101 were from the general population (PG) (51,06  15,52). DNA samples of the GP group belong to a DNA collection previously constructed In such group, which is equilibrium, the prevalence of the CT genotype was 53% and the allele frequency was 0,59. No difference was observed between whites and non-whites. DNA samples from patients belonging to CP and C groups were obtained after collecting buccal epithelial cells from the inner portion from the check with a swab. The region containing the polymorphism of interest was amplified by the PCR technique (polymerase chain reaction) using a pair of specific flanking oligonucleotides of the locus +3954 of IL-1B gene. PCR products were digested by TaqI enzyme and analyzed by 8% polyacrylamide gel and electrophoresis. The T allele frequency in severe chronic periodontitis group was significantly higher as compared to the control group. T allele frequencies in CP subgroups divided by skin color and smoking habit were significantly higher in whites and smokers as compared to the control group. Our results suggest that the T3954 allele of IL-1B may be considered a risk indicator for the occurrence of severe chronic periodontitis in this group of individuals from Rio de Janeiro State.
