These materials are present in several products used by society. Silver nanoparticles (AgNPs) are used as pesticides, food additives, makeup and skincare products, and electronic compounds. Although their presence in the market is consolidated, toxicological and environmental effects are unknown. For this reason, new studies and methods for control and monitoring are necessary. There is a lack of methods for the determination and sizing of NPs in biological samples. Traditional analytical techniques for sizing (such as Dynamic Light Scattering and Atomic Force Microscopy) present some disadvantages as the interferences of the matrix where the NPs are present and low analytical frequency. The single particle inductively coupled mass spectrometry (spICP-MS) method presents as a new tool to overcome the detection, sizing and quantification of metallic NPs in complex matrices. Therefore, the current work aimed the study AgNPs detection in human matrices: blood-plasma, sweat, urine and saliva. It was studied the influence of sample dilution, concentration of NPs in the samples and the storage time in AgNPs detection, sizing and quantification. Firstly, it was observed a 1:20 matrix dilution was able to allow AgNPs 2 ìg L -1 detection in urine, saliva and plasma; a 1:10 matrix dilution was able to allow AgNPs 2 ìg L -1 detection in sweat. Regarding the levels of AgNPs, concentrations ³ 5 ìg L-1 presented an adverse signal-background differentiation profile for detection of AgNPs. Higher concentrations of AgNPs in solution caused interference of matrix compatibility due to Ag+ detection by spICP-MS, generating false-positive results. Finally, it was observed that a 144h storage for AgNPs 2 ìg L -1 solutions in 1:20 diluted urine and water allowed the AgNPs detection. AgNPs 2 ìg L -1 did not present a good signal-background differentiation in a 1:20 diluted plasma solution after a 144h storage.
