One of the factors that contributes to biochemical damage is the oxidative stress, which is involved in neurodegenerative diseases as well as neuropsychiatric disorders. BPPs are structurally classified as oligopeptides, with a high proline content, and are present in the venom of Bothrops jararaca and Bitis nasicornis, both members of the Viperidae family. These peptides enhance the synthesis of Larginine, an amino acid that plays a crucial role in many metabolic pathways such as the formation of polyamines and agmatine, both with neuroprotective properties. The objective of There are few studies about the activity of Bradykinin-enhancing peptides (BPPs) against oxidative stress this study was to evaluate the cytoprotection mediated by peptide components from Bothrops jararaca and Bitis narcicornis snake venoms in astrocyte-type glial cells and neurons against oxidative stress caused by H2O2. In the present study, the use of conditioned medium from C6 cells subjected to oxidative stress for PC12 cells was explored. C6 cells were plated at densities 5 x 103 and 1 x 104 with different concentrations of 10, 1 and 0% FBS subjected to oxidative stress by H2O2 at concentrations of 2, 1, 0,5, and 0,3 mM for 24 h, so that the medium was later transferred to 5 x 103 PC12 cells and maintained for 24 h. The use of this protocol was considered inappropriate for carrying out cytoprotection tests with tested peptides, as the integrity of C6 cells was reduced. However, with the conditioning of culture medium there was an increase in the integrity of PC12 cells due to the presence of cellular secretomes of the C6 cells. In tests evaluating the cytotoxic effects of FBP (10, 1 and 0.01 µg.mL-1 ) and BPPs 7a and 10c (10, 1 and 0.01 µM) from Bothrops jararaca in C6 cells for 24 h, it was verified that FBP at concentrations 10, 1 and 0.01 µg.mL-1 increased cellular integrity, while BPP-7a did not show significant effects. BPP-10c showed an increase in cellular integrity in a dose-dependent manner, with the lower concentrations being more effective. The evaluation of cytotoxicity of BPPs 1, 2 and 8 (10, 1 and 0.01 µM) from Bitis nasicornis in C6 and PC12 cells for 24 h demonstrated that BPP1 was the only one capable of reducing cellular integrity at a concentration of 10 µM. In PC12 cells, it was noticed that BPPs 2 and 8 promoted an increase in cellular integrity at the lowest concentrations (0.01 µM). In cytoprotection assays, C6 cells (5 x 103 ) were pre-treated with different concentrations of FBP (25 to 0.04 µg.mL-1 ) and BPPs 7a or 10c (25 to 0.04 µM) for 4h, followed by the addition of H2O2 (0.4 mM) in the presence of the same concentrations of peptides, being maintained at 37°C for another 20h. FBP and BPPs 7a and 10c did not provide cytoprotection to C6 cells against oxidative stress, but were able to potentiate the cytotoxic effects of H2O2 at low concentrations. In cytoprotection assays in PC12 cells (5 x 103 ), BPPs 1, 2 and 8 (25 to 0.04 µM) were pretreated for 4h and then subjected to oxidative stress with H2O2 (0.5 mM) with the same peptide concentrations. BPP1 did not demonstrate a cytoprotective effect on PC12 cells, but BPPs 2 and 8 were effective in providing cytoprotection.
