Resumo: The endothelium plays a crucial role in the vascular tone, releasing relaxing and contracting factors – among the first ones the gas nitric oxide, produced from the amino acid L-arginine and a group of enzimes called nitric oxide synthases (NOS). This study evaluated the effects of L-arginine as well as NOS inibitors on vascular escape and tachyphylaxis of rabbit kidneys. Furthermore the effects of diabetes and hypothermia were studied also. Normal and alloxan treated diabetic rabbit kidneys were perfused with Krebs-Henseleit solution in a non-recirculating system and norepinephrine (NO) 1 microM was infused for three subsequent periods of 20 min each, with an inteval of 10 min for drug wash-out. In the control group L-arginine and D-arginine were added to the solution (both 1 microM) as well as the NOS inibitors L-NMA (10 microM) and LNOARG (10 microM). This group was also submitted to hypothermia (25o C). The kidneys from diabetic animals were separated in non treated and treated with GSH (1 microM) and rutin (1 microM). The treatment with the NOS inibitor LNOARG increased the basal (steady state) pressure (p<0.001). The L-arginine treatment decreased both basal (p<0.05) and maximal (p<0.01) pressures (after NOR infusion). The nitro-methyL-L-arginine treatment increased the maximal pressures (p<0.05). Diabetes increased both basal (p<0.001) and maximal (p<0.001) pressures. In the control kidneys the infusion of NOR promoted an intense vasoconstriction, which was less intense during the second and the third periods of infusion- what is called tachyphylaxis. The kidneys under hypothermia showed tachyphylaxis only in the second period. But the group treated with L-arginine didn’t show tachyphylaxis – instead the values became higher and higher at each period of infusion of NOR. However we should be cautious about these results since they didn’t reach a p<0.05 value. The diabetic group showed a substantially decreased escape when compared to control (p<0.05), but this effect was partially reversed by the treatment with GSH and rutin. Esse estudo avaliou os efeitos da L-arginina, bem como de inibidores de NOS sobre o escape vascular e a taquifilaxia de rins de coelhos. Além disso, os efeitos do diabetes e da hipotermia também foram estudados. Rins de coelhos normais e com diabetes aloxânico foram perfundidos com solução de Krebs-Henseleit em um sistema aberto e a norepinefrina (NOR) 1 microM foi infundida por três períodos consecutivos de 20 min cada, com intervalos de 10 min para lavagem. No grupo controle, L-arginina e D-arginina foram adicionadas (ambas a 1 microM), bem como os inibidores de NOS, nitro-metiL-L-arginina (10 microM) e n(omega)-nitro-L-arginina (10 microM). Esse grupo foi também submetido a hipotermia (25o C). Os rins de animais diabéticos foram agrupados em não tratados e tratados com glutationa reduzida (1 microM) e rutina (1 microM). O tratamento com LNOARG elevou a pressão basal (p<0.001). O tratamento com L-arginina reduziu tanto a pressão basal (p<0.05) como a máxima (p<0.01). O tratamento com nitro-metiL-L-arginina elevou a pressão máxima (p<0.05). O diabetes elevou tanto a pressão basal (p<0.001) como a máxima (p<0.01). Nos rins do grupo controle, a infusão de NOR promoveu uma intensa vasoconstrição, a qual foi menos intensa durante o segundo e o terceiro período de infusão – caracterizando o fenômeno de taquifilaxia. Os rins sob hipotermia exibiram taquifilaxia apenas no segundo período. Mas, o grupo tratado com L-arginina não mostrou taquifilaxia – ao contrário, os valores tornaram-se mais altos a cada período de infusão de NOR. Todavia, precisamos encarar esses resultados com reserva, pois não atingiram significância estatística ao nível desejado, ou seja, p<0.05. O grupo diabético mostrou um escape bastante reduzido se comparado ao grupo controle (p<0.05), mas esse efeito foi parcialmente revertido pelo tratamento com glutationa reduzida e rutina. Dentre os rins do grupo de animais euglicêmicos, aqueles tratados com L-arginina – mas não com D-arginina – exibiram um escape elevado se comparados aos do grupo controle. Contudo, esse resultado também não alcançou significância estatística, isto é, p<0.05.
