Resumo: Alpinia zerumbet is a plant from Zingiberaceae family, popularly known in Brazil as “Colônia”, used for anxiety and systemic arterial hypertension treatment. Several studies have already demonstrated its anti-hypertensive, vasodilator and antioxidant activities, among others, but no research aimed to verify the vasodilator effect of essential oil from Alpinia zerumbet (EOAz) in resistance vessels. Thus, the aim of this study was to evaluate the vasorelaxant effect of EOAz and to assess its mechanism of action in mesenteric small arteries isolated from Wistar rats. Therefore, the effect of EOAz (03 to 3000 µg/mL) was verified by isometric tension, measured in rings of second-order branch of the mesenteric artery (SOBMA), with intact endothelium, pre-contracted by KCl (80 mM) and U-46619 (03 µM). To study the mechanism of action, it was evaluated the influence of endothelium presence, and the influence of several inhibitors (TEA, 4-AP, Glybenclamide, Atropine, L-NAME, ODQ and indomethacin) on vasodilator effect of EOAz in SOBMA pre-contracted by U-46619. Some protocols were also performed targeting to study Ca2+ influx, through CaCl2 contraction in high K+ medium, as well as Ca2+ release from intracellular storages by caffeine and phenylephrine induced contraction in Ca2+ free medium. Results showed an endothelium-independent vasorelaxant effect of EOAz on SOBMA pre-contracted by KCl, with an EC50 of 33.17 µg/mL (CI95%: 28.11 to 39.38), and U-46619, with an EC50 of 21.52 µg/mL (CI95%: 17.68 to 25.95). Significantly higher potency (P<0,01) was observed for SOBMA pre-contracted by U-46619, when compared to those pre-contracted by KCl. Regarding the inhibitors, only ODQ and L-NAME produced significant alteration on EC50 when compared to Control, with values of 39.47 µg/mL (CI95%: 36.7 to 42.44; P<0,001), and 35.15 µg/mL (CI95%: 33.41 to 37.03), respectively. However, the inhibitors did not cause an Emax reduction. Concerning calcium assays, reduction on muscle contraction caused by incubation with EOAz was observed in all three protocols in a dose-dependent manner. Hence, our results suggest that essential oil of Alpinia zerumbet causes a vasodilator effect, mediated by inhibition of Ca2+ influx and Ca2+ release from intracellular storages, as well as an activation of NOS/GCs pathway.

Já foram realizadas diversas pesquisas com a Alpinia zerumbet mostrando atividade anti-hipertensiva, vasodilatadora, antioxidante, dentre outras, porém, nenhuma pesquisa verificou o efeito vasodilatador do óleo essencial de Alpinia zerumbet (OEAz) em vasos de resistência. Assim, o objetivo desse trabalho foi avaliar o efeito vasodilatador do OEAz em artérias de resistência isoladas de rato Wistar, bem como estudar o seu mecanismo de ação. Para tanto, foi verificado o efeito do OEAz (3 – 3000 µg/mL) na tensão isométrica mensurada em segmentos de segundo ramo de artéria mesentérica (SSRAM), com endotélio íntegro, pré-contraídos com KCl (80 mM) ou U-46619 (3 µM). Além disso, para se avaliar o mecanismo de ação do OEAz, foi verificada a influência da presença do endotélio, bem como a interferência da incubação com inibidores de diversas vias de relaxamento muscular (TEA, 4-AP, Glibenclamida, Atropina, L-NAME, ODQ, Indometacina) no efeito vasodilatador do OEAz em SSRAM pré-contraídas com U-46619. Foram realizados, ainda, alguns protocolos visando verificar o efeito do OEAz no influxo de Ca2+, mediante contração induzida por CaCl2 em meio com alta concentração de K+, bem como na liberação dos estoques intracelulares de Ca2+, através da contração induzida por Fenilefrina e por Cafeína em meio sem Ca2+. Foi observado um efeito vasodilatador independente de endotélio do OEAz em SSRAM, com CE50 de 33,17 µg/mL (IC95%: 28,11 – 39,38) em artérias pré-contraídas com KCl e CE50 de 21,52 µg/mL (IC95%: 17,68 – 25,95) em artérias pré-contraídas com U-46619, tendo sido observada uma potência significativamente maior (P<0,01) do OEAz em SSRAM pré-contraídas com U-46619, quando comparadas às pré-contraídas com KCl. Com relação aos inibidores, os únicos que causaram uma alteração significativa na CE50 do OEAz, quando comparados ao controle, foram o ODQ, com CE50 de 39,47 µg/mL (IC95%: 36,7 – 42,44) (P<0,001), e o L-NAME, com CE50 de 35,15 µg/mL (IC95%: 33,41 – 37,03) (P<0,001), entretanto os inibidores não causaram uma redução no Emax. Com relação aos protocolos de contração induzida por CaCl2 em meio rico em K+, e por cafeína e fenilefrina em meio sem Ca2+, foi observada uma redução da contração muscular, nos três protocolos, causada pela incubação com o OEAz, de maneira dose-dependente.
