Resumo: Blood pressure is regulated by several factors, such as vasoconstriction and peripheral resistance, and when conditions prevail that contract the arteries, has been the hypertension, in which values above 140/90mmHg. When analyzing the increasing number of deaths due to cardiovascular diseases it should be remembered that hypertension is associated with aging, eating habits and sedentary lifestyle. Nitric Oxide (NO), molecule produced endogenously from the conversion of L-arginine in L-citrulline, is the main regulator of blood pressure. Stable ruthenium nitrosyl complexes have been searched in order to release NO thus acting on vascular smooth muscle relaxation. In view of the foregoing, this paper aims to synthesize, characterize and perform reactivity studies of a new compound, the trans-[Ru(NH3)4(BPA)(NO)](PF6)3. The characterization of nitrosilocomplex was performed by means of spectroscopic techniques (Infrared region vibrational spectroscopy, electron spectroscopy in the ultraviolet and visible regions), 1H-nuclear magnetic resonance, and cyclic voltammetry. The synthesized complex was then studied as to its ability to release NO° and the application of electric potential suggests the formation of aquo-complex. The reduction by GSH indicates that there is the possibility of NO° release occurring. The spectrophotometric titration showed that the conversion pH of the NO+/NO2- species to the trans-[Ru(NH3)4(BPA)(NO)](PF6)3 complex is 11.59. In addition, reactivity of nitrosyl complex facing superoxide radical was studied. The results suggested that compound presented antioxidant activity against free radical. The results indicate the obtaining of a new compound with the potential to act as a NO-releasing agent in the body. Cytotoxicity studies, as well as vasodilation assay should be performed to assess its efficacy as a vasodilator potential.

Quando se analisa o número crescente de óbitos decorrentes de doenças cardiovasculares, deve-se lembrar que a hipertensão está associada ao envelhecimento, hábitos alimentares e sedentarismo. O óxido nítrico (NO), molécula produzida endogenamente a partir da conversão da L-arginina em L-citrulina, é o principal regulador da pressão sanguínea. Nitrosilocomplexos de rutênio estáveis, tem sido pesquisados a fim de liberar NO, agindo assim no relaxamento do musculo liso vascular. Diante do exposto, este trabalho tem por objetivo sintetizar, caracterizar e realizar estudos de reatividade frente a espécies oxidantes, de um novo composto, o trans-[Ru(NH3)4(BPA)(NO)](PF6)3. A caracterização do nitrosilocomplexo foi realizada por meios de técnicas espectroscópicas (espectroscopia vibracional na região do infravermelho, espectroscopia eletrônica na região do ultravioleta e do visível), ressonância magnética nuclear de 1H e voltametria cíclica. O complexo sintetizado foi então estudado quanto a sua capacidade de liberar NO° e a aplicação de potencial elétrico sugere a formação do aquo-complexo. O espectro de RMN de 1H do composto indica a coordenação do ligante BPA ao metal por meio do nitrogênio piridínico, evidenciado pelo deslocamento dos sinais referentes a quebra de simetria e alteração do ambiente químico do mesmo. A redução por GSH aponta que há a possibilidade de ocorrer a liberação de NO° visto que o monitoramento da banda característica indicou uma diminuição da mesma na forma coordenada. A titulação espectrofotométrica mostrou que o pH de conversão das espécies NO+/NO2- para o complexo trans-[Ru(NH3)4(BPA)(NO)](PF6)3 é de 11,59 . Testou-se também a reatividade do nitrosilo complexo frente ao radical superóxido, obtendo o IC50 para o composto de 215,22 μmol L-1, e a constante de velocidade para a reação determinada foi de 1,76 x 105 L mol-1 s-1, sugerindo que o composto apresenta atividade antioxidante frente a esta espécie radicalar. Os resultados indicam a obtenção de um novo composto com potencialidade de agir como liberador de NO° no organismo.
