Resumo: | The aim of this work was to prepare nanocarriers by self-assembly and Pickering emulsions from acylated cashew gum (CG), using propionic and butyric anhydrides, named GCAc, GCProp and GCBut, for Amphotericin B (AmB) encapsulation. The reaction was performed in three CG/anhydrides molar ratio conditions (1:9, 1:12 and 1:24) and the acylation reaction was confirmed by FTIR and NMR spectra. The degree of substitution was measured by potenciometric titration and it increases with increasing of anhydride amount for GCAc and GCBut derivatives. TGA analysis showed the acylation provides a better thermal stability in comparison with GC. Zebrafish toxicity showed biocompatibility for GCProp and GCBut. Nanoparticles (NP´s) with monomodal size distribution, very negative zeta potential and low PDI were produced. SEM showed a smooth and spherical surface for nanoparticles. Pickering emulsions were prepared by adding the organic phase in the aqueous phase, resulting the immediate emulsion formation. The particle size of emulsion presented unimodal distribution and a size decrease is observed after the isolation by centrifugation. The atomic force microscopy confirmed the hypothesis that nanoparticles are around the oil drop. After AmB encapsulation there is an increase in particle size, decrease PDI and zeta potential for NP´s and emulsions. The efficiency of encapsulation (EE) was higher for NP´s (maximum EE = 96.3 ± 4.6%) than for nano emulsions (maximum EE = 55.2 ± 0.1%), however AmB is less aggregated in the emulsion system. Two stability assays were performed for NP's and Pickering emulsions and, in both assays, the two systems had excellent stability. The GCProp loaded AmB exhibited best size stability over 180 days. The in vitro release profile of AmB for the NPs had its maximum release of 61.06 ± 1.4% after 72 h of assay. The hemolysis assay showed that the GCProp 9 emulsion shows a small extent of damage to the erythrocytes, indicating a system with good compatibility. |

Foram realizadas três condições reacionais para cada anidrido, 1:9, 1:12 e 1:24, variando a razão molar GC/anidrido. A confirmação da reação foi possível através dos espectros de infravermelho e ressonância magnética nuclear. O grau de substituição (GS) foi determinado por titulação potenciométrica, que aumentou com o aumento da proporção molar de anidrido para os derivados GCAc e GCBut. A análise termogravimétrica evidenciou que os produtos da reação de acilação têm maior estabilidade térmica do que a GC sem modificação. A toxicidade realizada com o peixe- zebra mostrou biocompatibilidade para GCProp e GCBut. As nanopartículas (NP´s) foram preparadas via auto-organização e todos os sistemas apresentaram distribuição de tamanho unimodal, sem sinais de agregação e os valores de IPD indicam homogeneidade e estabilidade. O MEV evidenciou superfície lisa e esférica para as nanopartículas. As emulsões de Pickering foram formadas por emulsificação espontânea, vertendo-se a fase orgânica sob a fase aquosa. O tamanho de partículas para as emulsões apresentou distribuição unimodal e diminuiu após o isolamento. A microscopia de força atômica confirmou a hipótese da disposição das nanopartículas ao redor da gota de óleo. Após o encapsulamento da AmB houve o aumento tamanho de partícula, aumento do IPD e diminuição do potencial zeta, tanto para as NP´s quanto para as emulsões. A eficiência de encapsulamento (EE) mostrou que as NP´s encapsularam melhor a AmB (EE máxima = 96,3 ± 4,6 %) do que as emulsões (EE máxima = 55,2 ± 0,1 %), entretanto a AmB está menos agregada no sistema de emulsão. Foram realizados dois ensaios de estabilidade para NP´s e emulsão de Pickering e, em ambos os ensaios, os dois sistemas sem fármaco mostraram excelente estabilidade. Após o encapsulamento da AmB as NP´s GCProp foram mais estáveis no período de seis meses para o ensaio de estabilidade estoque. O perfil de liberação in vitro da AmB para as NP´s teve liberação máxima de 61,06 ± 1,4 % após 72 h de ensaio.
