Resumo: The objective of this work was to obtain new isolates of N. caninum and characterize the same pathogenically. In Nerópolis (Goiás, Brazil), two Holstein-Friesian male calves, aged four months, positive for indirect immunoflourescence for Neospora with a titre of 1:400 and 1:800 were utilized for the isolation. Two new isolates, NC- Goiás 1 and NC-Goiás 2, were obtained. NC-Goiás 2 was discarded, however, due to fungal contamination. In addition to characterization of isolate NC-Goiás 1, pathogenic characterizations were done for six Spanish isolates in female mice of the lineage Balb/c. Symptomatology and mortality caused by the isolates, parasitaemia, detection of parasite DNA in the brain and lungs were verified, as well as parasite load in the brain throughout the experiment. Further, the development of humoral immune response of the animals was verified. Statistical analysis was carried analysing the effect of isolate and the infection phase. Differences between percentages encountered through nested-PCR were analysed using the Chi-square test. When significant differences were observed (P<0,05), the groups were compared two by two. The difference between parasite loads, optical density values for IgG1, IgG2a and the relation IgG1/IgG2a between the isolates were analysed by the Kruskal-Wallis test. When statistically significant differences were observed (P<0,05), the groups were compared two by two using the Mann-Whitney U test. In blood and lungs, the parasite was detected mainly in the acute phase (P<0,05). The DNA of N. caninum was also detected in the brain mainly in the chronic phase (P<0,05). On comparing parasite loads between isolates, statistically significant differences were observed between them in the chronic phase (P< 0,05). Antibodies of class IgG1 and IgG2a were detected starting from day 16 P.I. and 4 P.I. respectively, increasing gradually and remaining high until day 64 P.I. An analysis of the relation IgG1/IgG2a demonstrated predominance of IgG2a on day 8 e 16 P.I. and highest levels of IgG1 were detected on day 32 and 64 P.I. The isolate NC-Goiás 1 is the first isolate obtained in Goiás and the first obtained from an infected calf, though clinically healthy, reported from South América. These results are the first of a study of the diversity of the protozoon, N. caninum, and indicate that these isolates are of low pathogenicity in mice as compared with other known isolates

Este trabalho teve como objetivo obter novos isolados de N. caninum e realizar a caracterização patogênica dos mesmos. Em Nerópolis (Goiás, Brasil) dois bezerros machos de quatro meses de idade, da raça Holandês, positivos por Imunofluorescência indireta para Neospora com título de 1:400 e 1:800, foram utilizados para isolamento. Dois novos isolados denominados NC-Goiás 1 e NC-Goiás 2 foram obtidos porém, o isolado NC-Goiás 2 foi perdido devido a contaminação por fungos. Além da caracterização do isolado NC-Goiás 1 foram feitas caracterizações biológicas de seis isolados espanhóis em camundongos fêmeas, da linhagem BALB/c. Foi verificada a sintomatologia e mortalidade provocada pelos isolados, a parasitemia, detecção de DNA do parasito no cérebro e pulmão e determinada a carga parasitária no cérebro ao longo do experimento. Além disso, também se verificou o desenvolvimento da resposta imune humoral dos animais. A análise estatística foi realizada para verificar a influência do isolado e a fase da infecção. As diferenças entre as porcentagens encontradas pela nested-PCR foram analisadas aplicando o teste de Qui-quadrado. Quando observadas diferenças estatisticamente significativas (P<0,05), os grupos foram comparados dois a dois. A diferença entre as cargas parasitárias, valores de densidade óptica para IgG1, IgG2a e a razão IgG1/IgG2a entre os isolados foram analisadas pela prova de Kruskal-Wallis. Quando observadas diferenças estatisticamente significativas (P<0,05), os grupos foram comparados dois a dois pelo teste de U de Mann-Whitney. No sangue e pulmão o parasito foi detectado principalmente na fase aguda (P<0,05). Quanto ao cérebro, foi detectado DNA de N. caninum principalmente na fase crônica (P<0,05). Ao comparar as cargas parasitárias entre os isolados, foram observadas diferenças estatisticamente significativas na fase crônica (P< 0,005). Anticorpos da classe IgG1 e IgG2a foram detectados a partir do dia 16 P.I. e 4 P.I. respectivamente, aumentando gradualmente e se mantendo elevados até o dia 64 P.I. A análise da relação de IgG1/IgG2a mostrou predominância de IgG2a nos dias 8 e 16 P.I. e os níveis de IgG1 mais elevados foram detectados nos dias 32 e 64 P.I. O isolado NC-Goiás 1 é o primeiro isolado de N. caninum obtido em Goiás e o primeiro isolado obtido de um bezerro infectado porém clinicamente saudável relatado na América do Sul.
